Project description:The GNHS was an onging community-based prospective cohort study.In GNHS,a total of 4,048 Chinese participants aged 40-75 years living in the urban area of Guangzhou, China, for at least 5 years were recruited between 2008-2013. Participants were followed up every three years. During the period 2008-2019, blood samples were collected at up to three time points. In 2014-2019 follow-up visits, stool samples were collected at up to two time points.The information of diet, sociodemographic factors, lifestyle factors, and medications, etc. was collected at the baseline and follow-up visits. The aim of this study was to investigate the relationships among human nutrition, enviromental factors, gut microbiome and human diseases.

Project description:Comprehensive data on lifestyle-related modifiable factors, sociodemographic, anthropometric, economic, biochemical, and genetic markers related to the occurrence of cardiometabolic diseases as part of the observational cross-sectional survey (2015 Health Survey of Sao Paulo with Focus on Nutrition (2015 ISA-Nutrition), a population-based study. Data of 805218 SNPs for 841 individuals was genotyped using the Axioma 2.0 Precision Medicine Research Array in the Thermo Fisher Scientific laboratory (Affymetrix Inc, Santa Clara, CA).

Enhanced photorespiratory and TCA pathways by elevated CO2 to manage ammonium nutrition in tomato leaves

Project description:This work aims to study wether the increment of the atmospheric carbon dioxide (CO2) concentration, in the context of climate change, will potentially allow plants to better face ammonium nutrition. Tomato (Solanum lycopersicum L.) plants were grown for 4 week with 15 mM of nitrogen, supplied as nitrate or ammonium, in conditions of ambient (aCO2, 400 ppm) or elevated CO2 (eCO2, 800 ppm) atmosphere. Transcription profiling by array was carried out in leavesfor the four growth conditions assayed and gene expression comparisons were done between N sources and CO2 conditions: i) genes differentially expressed in response to the atmospheric CO2 concentration (eCO2 vs aCO2) under nitrate or ammonium nutrition; ii) genes differentially expressed in response to the N source (ammonium vs nitrate) at aCO2 or eCO2. 3 biological replicates for each growth condition were analysed.

Direct RNA ONT sequencing of pine roots under ammonium nutrition

Project description:Epitranscriptomics modifications constitute a gene expression checkpoint in all living organism including plants. Considering the relevance of nitrogen nutrition and metabolism for the correct plant growth and development, it can be hypothesized that epitranscriptome changes must regulate every biological process in plants including nitrogen nutrition. In the present work, the epritranscriptomics changes in maritime pine roots caused by ammonium nutrition have been monitored through direct RNA sequencing using Oxford Nanopore Technology. The main transcriptome responses to ammonium nutrition affected to transcripts involved in nitrogen and carbon metabolisms, defense response, hormone synthesis and signaling, and translation. Additionally to a global detection of epitranscriptomics marks, the m6A deposition and its dynamics have been identified, which seems to be important regulators of translation when compared with the proteomic profiles of the same samples. In this sense, the obtained results suggest that protein translation is finely regulated through the epitranscriptomics marks maybe through changes in mRNA polyA length, transcript amount and ribosome protein composition. The multiomics results in the present study suggest that the epitranscriptome must modulate the responses to development and environmental changes, including ammonium nutrition, through buffering, filtering and focusing the final products of the gene expression.

Maternal gestational nutrition perturbs offspring sperm epigenome in sheep [ncRNA-seq]

Project description:There is a growing body of evidence that inadequate maternal nutrition during gestation can have immediate and life-long effects on offspring. However, little is known about the reproductive effects of maternal gestational nutrition in offspring males. Here, using a sheep model of poor maternal nutrition (restricted- or over-feeding) during gestation, we found that poor maternal gestational nutrition does not affect semen characteristics (i.e. volume, sperm concentration, pH, sperm motility, sperm morphology) and scrotal circumference in offspring. However, by evaluating associations between poor maternal gestational nutrition and altered small non-cording RNAs (sncRNAs) and DNA methylation in offspring sperm, we demonstrated that poor maternal gestational nutrition alters sperm sncRNA composition and expression. Whole genome bisulfite sequencing further identified genomic regions with increased or decreased DNA methylation in sperm in response to poor maternal gestational nutrition. These findings imply that maternal diet-induced epigenetic errors can accumulate in sperm to worsen developmental outcomes of future generations.
