Environmental impacts on barley grain composition and longevity

Project description:<p>To counter projected reductions in yields of the major crop barley, it is essential to elucidate the mechanisms of its resilience. To assist such efforts, we collected grains from plants grown in fields at 12 testing stations, with suitable temperature and precipitation gradients for identifying environmentally induced changes in their protein and metabolite contents. We then subjected the grains to detailed molecular analysis. The results showed that numerous metabolites and at least a quarter of the grain protein content was modulated by the environment, and provided insights into barley seed production under abiotic stress, including alterations in ribosomal proteins, heatshock protein 70 family proteins, inhibitors, storage proteins and lipid droplet formation. Potential positive and negative markers of yield were also identified, including the phenolic compound catechin and storage protein levels, respectively. Complementary analyses of barley seedlings and Arabidopsis seeds, respectively, confirmed the role of the identified proteins in abiotic stress responses and highlighted evolutionarily conserved mechanisms. In addition, accelerated ageing experiments revealed that variations in the environment had stronger effects on seed longevity than the genotype. Finally, seeds with the highest longevity differed from the others in gibberellin contents, H2O2 metabolism and levels of >250 proteins, providing novel targets for improving resilience.</p>

RNA-seq of Sitobion avenae body parts and developmental stages

Project description:The English grain aphid, Sitobion avenae, is a major agricultural pest of wheat, barley and oats, and is a major vector of Barley Yellow Dwarf Virus (BYDV) leading to reductions in grain yield. RNA-seq data from a genotype (SA3) was generated from heads and bodies, and from winged and unwinged aphids. The primary goal was to generate evidence for genome annotation, and the secondary goal was to compare expression of genes between head and body, and also between winged and unwinged aphids.

Expression of Arabidopsis WRI1 and DGAT1 during soybean embryo development alters oil and carbohydrate composition

Project description:Soybean (Glycine max [L.] Merr.) is the largest feedstock for protein and second largest source of vegetable oil in the world. In order to evaluate whether the expression of ATWRI1 and AtDGAT1 could direct carbon flow towards lipid accumulation, we generated three independent transformation events (WD-1, -2, and -3). The transgenic events were evaluated in three field trials (2018, 2019, and 2020) as well as greenhouse conditions. RNAseq analysis was performed in immature embryos of both transgenic events grown under greenhouse conditions. We discovered that Glycolysis and FA metabolism enzymes are enriched in WD differentially expressed genes during seed development.

Transcriptomic and alternative splicing analyses reveal mechanisms of the difference in salt tolerance between barley and rice

Project description:Both barley (Hordeum vulgare) and rice (Oryza sativa) belong to Poaceae family, but differ greatly in salt tolerance. In order to understand molecular mechanisms in the difference of salt tolerance between the two species, the responses of transcriptomic profiles to salt stress were compared between rice (cultivar Nipponbare) and barley (accession XZ26) to reveal how alternative splicing (AS) coordinates with transcriptional regulation in adaptation to salt stress. Physiological study showed that XZ26 had higher salt tolerance than Nipponbare, as reflected by less growth inhibition, lower shoot Na+ concentration and higher K+/Na+ ratio when exposed to salt stress. Transcriptomic analysis showed that XZ26 had higher ROS scavenging ability, less degradation of protein kinases and enhanced anti-oxidation. Moreover, AS genes related to ion transporter genes and transcription factors could enhance and amplify K+/Na+ homeostasis and signal transduction cascades. We proposed that higher salt tolerance of barley accession XZ26 is attributed to its superior K+/Na+ homeostasis, tissue detoxication and less energy consumption. The present results provide insights at transcriptomic levels into reasons why barley has higher salt tolerance than rice.
