Project description:Adaptation to hypoxia is a complicated and important physiological course for organisms, but the genetic mechanism underlying the adaptation is not fully understood yet. Tibetan Chicken (T), an indigenous chicken breed in China which inhabit in high areas with an altitude above 2,900 meters. Shouguang Chicken(S) and Dwarf Recessive White Chicken (DRW), two lowland chicken breeds, were used as control groups. The heart was the first functional organ to develop during the embryonic development. Furthermore, the heart is an efficient energy converter utilizing the most appropriate fuel for a given environment. Therefore, GeneChip® Chicken Genome Array was employed to identify the differentially expressed genes in embryonic hearts of Tibetan Chicken and two lowland chicken breeds in both hypoxic and normoxic incubating environments with a genome wide profile. Project description:The shift from a hunter-gatherer (HG) to an agricultural (AG) mode of subsistence is believed to have been associated with profound changes in the burden and diversity of pathogens across human populations. Yet, the extent to which the advent of agriculture impacted the evolution of the human immune system remains unknown. Here we present a comparative study of variation in the transcriptional responses of peripheral blood mononuclear cells (PBMCs) to bacterial and viral stimuli between the Batwa, a rainforest hunter-gatherer, and the Bakiga, an agriculturalist population from Central Africa. We observed increased divergence between hunter-gatherers and farmers in the transcriptional response to viruses compared to that for bacterial stimuli. We demonstrate that a significant fraction of these transcriptional differences are under genetic control, and we show that positive natural selection has helped to shape population differences in immune regulation. Unexpectedly, we found stronger signatures of recent natural selection in the rainforest hunter-gatherers, which argues against the popularized notion that shifts in pathogen exposure due to the advent of agriculture imposed radically heightened selective pressures in agriculturalist populations.

Project description:Astrocytes may give raise to glioma, the most frequent primitive CNS tumours. TGFa, an EGF family member, is trophic for both astrocytes and gliomas cells and is frequently over-expressed in the early stages of glioma progression. Although its sole deregulation is insufficient to lead to cancerous transformation of astrocytes, it triggers their conversion into neural progenitor-like cells. We determined whether astrocytes converted into neural progenitor-like cells are more sensitive than their mature counterparts to cancerous transformation using gamma irradiation. Control and TGFa-treated astrocytes had identical cytogenomic profiles. Irradiation did not modify the lifespan of mouse astrocytes cultivated in serum-free medium. On the opposite TGFa-treated astrocytes were immortalized upon irradiation. Their ability to form colonies in methylcellulose medium, their cytogenomic anomalies, and the formation of high-grade glioma-like tumours after grafting into mice CNS testified to their cancerous transformation. Sham-irradiated TGFa-treated astrocytes displayed no evidence of transformation. These results demonstrate that instability of the mature astrocyte phenotype due to a single change in their environment is sufficient to sensitize them to an oncogenic stress. They allow proposing that TGFa does not only favour growth and survival of established gliomas but participates also to the earliest stages of their genesis. Project description:Tibetan chickens exhibit specific adaptations to high-altitude conditions compared with their lowland counterparts. To illustrate the genetic mechanisms of such adaptations in highland chickens, the genomes of four highland and four lowland chicken populations were resequenced. Our results showed that genes under positive selection in highland populations were related to cardiovascular and respiratory system development, DNA repair, response to radiation, inflammation, and immune response, indicating a strong adaptation to oxygen scarcity and high-intensity solar radiation. The distribution of allele frequencies of non-synonymous single nucleotide polymorphisms between highland and lowland populations was also analyzed by chi-square test. The results showed that several differentially distributed genes with missense mutations were enriched in several functional categories, especially in blood vessel development, which were related to adaptations to hypoxia and intense radiation. RNA sequencing also revealed that several differentially expressed genes were enriched in gene ontology terms related to blood vessel and respiratory system development. Additionally, an evident admixture found in Tibetan chickens suggested a history of introgression from lowland gene pools. Overall, our data provided new insights into the unique adaptation of highland animals to extreme environments.
