Project description:Polycystic ovary syndrome (PCOS) is a common disorder encompassing reproductive, metabolic, and endocrine abnormalities, affecting 5 to 10% of women of reproductive age.To reveal the differences in proteomic profiles of follicular fluid between patients with and without PCOS and explore possible mechanisms underlying PCOS.Follicular fluid samples were collected from 9 infertile patients with PCOS and 9 infertile patients without PCOS. Quantitative proteomics analysis based on mass spectrometry was used to measure the protein levels and understand the protein networks. The tandem mass tag (TMT) based proteomics technology and bioinformatics analysis were used to determine the differentially expressed proteins(DEPs). : In this study, we have identified a total of 1216 proteins,these included 70 DEPs, in which 32 proteins were upregulated, and 38 proteins were downregulated. The bioinformatics analyses revealed multiple biological functions associated with these DEPs, some DEPs were enriched in the immune and metabolic-related biological processes in PCOS patients.

Project description:Loss of ultrafiltration capacity in peritoneal dialysis (PD) patients is often associated with peritoneal vascular changes, manifest as diabetes-like vasculopathy and angiogenesis. The endothelial monolayer lining the vessel lumen controls vessel barrier function and thereby influences peritoneal substrate transport and ultrafiltration. The dipeptide alanyl-glutamine (AlaGln) has recently shown cytoprotective effects when added to PD fluid, including preservation of mesothelial cells in vitro (Kratochwill et al., Nephrol Dial Transplant, 2012, doi:10.1093/ndt/gfr459) and attenuation of the exuberant angiogenesis seen in long-term rodent models of PD (Ferrantelli et al., Kidney international, 2016, doi:10.1016/j.kint.2015.12.005). These effects have been reflected in early phase clinical trials as restoration of effluent cell stress responses and improved biomarkers of peritoneal health (Kratochwill et al., PLoS One, 2016, doi:10.1371/journal.pone.0165045; Vychytil et al., Kidney international, 2018, doi:10.1016/j.kint.2018.08.031). In a randomized clinical phase II trial, 8 weeks treatment with AlaGln in PD fluid decreased peritoneal protein loss. This clinically important effect might be explained by preserved peritoneal membrane and vessel integrity, but the role of AlaGln in preservation of peritoneal endothelial cell function remains unclear. In this study, we investigated to what extent in vivo observed biological processes and pathways are replicated by in vitro exposure of human umbilical vein endothelial cells (HUVEC) to conventional PD fluid. HUVECs were exposed to experimental solutions for up to 24 h. All test fluids were sterile-filtered before usage. Each experiment consisted of three independent samples in biological replicates on separate culture plates. For PD fluid incubation, cells were first exposed for 1 h to pure glucose-based PD fluid (Dianeal PD4 3.86% glucose, Baxter, Castlebar, Ireland), or to the same PD fluid supplemented with AlaGln dipeptide (8 mM, Dipeptiven; Fresenius Kabi, Bad Homburg, Germany), or to normal medium without growth factors as control. Cells were subsequently exposed for 24 h to the above solutions diluted 1:1 with culture medium and brought to 2% FCS. Cells were then washed three times (250 mM sucrose, 10 mM Tris/HCl, pH 7) and lysed in 125 µL per well of lysis buffer (30 mM Tris, pH 8.5, 7 M urea, 2 M thiourea, 4% CHAPS, 1 mM EDTA, one tablet of Complete Protease Inhibitor (Roche, Basel; Switzerland) per 100 ml, and one tablet of PhosStop Protease Inhibitor (Roche) per 100 mL). Total protein concentration was determined with the 2D-Quant kit (GE Healthcare, Uppsala, Sweden) per manufacturer’s manual. Lysates were stored at -80°C until further processing. We investigated the molecular mechanisms of endothelial cell pathology during in vivo and in vitro PD fluid exposure by proteomic and bioinformatic analysis of the endothelial cell response to hyperglycemic stress, integrating current understanding of PD-related cellular injury and stress responses (Bender et al., Nephrol Dial Transplant, 2011, doi:10.1093/ndt/gfq484; Kratochwill et al., BioMed research international, 2015, doi:10.1155/2015/628158; Kratochwill et al., J Proteome Res, 2009, doi:10.1021/pr800916s; Lechner et al., J Proteome Res, 2010, doi:10.1021/pr9011574). This strategy allowed characterization of endothelial cell injury and stress responses during exposure to conventional hyperglycemic PD fluid, and their modulation by added AlaGln.

Project description:Polycystic ovary syndrome (PCOS) is a complex endocrinopathy affecting reproductive aged women, whose etiology has not been fully understood yet. The follicular growth is arrested at preantral stage leading to cyst formation, consequently resulting in anovulatory infertility in these women. As follicular fluid provides the microenvironment for the growing oocyte, molecular profiling of the fluid may provide unique information about pathophysiology associated with follicular development in PCOS. Modification with oligosaccharide chains are known to influence functions of several secreted proteins and these glycoproteins also play a role in disease pathology. The glycoproteomic profile of follicular fluid of PCOS has not been explored in PCOS yet. In the present study, we performed comparative glycoproteomic analysis by first enriching glycoproteins using three different lectins viz. concanavalin A, wheat germ agglutinin and Jacalin from follicular fluid of women with PCOS and controls undergoing in vitro fertilization. Peptides generated by trypsin digestion were labeled with isobaric tags for relative and absolute quantification reagents and analyzed by liquid chromatography tandem mass spectrometry. We identified 10 differentially expressed glycoproteins, in the follicular fluid of women with PCOS compared to control. Two important differentially expressed proteins- SERPINA1 and ITIH4, were consistently upregulated and downregulated respectively, upon validation by Western blotting in follicular fluid and real-time polymerase chain reaction in granulosa cells. These proteins play a role in angiogenesis and extracellular matrix stabilization which are vital for follicle maturation. In conclusion, comparative glycoprotein profiling of follicular fluid from women with PCOS and controls revealed altered expression of proteins which may contribute to defects in follicle development in PCOS pathophysiology.

Project description:Polycystic ovary syndrome (PCOS) is an endocrine disease with metabolic disorders, and patients with PCOS often have impaired reproductive potential. Examining the composition of follicular fluid (FF) in these patients can help identify the specific changes that may impact oocyte quality and potentially cause infertility. This study sought to examine the differential variations in the metabolic composition of FF among normal-weight patients with PCOS and controls and to identify potential biomarkers that may offer insights into the early identification and management of these patients. We collected FF samples from 45 normal-weight women with PCOS and 36 normal-weight controls without PCOS who were undergoing in vitro fertilization-embryo transfer. Untargeted metabolomic study of collected FF from infertile women was performed using high-performance liquid chromatography–tandem spectrometry (LC-MS). The tendency of the two groups to separate was demonstrated through multivariate analysis. Univariate analysis and variable importance in the projection were used to screen out differential metabolites. Metabolic pathway analysis was conducted using the Kyoto Encyclopedia of Genes and Genomes (KEGG), and a diagnostic model was established using the random forest algorithm.

In this experiment neutrophil proteomes were investigated after migration towards JIA synovial fluid in an in vitro model of a synovial membrane, compared to neutrophils incubated in synovial fluid without migration. The migration model consisted of transwell inserts with human knee synoviocytes on the undersides and HMEC endothelial cells on the insides, placed in wells containing medium with 10 % JIA synovial fluid.

Project description:CpG-ODN demonstrated anti-tumor activity in IGROV-1 ascites tumor-bearing mice. To evaluate if soluble molecules present in ascitic fluid, presumably released by innate immune cells via TLR9 stimulation, is directly involved with gene expression modulation a microarray experiment was performed. IGROV-1 human ovarian carcinoma cells were adapted to growth intraperitoneally (i.p.) and maintained by serial i.p. passages of ascitic cells into healthy mice as described. Mice were injected i.p. with 2.5 x 106 ascitic cells in 0.2 ml of saline and treated starting 10-11 days later, when mice showed evident and established ascites, with CpG-ODN [phosphorothioated ODN1826 (5’-TCCATGACGTTCCTGACGTT-3’), TriLink Biotechnologies (San Diego, CA, USA)]. Delivered i.p. at a dose of 20 µg/mouse for 3 consecutive days or 1 time, control mice received saline (3 mice/group). Twenty-four hours after the last treatment with saline or CpG-ODN, ascites-bearing mice were sacrificed by cervical dislocation. Ascitic fluid was collected using a heparinized syringe and the volume recorded. The fluid was transferred to a centrifuge tube maintained on ice. After centrifugation, supernatant was removed and stored at -80°C for in vitro experiments. The ovarian cancer cell line IGROV-1 was obtained from the ATCC (Rockville, MD). Cells were routinely maintained in RPMI medium 1640 (Sigma, St. Louis, MO) supplemented with 10% FCS (Sigma) and 2 mM glutamine (Cambrex, East Rutherford, NJ). Cells were maintained at 37°C in a water-saturated atmosphere of 5% CO2 in air. 1x106 IGROV-1 cells were seeded in 6-well plates and, after seeding, cells were treated with 2 ml of culture medium in the presence of ascites from saline-treated mice or CpG-ODN-treated mice (ratio medium:ascites, 1:1) for 24 hours. At the end of treatment, cells were collected and RNA extracted.

According to the KEGG pathway analysis, these differential metabolites primarily participated in the metabolism of glycerophospholipids and the biosynthesis of steroid hormones. Based on the biomarker combination of the top 10 metabolites, the area under the curve value was 0.805. The concentrations of prostaglandin E2 in the FF of individuals with PCOS exhibited an inverse association with the proportion of high-quality embryos (p < 0.05).ConclusionsOur research identified a distinct metabolic profile of the FF from normal-weight women with PCOS. The results offer a broader comprehension of the pathogenesis and advancement of PCOS, and the detected differential metabolites could be potential biomarkers and targets for the treatment of PCOS.
