The AGAMOUS-LIKE 67 (AT1G77950) gene encodes a transcription factor that is nuclear-localized, as observed by transiently transformed epidermal onion cells, specifically expressed in seeds and involved in an ABA signaling pathway during seed germination (Fernández-Arbaizar et al. 2012). Project description:Abscisic acid (ABA) is a key hormone for plant growth, development and stress adaptation. Perception of ABA through four types of receptors has been reported. We show here that impairment of ABA perception through the PYR/PYL/RCAR branch reduces vegetative growth and seed production, and leads to a severely open stomata and dramatic ABA insensitive phenotype, even though other branches for ABA perception remain functional. Arabidopsis sextuple mutant impaired in 6 PYR/PYL receptors, namely PYR1, PYL1, PYL2, PYL4, PYL5 and PYL8, was able to germinate and grow even on 100 mM ABA. Whole-rosette stomatal conductance (Gst) measurements revealed that leaf transpiration in the sextuple pyr/pyl mutant was higher than in the ABA-deficient aba3-1 or ABA-insensitive snrk2.6 mutants. The gradually increasing Gst values of plants lacking three, four, five and six PYR/PYLs indicate quantitative regulation of stomatal aperture by this family of receptors. The sextuple mutant lacked ABA-mediated activation of SnRK2s and ABA-responsive gene expression was dramatically impaired as was reported in snrk2.2/2.3/2.6. In summary, these results show that ABA perception by PYR/PYLs plays a major role to regulate seed germination and establishment, basal ABA signaling required for vegetative and reproductive growth, stomatal aperture and transcriptional response to the hormone.

Project description:Summary: The non-protein amino acid beta-aminobutyric acid (BABA) primes Arabidopsis to respond more quickly and strongly to pathogen and osmotic stress. Here, we report that BABA also significantly enhanced acquired thermotolerance in Arabidopsis. This thermotolerance was dependent on heat shock protein 101, a critical component of the normal heat shock response. BABA did not enhance basal thermotolerance under a severe heat shock treatment. No roles for the hormones ethylene and salicylic acid in BABA-induced acquired thermotolerance were identified by mutant analysis. By global gene expression analysis, transcript levels for several transcription factors and DNA binding proteins regulating responses to the stress hormone abscisic acid (ABA) were found to be elevated in BABA-treated relative to water-treated plants. The role of ABA in BABA-induced thermotolerance was complex. BABA-enhanced thermotolerance was partially compromised in the ABA-insensitive mutant, abi1-1, but was augmented in abi2-1. In an unrelated process, BABA, like ABA, inhibited root growth and the level of inhibition was roughly additive in roots treated with both compounds. Root growth of both abi1-1 and abi2-1 was also inhibited by BABA. Unexpectedly, abi1-1 and abi2-1 root growth was inhibited more strongly by combined ABA and BABA treatments than by BABA alone. Our results together with previously published data suggest that BABA is a general enhancer of plant stress resistance and that cross-talk occurs between BABA and ABA signalling cascades. Specifically, the BABA-mediated accumulation of ABA transcription factors without concomitant activation of a downstream ABA response could represent one component of the BABA-primed state in Arabidopsis. Four biological replicates were generated for the 3 genotypes, pyr1pyl1pyl2pyl4pyl5pyl8 sextuple mutant, snrk2.2/2.3/2.6 triple mutant, and Col-0. All the samples were treated with ABA for 3 hours before harvesting. The four Col-0 samples were mix to create an unique reference Col-0 sample. Two comparasion were made, fisrt one, PYR/PYL sextuple mutant versus Col-0 reference sample and second one, SnRK2 triple mutant versus Col-0 reference sample. In each comparasion four biological replicates were made. Replicas number 1 and 2 were labeled with Cy5 for the mutant sample and Cy3 for the Col-0 reference sample, while the other two replicas,#3 and #4, were reversed-labeled.

Arabidopsis Col-0 plants were grown in ProMix HP (Premier Horticulture, Red Hill, PA, USA) for 14 days and the soil was drenched with a 0.25 mM BABA solution 1 day prior to harvesting samples for RNA preparation. These plants were grown in a growth chamber at 21?C with a 14-hr photoperiod. All plants were grown at a light intensity of about 100 ? 150 ?E m-2 s-1. BABA (mixed isomers, Sigma, Milwaukee, WI, USA) was diluted in water at indicated concentration. A biological sample represents a comparison between plants that were water-treated (green channel) to plants drenched with BABA (red channel). Project description:The goal of the RNA sequencing is to find out the signal pathway the VQ28 gene involved in. Uninfected 30-day-old detached Arabidopsis rosette leaves of wild type Col-0, knock-out line VQ28KO12 and overexpression line VQ28OE9 were collected to isolate total RNA. Then, we created a final cDNA library and performed the 2×150bp paired-end sequencing on an Illumina Novaseq™ 6000. After removed the low quality bases and undetermined bases, we mapped reads to the genome. The differentially expressed mRNAs were selected with fold change > 2 or fold change < 0.5 and p value < 0.05, and then analysis GO enrichment and KEGG enrichment to the differentially expressed mRNAs. In comparison with Col-0, constitutively overexpressing VQ28 caused the enrichment of most genes in MAPK signaling pathway and plant hormone signal transduction. Among the 11 genes up-regulated in VQ28OE9 but down-regulated in VQ28KO12, examining their expression by querying the Arabidopsis eFP Browser which contains an extensive collection of gene expression microarray data, six of them were involved in the response to abscisic acid (ABA). qRT-PCR analyses of expression presented that the six ABA-related genes were up-regulated in VQ28OE9 without infection.

Project description:Plant hormone brassinosteroids (BRs) and abscisic acids (ABA) antagonistically regulate many aspects of plant growth and responses. This study analyzes the molecular mechanisms by which regulate the crosstalk between BR and ABA signaling.Various BR deficient and gain-of-function signaling mutants were used to analyze their responses to ABA inhibited primary root growth. RNA sequencing was performed to identify the ABA regulated root genes that are also regulated by BR signaling components.Our result demonstrated that BR signaling negative regulates plant ABA response, and the crosstalk is mediated by BIN2 and BZR1. RNA sequencing has identified subsets of ABA responsive root genes that were regulated by BIN2 and/or BZR1. ChIP-qPCR and EMSA assays showed that BZR1 could bind directly with several G-box cis-elements on ABI5 promoter, suppress the expression of ABI5 and makes plant insensitive to ABA.These data demonstrated that ABI5 is a BZR1 direct targeted gene. Regulation of ABI5 expression by BZR1 plays important roles in regulating the crosstalk between BR and ABA signaling pathways.

Project description:Abscisic acid (ABA) plays a key role in plant development and responses to abiotic stress. A wide number of regulatory mechanisms on ABA perception and signaling are known, including transcriptional regulation and post-translational regulations; however, less to know about the post-transcriptional regulations. In this work, we have found that SKIP, a splicing factor in spliceosome, positively regulates ABA signal transduction. Mutation of SKIP, skip-1, confers ABA insensitive phenotype in seed germination, root growth, and ABA-responsive gene expression. Transformation of SKIP genomic DNA to skip-1 is able to recover its defects. SKIP binds to the pre-mRNA of genes in ABA signaling, such as PYL8, to regulate their splicing. The alternative splicing of PYL7, PYL8, ABF2, and ABI5 in ABA pathway is disrupted by skip-1, reducing the mRNA levels of them. The abnormal splicing of PP2Cs activates their transcription by skip-1, suggesting there is a feedback regulation for PP2Cs caused by skip-1. The down-regulation of PYL ABA receptors, ABF2 and ABI5 ABA response transcription factors, and the up-regulation of PP2Cs through alternative splicing reduce the plant responses to ABA in skip-1. SKIP is required for ABA-mediated genome-wide alternative splicing. ABA treatment enhances the novel splicing events in WT genome-widely. The novel alternative splicing events are increased by skip-1 in the absence and present of ABA. Our results first reveal a principle on how a splicing factor affects the ABA signaling.
