Project description:Hepatic ischemia/reperfusion (I/R) injury represents a major risk factor for liver transplantation and is related to graft dysfunction and acute/chronic rejection. However, a significant part of these processes remain poorly characterized. To reveal differences in the proteome during liver I/R injury, we collected human liver biopsy samples during hepatectomy before and after the Pringle maneuver and conducted a TMT-based proteomic analysis through quantitative high-throughput mass spectrometry. We used a fold-change threshold of 1.3 and a t-test p-value < 0.05 as the criteria to identify 5,257 total quantifiable proteins. The levels of 142 proteins were increased, while the levels of 103 proteins were decreased in response to hepatic I/R treatment. Bioinformatic analysis further revealed that these differentially expressed proteins are mainly involved in multiple biological functions and enzyme-regulated metabolic pathways. Most proteins whose expression was changed are related to the defense, immune and inflammatory responses as well as lipid and steroid metabolic processes. Based on this finding, we developed a panel for targeted proteomic analysis and used the parallel reaction monitoring (PRM) method, qPCR and western blotting experiments to validate alterations in the expression of some of the identified proteins. The upregulated proteins were found to be involved in immunity and inflammatory responses, and downregulated proteins were enriched in metabolic pathways. This study therefore may provide a research direction for the design of new therapeutic strategies for hepatic ischemia/reperfusion injury.

Project description:BackgroundThe treatment approach for aggressive fibromatosis is changing. Although surgery is the mainstay in common practice, recent literature is reporting a more conservative approach. We compared the local control rate for surgery, surgery with radiotherapy, radiotherapy alone and a wait and see policy in a systematic review.MethodsA comprehensive search of the databases PubMed/Medline, Embase and Cochrane, of the medical literature published in 1999 till March 2017 was performed by two reviewers, including articles about extra abdominal aggressive fibromatosis without the genetical variants. A total of 671 studies were assessed for eligibility, and 37 studies were included for analysis, representing 2780 patients.ResultsThe local control rates for surgery alone, surgery and radiotherapy, radiotherapy alone and observation were 75%, 78%, 85% and 78%, respectively. For patients with recurrent disease observation had a better local control rate than surgery alone (p?=?0.001). In the observation group, stabilization of the tumor was seen in median 14 (range 12-35)?months. The time to local recurrence in the treatment group was median 17 (range, 11-52)?months.ConclusionA watchful conservative first line approach with just observation and closely monitoring, by means of physical examination and MRI, appears to be justified in a subgroup of patients without clinical symptoms and no possible health hazards if the tumor would progress.

Project description:Farnesol is a nonsterol isoprenoid produced by dephosphorylation of farnesyl pyrophosphate, a catabolite of the cholesterol biosynthetic pathway. These isoprenoids have been reported to inhibit proliferation and induce apoptosis in neoplastic cell lines as well as to be effective in chemotherapy in several in vivo cancer models. Recently, it was shown that farnesol triggers morphological features characteristic of apoptosis in the filamentous fungus Aspergillus nidulans. In order to investigate which pathways are involved under Farnesol treatment, we determined the transcriptional profile of A. nidulans wild type strain. Conidia were incubated at 37°C in complete medium for 16 hours and were esposed or not to 100 μM Farnesol for 2 hours. The mycelia were harvested by centrifugation and used for competitive microarray hybridizations. We detected differential regulation of genes involved in a variety of cellular processes whose specific modulation is likely to be implicated with A. nidulans adaptation to farnesol. We observed decreased mRNA abundance of several genes involved in RNA processing and modification, transcription, translation, ribosomal structure and biogenesis, amino acid transport and metabolism. Interestingly, several genes involved in the ergosterol biosynthesis (such as the homologues of erg4/24, 11A, -11B, -13, -25, and –28) have decreased mRNA accumulation and other genes encoding mitochondrial proteins [such as AN9103.3, AN4500.3, and AN5440.3, encoding the Apoptosis Inducing Factor (AIF)-like mitochondrial oxidoreductase, the mitochondrial ATPase inhibitor, and the cytochrome c peroxidase, respectively] have increased mRNA expression when A. nidulans was exposed to farnesol. We also observed as more expressed several genes encoding proteins involved in trehalose metabolism and chaperones. Keywords: farnesol effect
