Project description:Sib mating increase homozygosity and can lead to inbreeding depression. Selective pressures have favored the evolution of kin recognition and sib avoidance in mate choice. Such avoidance behavior has been recorded in the parasitoid wasp Venturia canescens where the sex determining system (single-locus complementary sex determination, sl-csd) introduces an extra short-term cost to inbreeding; females preferentially mate with unrelated males. The genetics underlying mate choice of females and kin recognition remains largely unexplored. Here we analyzed the head transcriptomic changes after exposure to distinct stimulus: courtship by unrelated male or courtship by brother, while transcriptome of females alone was used as control. Our results demonstrate that male courtship provokes a major transcriptome reprogramming in females heads. The transcriptomic response is highly dependent on the compatibility of the courting male. We subdivided the socially responsive genes using an integrative gene network analysis, we highlighted genes regulated by courtship, whatever the relatedness of the courting male. We also showed that the regulation of some peculiar group of genes is very specific of compatible male courtship, while others are regulated only by incompatible male courtship. We suggest that those different transcriptomic responses lead to mating decision or sib mating avoidance behavior observed in this species.

Project description:Sib mating increases homozygosity, which therefore increases the risk of inbreeding depression. Selective pressures have favoured the evolution of kin recognition and avoidance of sib mating in numerous species, including the parasitoid wasp Venturia canescens. We studied the female neurogenomic response associated with sib mating avoidance after females were exposed to courtship displays by i) unrelated males or ii) related males or iii) no courtship (controls). First, by comparing the transcriptional responses of females exposed to courtship displays to those exposed to controls, we saw a rapid and extensive transcriptional shift consistent with social environment. Second, by comparing the transcriptional responses of females exposed to courtship by related to those exposed to unrelated males, we characterized distinct and repeatable transcriptomic patterns that correlated with the relatedness of the courting male. Network analysis revealed 3 modules of specific 'sib-responsive' genes that were distinct from other 'courtship-responsive' modules. Therefore, specific neurogenomic states with characteristic brain transcriptomes associated with different behavioural responses affect sib mating avoidance behaviour.

Larval host gene expression study in Drosophila post parasitic wasp-infection

Project description:Upon pathogenic infection, drosophila larval host mounts an immune response. Parasitic wasps inject venom that contain virulence factors during oviposition, which can elicit host immune response, and in some cases, suppress host immune responses altogether. Several microarray experiments have been performed on different classes of parasitic wasps. We wanted to compare how Ganaspis xanthopoda-infected hosts respond compared to other classes of parasitic wasps. Third instar y w larvae from a 2-day egglay were infected with G. xanthopoda for three and six hours, respectively, by introducing waps in petri-dish containing larvae. Controls were handled side-by-side without introducing wasps. Host larvae were immediately dissected, infection confirmed by presence of wasp egg, and frozen in liquid nitrogen and ground in Trizol. RNA was isolated and checked by agarose gel-electrophoresis. Samples were then sent to the Microarray Core Facility at Weill Cornell Medical College.

Transcription profiling of bone marrow derived mast cells from WASP negative and wild type mice to investigate the role of WASP in Kit signalling

Project description:The pleiotropic RTK Kit can provide cytoskeletal signals that define cell shape, positioning and migration, but the underlying mechanisms are less well understood. Here we provide evidence that Kit signals through WASP (Wiskott-Aldrich Syndrome Protein), the central hematopoietic actin nucleation- promoting factor and regulator of the cytoskeleton. KL-mediated gene expression in WT and WASP-deficient BMMCs was compared and revealed that approximately 30% of all Kit-induced changes were WASP-dependent. The results indicate that Kit signaling through WASP is necessary for normal Kit-mediated filopodia formation, cell survival and gene expression and provide new insight in the mechanism how WASP exerts a strong selective pressure in hematopoiesis.

Global gene occupancy by nuclear WASp during T cell development

Project description:To investigate a role of nuclear WASp in T cell development we performed WASp chromatin immunoprecipitation and deep sequencing (ChIP-Seq) in thymocytes and spleen CD4+ T cells. To pre-process raw ChIP-Seq data, the total number of reads were normalized and aligned against the mouse genome. WASp was enriched at transcription start sites of a large number of protein-coding genes. Many of the WASp-enriched genes were associated with RNA Polymerase II-enriched genes and active epigenetic marks of transcription; H3K4m3, H3K9a, H3K27a, and with the epigenetic mark for active enhancers H3K4m1. To study the distribution of overactive WASpI296T in the thymocyte genome and to identify regions enriched in WASpI296T binding, we performed second round of ChIP-Seq analysis using the WASp F-8 antibody. To detect differences in gene enrichment between thymocytes expressing wildtype WASp or WASpI296T, we applied stringent conditions and subtracted common genes between the two samples. Using this approach, we identify 70 WASpI296T-enriched genes. Functional clustering of these genes revealed that WASpI296T was associated with RNA Polymerase II genes in 11 functional groups of genes.thymocytes and spleen CD4+ T cells. WASp was enriched at transcription start sites of a large number of protein-coding genes.

Neurogenomic insights into paternal care and its relation to territorial aggression

Project description:Motherhood is characterized by dramatic changes in brain and behavior, but less is known about fatherhood. Here we report that male sticklebacks – a small fish in which fathers provide care – experience dramatic changes in neurogenomic state as they become fathers. Some genes are unique to different stages of paternal care, some genes are shared across stages, and some genes are added to the previously acquired neurogenomic state. Comparative genomic analysis suggests that some of these neurogenomic dynamics resemble changes associated with pregnancy and reproduction in mammalian mothers. Moreover, gene regulatory analysis identified transcription factors that are regulated in opposite directions in response to a territorial challenge versus during paternal care. Altogether these results show that some of the molecular mechanisms of parental care might be deeply conserved and might not be sex-specific, and suggest that tradeoffs between opposing social behaviors are managed at the gene regulatory level.
