Project description:Adaptation of long-living forest trees to respond to environmental changes is essential to secure their performance under adverse conditions. Water deficit is one of the most significant stress factors determining tree growth and survival. Maritime pine (Pinus pinaster Ait.), the main source of softwood in southwestern Europe, is subjected to recurrent drought periods which, according to climate change predictions for the years to come, will progressively increase in the Mediterranean region. The mechanisms regulating pine adaptive responses to environment are still largely unknown. The aim of this work was to go a step further in understanding the molecular mechanisms underlying maritime pine response to water stress and drought tolerance at the whole plant level. A global transcriptomic profiling of roots, stems, and needles was conducted to analyze the performance of siblings showing contrasted responses to water deficit from an ad hoc designed full-sib family. Although P. pinaster is considered a recalcitrant species for vegetative propagation in adult phase, the analysis was conducted using vegetatively propagated trees exposed to two treatments: well-watered and moderate water stress. The comparative analyses led us to identify organ-specific genes, constitutively expressed as well as differentially expressed when comparing control versus water stress conditions, in drought-sensitive and drought-tolerant genotypes. Different response strategies can point out, with tolerant individuals being pre-adapted for coping with drought by constitutively expressing stress-related genes that are detected only in latter stages on sensitive individuals subjected to drought.

Project description:Bark thickness is a key structural feature in woody plants in the protection against fire. We used 19 provenances of Pinus halepensis, an obligate-seeder species, in a replicated common garden at two environments contrasting in water availability to assess the interacting effects of site environment and population in the relative allocation to bark, expecting lower allocation at the drier site. Secondly, given the average fire frequency, we analyzed whether trees reached the critical absolute thickness soon enough for population persistence via aerial seed bank. Our analyses indicated that trees at the moister site allocated a rather fixed quantity of resources independent of tree size, and almost all populations reached critical absolute bark thickness to eventually survive fire. In contrast, at the drier site allocation to bark reduced with tree size, and most populations did not reach the critical bark thickness. Populations from areas with higher fire frequency had thicker basal bark, while those from areas with severe droughts and short vegetative periods, had thinner bark. In conclusion, drought-stressed trees have a higher risk to die from fires before achieving reproduction and building a sufficient aerial seed bank.

Project description:Animal longevity widely differs across species, and even individuals from the same species may exhibit different rates of ageing. In different species, the rate at which individuals actually age is related to the level of their social interactions, but this was still not known in ants. In a given colony, ant individuals are close genetic relatives, exhibit very different behaviours and a contrasted lifespan according to the Caste. Such characteristics constitute main advantages to study relationships between sociality and ageing. Therefore, the aim of this study was to characterize differences in the proteome of Lasius niger queens versus that of domestic and foraging workers. Proteomic data were put in relation with the behaviour of individuals from the three Castes. Hence, it was found that sociality correlates with ant longevity, with i) social immunity enabling the queen to mainly invest in soma protection, and ii) marked exposition of workers to the environment and nutrients inducing metabolic pathways that reduce their lifespan.

Project description:Background and aimsThe study of local adaptation in plant reproductive traits has received substantial attention in short-lived species, but studies conducted on forest trees are scarce. This lack of research on long-lived species represents an important gap in our knowledge, because inferences about selection on the reproduction and life history of short-lived species cannot necessarily be extrapolated to trees. This study considers whether the size for first reproduction is locally adapted across a broad geographical range of the Mediterranean conifer species Pinus pinaster. In particular, the study investigates whether this monoecious species varies genetically among populations in terms of whether individuals start to reproduce through their male function, their female function or both sexual functions simultaneously. Whether differences among populations could be attributed to local adaptation across a climatic gradient is then considered.MethodsMale and female reproduction and growth were measured during early stages of sexual maturity of a P. pinaster common garden comprising 23 populations sampled across the species range. Generalized linear mixed models were used to assess genetic variability of early reproductive life-history traits. Environmental correlations with reproductive life-history traits were tested after controlling for neutral genetic structure provided by 12 nuclear simple sequence repeat markers.Key resultsTrees tended to reproduce first through their male function, at a size (height) that varied little among source populations. The transition to female reproduction was slower, showed higher levels of variability and was negatively correlated with vegetative growth traits. Several female reproductive traits were correlated with a gradient of growth conditions, even after accounting for neutral genetic structure, with populations from more unfavourable sites tending to commence female reproduction at a lower individual size.ConclusionsThe study represents the first report of genetic variability among populations for differences in the threshold size for first reproduction between male and female sexual functions in a tree species. The relatively uniform size at which individuals begin reproducing through their male function probably represents the fact that pollen dispersal is also relatively invariant among sites. However, the genetic variability in the timing of female reproduction probably reflects environment-dependent costs of cone production. The results also suggest that early sex allocation in this species might evolve under constraints that do not apply to other conifers.

Project description:In Mediterranean environments in western North America, historic fire regimes in frequent-fire conifer forests are highly variable both temporally and spatially. This complexity influenced forest structure and spatial patterns, but some of this diversity has been lost due to anthropogenic disruption of ecosystem processes, including fire. Information from reference forest sites can help management efforts to restore forests conditions that may be more resilient to future changes in disturbance regimes and climate. In this study, we characterize tree spatial patterns using four-ha stem maps from four old-growth, Jeffrey pine-mixed conifer forests, two with active-fire regimes in northwestern Mexico and two that experienced fire exclusion in the southern Sierra Nevada. Most of the trees were in patches, averaging six to 11 trees per patch at 0.007 to 0.014 ha(-1), and occupied 27-46% of the study areas. Average canopy gap sizes (0.04 ha) covering 11-20% of the area were not significantly different among sites. The putative main effects of fire exclusion were higher densities of single trees in smaller size classes, larger proportion of trees (? 56%) in large patches (? 10 trees), and decreases in spatial complexity. While a homogenization of forest structure has been a typical result from fire exclusion, some similarities in patch, single tree, and gap attributes were maintained at these sites. These within-stand descriptions provide spatially relevant benchmarks from which to manage for structural heterogeneity in frequent-fire forest types.

Project description:Understanding tree recruitment is needed to forecast future forest distribution. Many studies have reported the relevant ecological factors that affect recruitment success in trees, but the potential for genetic-based differences in recruitment has often been neglected. In this study, we established a semi-natural reciprocal sowing experiment to test for local adaptation and microenvironment effects (evaluated here by canopy cover) in the emergence and early survival of maritime pine (Pinus pinaster Aiton), an emblematic Mediterranean forest tree. A novel application of molecular markers was also developed to test for family selection and, thus, for potential genetic change over generations. Overall, we did not find evidence to support local adaptation at the recruitment stage in our semi-natural experiment. Moreover, only weak family selection (if any) was found, suggesting that in stressful environments with low survival, stochastic processes and among-year climate variability may drive recruitment. Nevertheless, our study revealed that, at early stages of recruitment, microenvironments may favor the population with the best adapted life strategy, irrespectively of its (local or non-local) origin. We also found that emergence time is a key factor for seedling survival in stressful Mediterranean environments. Our study highlights the complexity of the factors influencing the early stages of establishment of maritime pine and provides insights into possible management actions aimed at environmental change impact mitigation. In particular, we found that the high stochasticity of the recruitment process in stressful environments and the differences in population-specific adaptive strategies may difficult assisted migration schemes.

Project description:Drought stress is one of the main factors influencing the growth and development of an organism. Auricularia fibrillifera has strong dessication resistance. In A. fibrillifera under dessication-stress, the melanin content of fruiting bodies elevated significantly by >10-fold compared with the control. Folate content also increased sharply but decreased significantly after rehydration, and amino acid and biotin levels increased by 40.11% and 22.14%, respectively. In proteomic analysis, 1,572 and 21 differentially abundant proteins (DAPs) were identified under dessication-stress and rehydration, respectively. A large number of DAPs were annotated in “amino acid metabolism,” “carbohydrate metabolism,” and “translation” pathways, and the DAPs related to osmotic regulation and antioxidant enzymes were significantly increased in abundance. Transcriptome-proteome association analysis showed that most DAPs (30) were annotated in the “biosynthesis of antibiotics” pathway. DAPs and corresponding differentially expressed genes were all up-regulated in the “biotin biosynthesis” pathway and associated with “folate biosynthesis” and “phenylalanine, tyrosine, and tryptophan biosynthesis.” In the analysis of protein–protein interactions, the DAPs annotated in the “phenylalanine, tyrosine, and tryptophan biosynthesis” pathway had the strongest interactions with other DAPs. These enriched pathways could enhance amino acid, folate, biotin, and melanin levels during desiccation stress, which is consistent with the physiological data (amino acid, folate, biotin, and melanin contents). In addition, many DAPs related to the cytoskeleton were significantly increased in abundance under dessication-stress. Physiological and transcriptome data were in agreement with proteomic results. This work provides valuable insight into the dessication-tolerant mechanisms of A. fibrillifera.
