Project description:We found that approximately 200 genes were differentially expressed between CD11b+ lung-migratory dendritic cells (CD11b+ mDCs) from adult and infant mice treated with house dust mite allergen + LPS, and that the TNFa-via NFkB signaling pathway was significantly enriched in CD11b+ mDCs from adults relative to those from infants. Specifically, out of the 200 genes in the hallmark TNFa-via NFkB signaling pathway that were analyzed, 31 genes were significantly downregulated in CD11b+ mDCs from infants compared to CD11b+ mDCs from adults.

Global transcriptional profiles of WT and HOIL-1L-/- BMDMs in response to LPS and TNFa

Project description:HOIL-1L is an essential member of the linear ubiquitin assembly complex (LUBAC), which targets Nemo for linear ubiquitination during NF-kB activation in response to a variety of simuli, including LPS and TNFa treatment. HOIL-1L has also been suggested to function as a transcription factor. Here we analyzed changes in the global transcriptional profiles of primary bone marrow derived macrophage (BMDMs) from WT and HOIL-1L-/- mice upon treatment with NF-kB activating simuli. BMDMs were differentiated for 1 week and then treated with mock (untreated), 10ng/ml LPS, or 1ng/ml TNFa for 4 hours. RNA was immediately collected and analyzed in biological duplicates by the Agilent 8x60 mouse array.

Project description:To understand the transcripts induced by TNFa and how these may interact in inflammatory networks, the transcriptome of HUCECs stimulated with TNFa was examined in 3 different pools each cosisting of 10 different HUVEC isolates. In the study present here, HUVEC isolates from 3 pools of 10 different individuals were cultured until passage 4 in fully supplemented growth conditions. Passage 4 HUVEC isolates were then treated with TNFa (10ng/ml), with RNA extracted at time points 0, 1, 1.5, 2, 4, 5, and 6hrs after TNFa treatment . RNA was extracted and hybridised onto CodeLink microarrays.
