IodoTMT 6plex analysis of cysteine oxidation following eccentric contractions

ABSTRACT: Two iodoTMT 6-plex screens were used to assess the changes in protein cysteine oxidation following eccentric contractions in extensor digitorum longus (EDL) muscles that were extracted from wild type and mdx mice and treated with and without 750uM sodium hydrogen sulfide (NaHS). N=5 for each group: WT, WT+ECC, mdx, mdx+ECC, and mdx+ECC+NaHS.

Hydrogen sulfide, a novel small molecule signalling agent, participates in GAs biosynthesis under heat stress and the regulation of multiple signaling pathway genes in Ganoderma lucidum

Project description:As one of the most important environmental factors, heat stress (HS) has been found to affect various biological activities of organisms such as growth, signal transmission, primary metabolism and secondary metabolism. Ganoderma lucidum has become a potential model system for evaluating how environmental factors regulate the secondary metabolism of basidiomycetes. Previous research showed that HS can induce the biosynthesis of ganoderic acids (GAs). In this study, we found the existence of hydrogen sulfide in Ganoderma lucidum; moreover, HS increased GAs biosynthesis and could affect the hydrogen sulfide content. We found that sodium hydrosulfide (NaHS), an exogenous donor of hydrogen sulfide, could revert the increased GAs biosynthesis elicited by HS. This result indicated that an increased content of hydrogen sulfide, within limits, was associated with HS-induced GAs biosynthesis. Our results further showed that the GAs content was increased in CBS-silenced strains and could be reverted to WT strain levels by the addition of NaHS. Transcriptomic analyses indicated that that H2S can affect various intracellular signal pathways and physiological processes in G. lucidum. Further studies showed that H2S could affect the intracellular calcium concentration and thus regulate the biosynthesis of GAs. This study demonstrated that hydrogen sulfide is involved in the regulation of secondary metabolic processes induced by heat stress in filamentous fungi.

The effect of slow and rapid H2S production on the levels of LPS-induced proinflammatory mediators and transcription in different human cell cultures

Project description:In this study we investigated the effect of two different agents producing hydrogen sulfide (H2S) on LPS-induced inflammatory mediators such as TNFa, NO and ROS. The agents (GYY4137 and sodium hydrosulfide) applied differ significantly by the kinetics of H2S release. In the investigation we compared the effects of H2S release on the LPS-induced inflammation exploring SH-SY5Y human neuroblastoma cell line and human promonocytic cell line, THP-1. Our data clearly show that both hydrogen sulfide producing agents significantly reduced the production of the investigated proinflammatory mediators when applied both before and after LPS administration. Furthermore, transcriptomic studies demonstrated that H2S release ameliorates the expression of multiple proinflammatory genes up-regulated due to LPS administration. However, the pattern of changes observed in transcriptome differs dramatically depending on the time of H2S release (before or after LPS-challenge).

Sulfide Regulates Autophagy by Reversible Persulfidation of the Cysteine Protease ATG4

Project description:Hydrogen sulfide is a signaling molecule that regulates essential processes for plant performance, such as autophagy. In Arabidopsis thaliana, hydrogen sulfide negatively regulates autophagy independently of reactive oxygen species, but the underlying mechanism remains to be understood. To determine whether persulfidation, an emergent posttranslational modification, has a main role in the control of autophagy by sulfide, we have used a proteomic approach targeting ATG4, a cysteine protease that plays a crucial role in autophagy progression. We showed that AtATG4a from A. thaliana, which is the predominant ATG4 protease in this plant species, contains a specific site for persulfidation, the residue Cys170, included in the characteristic catalytic triad Cys-His-Asp of cysteine proteases. We tested whether persulfidation regulates the activity of ATG4 by setting up a heterologous assay using the Chlamydomonas reinhardtii CrATG8 protein as a substrate. Our findings demonstrate that sulfide significantly inactivates AtATG4a cleavage activity in a reversible manner. The biological significance of the reversible inhibition of the ATG4 protease by sulfide is supported by our findings in Arabidopsis leaves under both basal and autophagy-inducing conditions. We also observed a significant increase in the overall ATG4 activity in Arabidopsis under nitrogen starvation and osmotic stress, which is inhibited by sulfide. Therefore, our data strongly suggest that the negative regulation of autophagy by sulfide is mediated, at least, by the specific persulfidation of the protease ATG4.

Project description:The aim of this experiment was to determine how exposure of Hydrogen sulfide impacts gene expression in Mycobacterium tuberculosis. RNA was isolated from actively growing mycobacterial cells (0.6-0.8 OD600) using Trizol according to established protocols (27). Briefly, cells were exposed to 25 µM GYY4137 for 1 hr under carefully controlled conditions (n=3/group) and RNA isolated. Unexposed cells received spent GYY4137 (without any capacity to produce Hydrogen sulfide).

Transcriptome profiles with and without L-cysteine supplementation

Project description:CyuR (b0447, also known as DecR or YbaO) is a recently characterized transcription regulator responsible for the generation of hydrogen sulfide (H2S) from L-cysteine (Cys), which decreases oxidative stress. Given that mitigation of oxidative stress is an important survival mechanism to achieve antibiotic resistance (AR) in many pathogenic bacteria such as Escherichia coli and Salmonella enterica, detailed studies about its effect and the regulatory network of CyuR are imperative. In this study, we investigated the roles of CyuR and its regulon genes in a Cys-dependent AR mechanism in E. coli in a microaerobic condition. We show that (1) CyuR negatively controls the expression of mdlAB encoding a transporter that may export antibiotics. (2) Cys metabolism has a significant role in AR and its effect is conserved in many E. coli strains including clinical isolates. (3) CyuR binds ‘GAAwAAATTGTxGxxATTTsyCC’ in the absence of Cys found by performing an in vitro binding assay. (4) CyuR may regulate 14 genes, in addition to previously reported 2 genes, as we suggested by in silico motif scanning and transcriptome sequencing. Collectively, our findings confirm the significant roles of CyuR and its relevance to antibiotic resistance associated with Cys.

In mammals, H2S is mainly generated by two pyridoxal-5'-phosphate-dependent enzymes, termed cystathionine β-synthase (CBS) and cystathionine γ-lyase (CSE). CBS-deficient mice showed autoimmune disorders. H2S play important roles in T cell development and differentiation, especially Treg cells development and differentiation. We used microarrays to detail the global gene expression of WT and CBS-deficient CD4+ T cells.

Global gene profiling comparison between hydrogen sulfide- and N-methyl-D-aspartate-induced neuronal deaths

Project description:Hydrogen sulfide (H2S), present in abundance in the mammalian brain, has recently been demonstrated to induce a dose- and time-dependent apoptotic-necrotic continuum in murine primary cortical neurons, which was successfully attenuated upon application of N-methyl-D-aspartate (NMDA) receptor antagonist. The current study focused on gaining an insight into the molecular mechanisms of H2S–mediated neuronal death pertaining to NMDA receptors activation through global gene expression comparisons. A total of 24 RNA samples were analyzed. There are 2 treatment conditions, namely 200uM sodium hydrosulfide and 200uM N-methyl-D-asparate. 3 replicates were collected for each of the selected time-points (5h, 15h and 24h), in addition to 6 replicates of shared vehicle control. The supplementary file 'GSE16035_non-normalized_data.txt' contains non-normalized data for Samples GSM401312-GSM401335.
