Novel Brutons Tyrosine Kinase (BTK) substrates for time-resolved luminescence assays


ABSTRACT: BTK substrate profile determined by treating digest KG-1 cell lysate with alkaline phosphatase followed by recombinant BTK. Samples were enriched with PolyMac phosphoenrichment kits prior to analysis. We have organized our uploaded files into three main folders: 1. BTK KALIP MS raw files: raw mass spectrometer files in .RAW format. 2. BTK KALIP PEAKS analysis: project files from data analysis using PEAKS Studio X pro. These files are formatted as a PEAKS projects and can be opened in PEAKS software. 3. BTK PEAKS output files: exported files from the PEAKS Studio X pro analysis, including protein-peptides.csv files for each replicate.

Bruton's tyrosine kinase (BTK) is a well-documented target for cancer therapeutics due to its role in B-cell signaling pathways. However, inhibitor design is hindered by lack of tools to assess kinase activity. We used in vitro phosphoproteomics to determine BTK's substrate preferences and applied this information to our updated data processing pipeline, KINATEST-ID 2.1.0.
