Project description:Setd2 methylate the nucleosome to form H3K36me3. Here we utilized the Cryo-EM to elucidate the structure of SETD2/Set2 bound with nucleosomes. Through this structure analysis, we found that histone H1 may interfere the enzymatic activity of SETD2/Set2 by inhibiting their binding affinity.

High-resolution cryo-EM structure of the respiratory alternative complex III from Rhodothermus marinus

Project description:Electron transfer in respiratory chains generates the electrochemical potential that serves as the energy source for ATP synthesis, solute transport and motility. In eukaryotes and many bacteria, the respiratory chain consists of four electron transport complexes, known as complex I to IV. Respiratory chains of some prokaryotes differ in composition and organization. They can use a wide range of electron donors and acceptors and may have complexes performing the same catalytic reaction. The diversity and apparent redundancy of prokaryotic respiratory chains reflects the versatility and robustness of the organisms. Many of these alternative respiratory chain complexes are either unknown or their structures and mechanisms remain elusive. In this work we describe a 3.9 A cryo-EM structure of the alternative complex III (ACIII) from Rhodothermus marinus, which we demonstrated takes over the role of canonical respiratory complex III (bc1 complex), even though it is structurally unrelated. Our structure reveals that ACIII is an integral membrane protein complex of at least 7 subunits (ActABCDEFH). The periplasmic domain, with ActA, B, E and H, harbours four iron-sulphur clusters and six C-type hemes. The cofactors form two electron wires that converge on the putative quinol-binding site in subunit ActC. The two homologous subunits, ActC and ActF, each have two four-helix bundles in the membrane, with several conserved polar residues that delineate putative proton channels. ACIII meets all requirements for an energy-transducing machine that couples electron transfer from quinol to the oxygen reductase, to translocation of protons across the membrane.

Project description:The COVID-19 pandemic prompted an unprecedented effort to develop effective countermeasures against SARS-CoV-2. While efficacious vaccines and certain therapeutic monoclonal antibodies are available, here, we report the development, cryo-EM structures and functional analyses of distinct potent monoclonal antibodies (mAbs) that neutralize SARS-CoV-2 and its variant B.1.351. We established a platform for rapid identification of highly potent and specific SARS-CoV-2-neutralizing antibodies by high-throughput B cell receptor single cell sequencing of spike receptor binding domain immunized animals. We identified two highly potent and specific SARS-CoV-2 neutralizing mAb clones that have single-digit nanomolar affinity and low-picomolar avidity. We also generated a bispecific antibody of these two lead clones. The lead monospecific and bispecific antibodies showed strong neutralization ability against prototypical SARS-CoV-2 and the highly contagious South African variant B.1.351 that post a further risk of reducing the efficacy of currently available therapeutic antibodies and vaccines. The lead mAbs showed potent in vivo efficacy against authentic SARS-CoV-2 in both prophylactic and therapeutic settings. We solved five cryo-EM structures at ~3 resolution of these neutralizing antibodies in complex with the ectodomain of the prefusion spike trimer, and revealed the molecular epitopes, binding patterns and conformations between the antibodies and spike RBD, which are distinct from existing antibodies. Our recently developed antibodies expand the repertoire of the toolbox of COVID-19 countermeasures against the SARS-CoV-2 pathogen and its emerging variants.

Cryo-EM structure of the volume-regulated anion channel LRRC8D isoform identifies features important for substrate permeation

Project description:Members of the leucine-rich repeat-containing 8 (LRRC8) protein family, composed of the five LRRC8A-E isoforms, are pore-forming components of the volume-regulated anion channel (VRAC), which is activated by cell swelling and transports halide anions and small organic compounds. Despite the recent availability of the cryo-EM structures of homo-hexameric LRRC8A, the structural basis of how LRRC8 isoforms other than LRRC8A contribute to the functional diversity of VRAC has remained elusive. In this study, we confirmed the absence of LRRC8 isoforms in the purified HsLRRC8D using mass spectrometry, and the structure of the HsLRRC8D homomer was analyzed using the cryo-EM. This work provides structural insights into the functional diversity of the LRRC8 protein family.

Cryo-EM structure of ex vivo fibrils associated with extreme AA amyloidosis prevalence in a cat shelter

Project description:We present the 3.3 Å cryo-EM structure of an AA amyloid extracted post-mortem from the diseased kidney of a DSH cat with renal failure. The structure reveals a cross-beta architecture assembled from two 76-residue long proto-filaments. Despite >70% sequence homology to mouse and human SAA, the cat SAA variant adopts a unique amyloid fold exhibiting type-2 polymorphism. Based on shared disease profiles and almost identical sequences, we propose a similar amyloid fold of deposits identified previously in captive cheetah.

Project description:The Tthyroglobulin (Tg) protein is the essential fundament toto thyroid hormone synthesis, a key player playing a vital role in the regulation of metabolism, development and growth and serving as intraglandular iodine storage. Its structure is conserved among vertebrates. Tg is delivered through the secretory pathway of the thyroid follicular unit to the central colloid depository, where it is iodinated at specific tyrosine sites to form mono- or diiodotyrosine, which combine to produce triiodothyronine (T3) and thyroxine (T4), respectively. Synthesis of these hormones depends on the precise 3D structure, iodination capacity and post-translational modification of Tg. The Tg structure is overall conserved among vertebrates and , which and structural information has long been remained unknown missing despite decades of research until recently structural information has been made available on human thyroglobulin (hTg) originating from recombinant hTg and endogenous hTg from patients with goitres. Here, we present the cryo-electron microscopy structure of native and fully functional human thyroglobulin (hTg) originating from healthy human thyroid glands, to a global resolution of 3.2 Å. We used LC-MS experiments to The structure provides detailed information on the location of the hTg hormonogenic sites by revealing native iodination, and reveals the position as well as the role of many of its glycosylation sites and other post-translational modifications. Our results offer structural insights into thyroid hormonogenesis of natively functional hTg and provide a fundamental understanding of clinically relevant hTg mutations, which can improve treatment of thyroid diseases.

Structural basis of respiratory complexes adaptation to cold temperatures

Project description:In response to cold, mammals activate brown fat for respiratory-dependent thermogenesis reliant on the electron transport chain. Yet, the structural basis of respiratory complex adaptation to cold remains elusive. Herein we combined thermoregulatory physiology and cryo-EM to study endogenous respiratory supercomplexes exposed to different temperatures. A cold-induced conformation of CI:III2 (termed type 2) was identified with a ~25 rotation of CIII2 around its inter-dimer axis, shortening inter-complex Q exchange space, and exhibiting different catalytic states which favor electron transfer. Large-scale supercomplex simulations in lipid membrane reveal how unique lipid-protein arrangements stabilize type 2 complexes to enhance catalytic activity. Together, our cryo-EM studies, multiscale simulations and biochemical analyses unveil the mechanisms and dynamics of respiratory adaptation at the structural and energetic level.
