Proteomic and Direct Analysis in Real Time Mass Spectrometry Analysis of a Native American Ceremonial Hat

ABSTRACT: Direct analysis in real time (DART) surface analysis files and LC-MS proteomics files of a Tlingit ceremonial hat. DART was performed to evaluate what was present on the surface and also to help understand connection between two components of the hat. Proteomics was performed on small samples of material from around the hat to characterize the original adornment. The hat is housed at the Smithsonian Natural Museum of Natural History (E74339, E74441). Proteomics data were searched with PEAKS 8.5 and Prosight Lite v1.4 Build 1.4.6

Species identification of ivory and bone museum objects using minimally invasive proteomics

Project description:Ivory is a highly prized material in many cultures since it can be carved into intricated designs and have a highly polished surface. Due to its popularity, the animals from which ivory can be sourced have started to come under threat. Identification of the ivory species is not only important for compliance with the Convention on International Trade in Endangered Species of Wild Fauna and Flora (CITES), it can also provide important information about the context in which the work was created. In this work, we have developed a minimally invasive workflow to remove minimal amounts of material from precious objects, and, using high-resolution mass spectrometry-based proteomics, identified the taxonomy of several ivory and bone objects from the collection of The Metropolitan Museum of Art dating from as early as 4000 B.C. We built an inhouse proteomic databases of underrepresented species based on exemplars obtained from the Mammology American Museum of Natural History collection and proposed alternative data analysis workflows for rare samples containing sparse and inconsistently preserved organic material. This is a first application demonstrating extensive and accurate ivory species identification using proteomics to unlock sequence uncertainties, e.g. Leu/Ile-discrimination.

Incorporation of In Vitro Techniques for Botanicals Dietary Supplement Safety Assessment – Towards evaluation of Developmental and Reproductive Toxicity (DART)

Project description:As complex mixtures, botanicals present unique challenges when assessing safe use, particularly when endpoint gaps exist that cannot be fully resolved by existing toxicological literature. Obtaining data for developmental and reproductive toxicity (DART) can be particularly difficult as this is a frequent data gap for botanicals. Here we explore in vitro receptor binding and enzyme activity assays as well gene expression studies as alternative tools to inform on DART relevent modes of action. Botanicals suspected to have DART effects in addition to herbs with a significant history of use were tested at biologically relevant doses to probe activity at a molecular level. Additionally, gene expression changes in a number of different cell types were analysed using the connectivity mapping approach (CMap) to identify modes of action through a functional read across approach. Together, these two data streams increase the confidence in predictions of biological mode of action and inform potential DART risk by way of potency comparisons and/or a margin of internal exposure approach.

Assessing the hidden potential of beeswax specimens in Natural History Museum collections

Project description:Natural history museum specimens of historical honeybees have been successfully used to explore the genomic past of the honeybee, indicating fast and rapid changes between historical and modern specimens, possibly as a response to current challenges. In our study we explore a potential untapped archive from natural history collections - specimens of beeswax. We examine an Apis mellifera mellifera queen cell specimen from the 19th century. The intact and closed cell was analysed by X-ray Computed Tomography (CT) to reveal a perfectly preserved queen bee inside her cell. Subsequently, a micro-destructive approach was used to evaluate the possibility of protein extraction from the cell. Our results show that studies on specimens such as these provide valuable information about the past rearing of queens, their diet and development, which is relevant for understanding current honeybee behaviour. In addition we evaluate the feasibility of using historical beeswax as a biomolecular archive for ancient proteins to study honeybees.

Expression profile comparison of African American and American Caucasian esophageal squamous epithelium from subjects with and without Barrett's Esophagus

Project description:Histologically normal biopsies of the esophageal squamous epithelium were collected from consented individuals of either African American (AA) or American Caucasion (Cau) ethnicity. Total RNA was extracted and used to generate Affymetrix expression array profiles. For each racial group collected samples were either controls (no history of BE or EAC) or those with a history of BE (Barrett's Esophagus) and/or EAC (esophageal adenocarcinoma). This allowed us to identify gene with tissue specific expression differences between the two racial groups, as well as those that differed between control and disease groups.
