Human immune cell proteomic library

ABSTRACT: Comprehensive human immune cell proteomic spectral library which contain CD4+T cell, CD8+ T cell, NK cell, B cell, PBMC, Daudi, Jurkat, stimulated Jurkat, Molt-4, Ramos, and THP-1 were generated. This library includes 10,544 proteins groups, and 127,106 peptides.

Although proteomics is extensively used in immune research, there is currently no publicly accessible spectral assay library for the comprehensive proteome of immune cells. Proteomics of immune cells from liver tumors reveals immunotherapy targets

Project description:Elucidating the mechanisms by which immune cells become dysfunctional in tumors is critical to developing next-generation immunotherapies. We profiled proteomes of cancer cells, monocyte/macrophages, CD4+ and CD8+ T cells, and NK cells isolated from tumors, liver, and blood of 48 patients with hepatocellular carcinoma. We found that tumor macrophages induce the sphingosine-1-phospate-degrading enzyme SGPL1, which dampened their inflammatory phenotype and anti-tumor function in vivo. We further discovered that the signaling scaffold protein AFAP1L2, typically only found in activated NK cells, is also upregulated in chronically stimulated CD8+ T cells in tumors. Ablation of AFAP1L2 in CD8+ T cells increased their viability upon repeated stimulation and enhanced their antitumor activity synergistically with PD-L1 blockade in mouse models. Our data revealed new targets for immunotherapy and provide a resource on immune cell proteomes in liver cancer (www.immunomics.ch/liver).

Transcription profiling of human immune cell line mixtures to test deconvolution

Project description:Microarray deconvolution is a technique for quantifying the relative abundance of constituent cells in a mixture based on that mixture's microarray signature and the signatures of the purified constituents. Its ability to discriminate related human cells is unknown. Here we test the ability of this technique to determine the fractions of transformed cells of immune origin in mixed samples. Experiment Overall Design: Four immune cell lines were grown and run on microarrays either by themselves or in mixtures of various relative proportions. Mixtures of cells were performed in triplicate. Expression profiles from a variety of resting and activated human immune cells

Project description:Immune cell-specific expression is one indication of the importance of a gene's role in the immune response. In order to identify such patterns, we set out to broadly profile gene expression in a variety of immune cells. We isolated twelve different types of human leukocytes from peripheral blood and bone marrow, treated them to induce activation and/or differentiation, and profiled their gene expression before and after treatment. The twelve cell types are: B cells, CD14+ cells, CD4+ CD45RO+ CD45RA- T cells, CD4+ T cells, CD8+ T cells, IgG/IgA memory B cells, IgM memory B cells, Monocytes, NK cells, Neutrophils, Plasma cells from bone marrow, and Plasma cells from PBMC.

Optimized minimal genome-wide human sgRNA library

Project description:To minimize the human genome-wide CRISPR/Cas9 library size, we established H-mLib which recruited a novel sgRNA design method and applied with dual plasmid based strategy. The performance of the H-mLib was benchmarked to other CRISPR libraries in a proliferation screening conducted in K562 cells. We also identified human core essential genes and cell-type specific essentials genes in K562 and Jurkat cells.
