Liver transcriptomic and plasma metabolic profile of fattening lambs is modified by feed restriction during the suckling period.

Project description:Purpose: The present study was designed to identify both differentially expressed (DE) genes in the liver tissues of fattening Merino lambs and differences in metabolites accumulated in plasma, thus trying to identify modified metabolic pathways as a consequence of milk restriction during the suckling period. Methods: Twenty-four male Merino lambs were assigned to 2 different groups (n=12 per dietary treatment). The first group (ad libitum, ADL) was kept permanently with the dams whereas the other group (restricted, RES) was milk restricted. When they reached 15 kg of live body weight, all the lambs were offered the same complete pelleted diet at the same level to ensure no differences in dry matter intake. All the lambs were slaughtered with 27 kg. For transcriptomic analysis, 4 liver samples representative from each group (8 samples in total) were selected for RNA sequencing methodology (RNA-seq). Results: 38 DE annotated genes were identified by RNA-seq, with 23 DE genes being down-regulated and 15 up-regulated in the liver of RES relative to the ADL group (P < 0.10). In general, those genes and pathways involved in protein synthesis or protease inhibitors were down-regulated in the RES group, whereas those related to proteolytic degradation were up-regultated, thus suggesting a higher catabolism of proteins in these lambs. Contrarily, RES lambs showed over-expression of xenobiotic metabolism pathways, whereas those genes related to β-oxidation of fatty acids were down expressed. Conclusions: According to the data obtained, early feed restriction during the suckling period of Merino lambs promoted long-term effects on hepatic transcriptomic profile and plasma metabolic profile wich might have modified fatty acids metabolism, catabolism of proteins and detoxification of xenobiotics, thus reducing feed efficiency of fattening period.

Effect of pelleted or non-pelleted high-grain diet on ruminal epithelium in Hu-lambs

Project description:Fifiteen male Hu-lambs were randomly assigned to three groups (n = 5 for each group). Lambs in the control (CON), HG, and HP groups received low-grain nonpelleted diet (30% concentrate), HG diet (70% concentrate), and HP diet containing the same ingredients and nutritions with HG group, respectivley. After 60-day treatment, all the lambs were slaughtered to collect ruminal epihelium samples for transcriptome analysis.

Feed restriction, but not L-carnitine infusion, affects the liver transcriptome with an evident and inhibition of energy production and sterol synthesis and increase of gluconeogenesis in mid-lactating dairy cows

Project description:Feed restriction and L-carnitine infusion are known to affect the liver metabolism of dairy cows. In the present experiment the effects on liver transcriptome of feed restriction and L-carnitine abomasal infusion and the interaction of the two in mid-lactation Holstein dairy cows was assessed. Data clearly indicated a lack of transcriptomics effect by L-carnitine but a strong effect due to feed restriction. The functional analysis identified a overall reduction of cholesterol synthesis and oxidative phosphorylation and data suggested an increase flux toward gluconeogenesis and fatty acid oxidation.

Project description:In this study, we studied the fibrolytic potential of the rumen microbiota in the rumen of 6 lambs separated from their dams from 12h of age and artificially fed with milk replacer (MR) and starter feed from d8, in absence (3 lambs) or presence (3 lambs) of a combination of the live yeast Saccharomyces cerevisiae CNCM I-1077 and selected yeast metabolites. The fibrolytic potential of the rumen microbiota of the lambs at 56 days of age was analyzed with a DNA microarray (FibroChip) targeting genes coding for 8 glycoside hydrolase (GH) families.

The pituitary hormonal changes related to feed efficiency in pigs

Project description:In Poland, conducted for years porcine breeding works focused on improving meat content in the carcass without undertaken to accomplish the more effectiveness of fattening indicators that impact on feed efficiency. At present, feeding and feed conversion became important factors that are especially evident in the countries where the production of fattening pigs is conducted in large farms. In the present study, was attempted to indicate pituitary candidate genes depending on feed conversion ratio (FCR) in native Polish pig breed, Złotnicka White. The whole pituitary transcriptome was sequenced using next-generation sequencing technology. The obtained results show the differences in gene expression for pathways associated with hormonal regulation and also with Notch and Wnt signaling. Moreover, it was observed that worse FCR in Złotnicka pigs was associated with a higher fat level in a carcass and a significant higher level of prolactin expression in the pituitary.

Effect of lifelong carnitine supplementation on plasma and tissue carnitine status, hepatic lipid metabolism and stress signaling pathways and skeletal muscle transcriptome in mice at advanced age

Project description:Despite this large body of evidence from clinical studies that an improvement of carnitine status has beneficial effects on metabolic health, serious safety concerns with carnitine supplementation have been raised based on the recent observation that longterm carnitine supplementation (from weaning to 5 mo of age) in a transgenic mouse model of atherosclerosis (Apoe-/- mice) promotes atherosclerosis through gut microbiota-dependent formation of trimethylamine (TMA) and subsequent hepatic conversion into proatherogenic TMA-N-oxide (TMAO) (Koeth et al., 2013). In addition, Lu et al. (2017) found that carnitine supplementation to normal mice for 3 mo induces oxidative stress, liver inflammation, liver lipid accumulation and even liver damage as well as hypercholesterinemia. However, it has to be mentioned that the carnitine dose administered to the mice in the abovementioned studies was extremely high. In order to address whether safety concerns are founded under conditions of longterm supplementation of a carnitine dose used in clinical trials, the present study aimed to investigate the effect of almost lifelong daily administration of a carnitine-supplemented diet (1 g/kg diet) to mice from weaning to 18 mo of age. Considering a body weight of 50 g for an adult mice and a daily feed intake of 5 g, the daily intake of this diet results in a daily carnitine dose of 100 mg/kg body weight which better reflects the dose used in clinical trials. In order to evaluate possible detrimental effects of carnitine supplementation, we investigated parameters of lipid metabolism and stress signaling pathways in the liver and performed a genome-wide transcriptome analysis of skeletal muscle in the mice at advanced age.
