Project description:Total RNA versus genomic DNA hybridization on custom arrays designed for all Debaryomyces hansenii genes Total RNA was collected in mid-log phase from Debaryomyces hansenii cells grown in rich medium (abbreviated CM, in house recipe). RNA was then converted to cDNA, Cy3-labeled and hybridized competitively against Cy5 labeled genomic DNA from Debaryomyces hansenii.
Project description:In the frame of a multispecies project, Debaryomyces hansenii cells were treated with 0.5 mM sodium selenite. Cells were collected 10, 20, 30, 40, 50, 60, 70 and 80 minutes after the treatment and their transcriptomes were compared to those of mock-treated cultures. Four independent biological replicates were performed.

For this, a control fishmeal-based diet (CD) and experimental diets containing 0.1% of Debaryomyces hansenii were fed to fish for 8 weeks in fresh water. Project description:We found that the fungus, Debaryomyces hansenii (D. hansenii), is enriched in inflamed intestinal tissue from patients of Crohn's disease and its administration to mice impairs colonic wound healing in multiple models of colonic injury and repair. To understand the mechanism, we isolated bone marrow derived macrophages from mice and stimulated them in vitro with a pure isolate of D. hansenii. Total RNA was isolated at multiple time points and assayed for transcriptomic analysis.
