Project description:Medieval Europe was repeatedly affected by outbreaks of infectious diseases, some of which reached epidemic proportions. A Late Medieval mass burial next to the Heiligen-Geist-Hospital in Lübeck (present-day Germany) contained the skeletal remains of more than 800 individuals who had presumably died from infectious disease. From 92 individuals, we screened the ancient DNA extracts for the presence of pathogens to determine the cause of death. Metagenomic analysis revealed evidence of Salmonella enterica subsp. enterica serovar Paratyphi C, suggesting an outbreak of enteric paratyphoid fever. Three reconstructed S. Paratyphi C genomes showed close similarity to a strain from Norway (1200 CE). Radiocarbon dates placed the disease outbreak in Lübeck between 1270 and 1400 cal CE, with historical records indicating 1367 CE as the most probable year. The deceased were of northern and eastern European descent, confirming Lübeck as an important trading center of the Hanseatic League in the Baltic region.

Quantitative metaproteomics of medieval dental calculus reveals individual oral health status

Project description:The composition of the ancient oral microbiome has recently become possible to investigate by using advanced biomolecular methods such as metagenomics and metaproteomics. This study presents a look at the individuality of the metaproteomes from 22 medieval Danish dental calculus samples. The proteomics data suggest two distinct groups; a healthy and disease-susceptible. Comparison to modern healthy calculus samples supports this hypothesis. The osteological inspections of the samples does not immediately support the grouping made by proteomics data, making us believe that this will add a new and exciting level of information. We identify 3671 protein-groups across all medieval samples and thus expanding the depth of previous studies more than ten times. As a part of future perspective for further depth in these types of samples we performed offline high pH fractionation in combination with TMT labelling and achieved ~30% more protein identifications and reduced costly mass spectrometry time.

Medieval birth girdle stains LC-MSMS

Project description:This project applied a dry non-invasive method to detect palaeoproteomic evidence from stained manuscripts. The manuscript analysed in this study is a medieval parchment birth girdle (Wellcome Collection Western MS. 632) made in England and thought to be used by pregnant women while giving birth. Using a dry non-invasive sampling method we were able to extract both human and non-human peptides from the stains, including evidence for the use of honey, cereals, ovicaprine milk and legumes. In addition, a large number of human peptides were detected on the birth roll, many of which are found in cervico-vaginal fluid. This suggests that the birth roll was actively used during childbirth. This study is the first to extract and analyse non-collagenous peptides from a parchment document using a dry non-invasive sampling method and demonstrates the potential of this type of analysis for stained manuscripts, providing direct biomolecular evidence for active use.

The trade of glass beads in early medieval Illyricum: towards an Islamic monopoly.

Project description:The trade of glass beads has long been assumed to have been under Islamic dominance during the early centuries following the Arab conquest of the Middle East, judged by the prevalence of Islamic beads in the archaeological contexts from Viking Scandinavia to medieval Morocco. This paper explores the impact of the Byzantine-Slavic transition on the use and by extension trade of glass beads in the Balkans from the seventh to the ninth century CE. A series of 48 glass beads and 4 vessel fragments from two excavated sites in modern day Albania have been analysed morphologically, technologically and chemically by LA-ICP-MS. The seventh-century beads from Lezha have typological parallels among central European assemblages and are made from recycled natron-type glass. The presence of a high lead-iron-natron variant is of particular interest as it potentially reflects a regional production. The ninth-century beads from Komani are made from soda-rich plant ash glass from the eastern Mediterranean and Mesopotamia and correspond to an Islamic typology. The chronological and geographical differences are reflected in the distinctive cobalt sources used for the two groups. While the beads from Lezha are coloured with a cobalt not correlated with any particular element, the cobalt source of the Komani samples is associated with zinc, typical of Islamic glass making. It thus appears that the supply of beads during the seventh century when the Balkans were under Slavic occupation relied on regional production and recycled material, and that a long-distance trade with the eastern Mediterranean was revived following the Byzantine re-conquest of the south-eastern Adriatic in the ninth century. Intriguingly, the Albanian finds confirm the Islamic control of the production and trade of glass beads during this period and highlight the mediatory role of the Byzantine Empire.
