Project description:Large quantities of kitchen waste are produced in modern society and its disposal poses serious environmental and social problems. The aim of this study was to isolate degradative strains from kitchen waste and to develop a novel and effective microbial agent. One hundred and four strains were isolated from kitchen waste and the 84 dominant strains were used to inoculate protein-, starch-, fat- and cellulose-containing media for detecting their degradability. Twelve dominant strains of various species with high degradability (eight bacteria, one actinomycetes and three fungi) were selected to develop a compound microbial agent "YH" and five strains of these species including H7 (Brevibacterium epidermidis), A3 (Paenibacillus polymyxa), E3 (Aspergillus japonicus), F9 (Aspergillus versicolor) and A5 (Penicillium digitatum), were new for kitchen waste degradation. YH was compared with three commercial microbial agents-"Tiangeng" (TG), "Yilezai" (YLZ) and Effective Microorganisms (EM), by their effects on reduction, maturity and deodorization. The results showed that YH exerted the greatest efficacy on mass loss which decreased about 65.87% after 14 days. The agent inhibited NH3 and H2S emissions significantly during composting process. The concentration of NH3 decreased from 7.1 to 3.2ppm and that of H2S reduced from 0.7 to 0.2ppm. Moreover, E4/E6 (Extinction value460nm/Extinction value665nm) of YH decreased from 2.51 to 1.31, which meant YH had an obvious maturity effect. These results highlighted the potential application of YH in composting kitchen waste.
Project description:This study measured the total quantity and composition of waste generated in a large, New York City (NYC) hospital kitchen over a one-day period to assess the impact of potential waste diversion strategies in potential weight of waste diverted from landfill and reduction in greenhouse gas (GHG) emissions. During the one-day audit, the hospital kitchen generated 1515.15 kg (1.7 US tons) of solid waste daily or 0.23 kg of total waste per meal served. Extrapolating to all meals served in 2019, the hospital kitchen generates over 442,067 kg (487 US tons) of waste and emits approximately 294,466 kg of CO2e annually from waste disposal. Most of this waste (85%, 376,247 kg or 415 US tons annually) is currently sent to landfill. With feasible changes, including increased recycling and moderate composting, this hospital could reduce landfilled waste by 205,245 kg (226 US tons, or 55% reduction) and reduce GHG emissions by 189,025 kg CO2e (64% reduction). Given NYC's ambitious waste and GHG emission reduction targets outlined in its OneNYC strategic plan, studies analyzing composition, emissions, and waste diversion potential of large institutions can be valuable in achieving city sustainability goals.
Project description:We report here the complete genome sequence of Bacillus subtilis subsp. subtilis strain IITK SM1, isolated from kitchen waste compost. We have sequenced the whole genome of this strain to identify and characterize the enzymes that participate in efficient composting activity.
Project description:Background:An economical strategy for producing microalgae as biofuel feedstock is driven by the freshwater and nutrients input. In this study, seawater was applied to limnetic algal cultivation and the behavior of algae in seawater media was observed including growth, lipid synthesis, and ultrastructure. To make seawater cater algae, a kind of wastewater, anaerobically digested effluent from kitchen waste (ADE-KW), was used as nutrient sources. Results:Pure seawater cannot support the growth demand of freshwater microalga, due to high salinity and lack of nutrients. However, it is the conditions triggered the algae to synthesize lipids of 60%, double of lipid content in standard medium BG11. Introducing 3 or 5% ADE-KW (volume percentage) into seawater made algal growth reach the level attained in BG11, while lipid content compared favourably with the level (60%) in pure seawater. This method achieved the goal of fast growth and lipid accumulation simultaneously with the highest lipid productivity (19 mg/L ?day) at the exponential stage, while BG11 obtained 10.55 mg/L ?day at the stationary stage as the highest lipid productivity, almost half of that in seawater media. Moreover, the condition for highest lipid productivity enlarged algal cells compared to BG11. Under the condition for highest lipid productivity, Chlorella sorokiniana SDEC-18 had enlarged cells and increased settling efficiency compared to BG11, which facilitated harvest in an energy saving way. Conclusions:The results suggested that combining seawater with ADE-KW to cultivate microalgae had a double function: nutrients and water for algal growth, and high salinity for stimulating lipid accumulation. If this technology was operated in practice, freshwater and non-waste nutrient consumption would be completely obviated.

Project description:The production of insect biomass from organic waste is a major challenge in terms of reducing the environmental impacts of waste and maintaining feed and food security. The feasibility of the co-conversion of soybean curd residue (SCR) and kitchen waste (KW) to breed black soldier fly (BSF, Hermetia illucens) larvae was evaluated so as to enhance biomass conversion efficiency and supply animal feed and allow it to be used in biodiesel production. Co-digestion was found to significantly increase larval yield, bioconversion rate, and bioaccumulation of lipid. Partial least squares regression showed that the conversion of 30% SCR with 70% KW is an appropriate proportion. The appropriate performance parameters of BSF were: survival rate (98.75%), prepupal rate (88.61%), larval biomass (30.32 g fresh and 11.38 g dry mass), bioconversion rate (18.45%), efficiency conversion of ingested food (ECI) (28.30%), and FCR (2.51). Our results show that conversion of mixtures (e.g., SCR with KW) by BSF larvae (BSFL) could play an important role in various organic materials management.
Project description:Based on phylogenetic analysis of sequence data, Aspergillus section Usti includes 21 species, inclucing two teleomorphic species Aspergillus heterothallicus (= Emericella heterothallica) and Fennellia monodii. Aspergillus germanicus sp. nov. was isolated from indoor air in Germany. This species has identical ITS sequences with A. insuetusCBS 119.27, but is clearly distinct from that species based on ?-tubulin and calmodulin sequence data. This species is unable to grow at 37 °C, similarly to A. keveii and A. insuetus. Aspergillus carlsbadensis sp. nov. was isolated from the Carlsbad Caverns National Park in New Mexico. This taxon is related to, but distinct from a clade including A. calidoustus, A. pseudodeflectus, A. insuetus and A. keveii on all trees. This species is also unable to grow at 37 °C, and acid production was not observed on CREA. Aspergillus californicus sp. nov. is proposed for an isolate from chamise chaparral (Adenostoma fasciculatum) in California. It is related to a clade including A. subsessilis and A. kassunensis on all trees. This species grew well at 37 °C, and acid production was not observed on CREA. The strain CBS 504.65 from soil in Turkey showed to be clearly distinct from the A. deflectus ex-type strain, indicating that this isolate represents a distinct species in this section. We propose the name A. turkensis sp. nov. for this taxon. This species grew, although rather restrictedly at 37 °C, and acid production was not observed on CREA. Isolates from stored maize, South Africa, as a culture contaminant of Bipolaris sorokiniana from indoor air in Finland proved to be related to, but different from A. ustus and A. puniceus. The taxon is proposed as the new species A. pseudoustus. Although supported only by low bootstrap values, F. monodii was found to belong to section Usti based on phylogenetic analysis of either loci BLAST searches to the GenBank database also resulted in closest hits from section Usti. This species obviously does not belong to the Fennellia genus, instead it is a member of the Emericella genus. However, in accordance with the guidelines of the Amsterdam Declaration on fungal nomenclature (Hawksworth et al. 2011), and based on phylogenetic and physiological evidence, we propose the new combination Aspergillus monodii comb. nov. for this taxon. Species assigned to section Usti can be assigned to three chemical groups based on the extrolites. Aspergillus ustus, A. granulosus and A. puniceus produced ustic acid, while A. ustus and A. puniceus also produced austocystins and versicolorins. In the second chemical group, A. pseudodeflectus produced drimans in common with the other species in this group, and also several unique unknown compounds. Aspergillus calidoustus isolates produced drimans and ophiobolins in common with A. insuetus and A. keveii, but also produced austins. Aspergillus insuetus isolates also produced pergillin while A. keveii isolates produced nidulol. In the third chemical group, E. heterothallica has been reported to produce emethallicins, 5'-hydroxyaveranthin, emeheterone, emesterones, 5'-hydroxyaveranthin.

Three groups of samples were considered: P group (three replicate cultures with OVWPE extract diluted in ethanol 20%), E group (three replicate cultures with ethanol 20% alone) C group (three control replicate cultures). 1 ml of VWPE extract (1 mg/ml), 1 ml of ethanol 20% and 1 ml of LB broth were added respectively to the three groups when the OD600 of each culture reached 0.4. Cultures were sampled (2 ml) at the time of treatment (t1), 20 (t2) and 40 (t3) minutes after treatment.
