Glucose Shortens the Lifespan of Caenorhabditis elegans by Down-Regulating Aquaporin Gene Expression

Project description:Many studies have addressed the effect of dietary glycemic index on obesity and diabetes, but little is known about its effect on lifespan itself. We found that adding a small amount of glucose to the medium (0.1-2%) shortened the lifespan of C. elegans. Glucose shortened lifespan by inhibiting the activities of lifespan-extending transcription factors that are also inhibited by insulin signaling: the FOXO family member DAF-16 and the heat shock factor HSF-1. This effect involved the down-regulation of an aquaporin glycerol channel, aqp-1. We show that changes in glycerol metabolism are likely to underlie the lifespan-shortening effect of glucose, and that aqp-1 may act cell non-autonomously as a feedback regulator in the insulin/IGF-1 signaling pathway. Insulin down-regulates similar glycerol channels in mammals, suggesting that this glucose-responsive pathway might be conserved evolutionarily. Together these findings raise the possibility that a low-sugar diet might have beneficial effects on lifespan in higher organisms. Refer to individual Series. Characterization of the genes that were regulated by feeding with Lactococcus kimchii

Project description:In the present study, we investigated the effect of Lactococcus kimchii on healthspan using Caenorhabditis elegans as a model animal. The healthspan of the adult C. elegans fed the mixture of E. coli OP50 and Lactococcus kimchii was significantly longer than that of animals fed sole OP50 (control). Transcriptional profiling comparing Lactococcus kimchii- and control-fed animals suggested that genes related to insulin-like signaling pathway were upregulated by Lactococcus kimchii.

Transcriptomics of C. chinensis and E. ulmoides treated aged C. elegans

Project description:The nematode Caenorhabditis elegans was treated with extracts of the Traditional Chinese Medicine plants Cuscuta chinensis and Eucommia ulmoides from the L4 stage. In aged worms (on the 7th and 12th day of adulthood) different health parameters were determined. Besides the prolongation of lifespan, we found that C. chinensis improved the swimming behavior, pharyngeal pumping rate, stress resistance, mechanosensation and memory of aged C. elegans. Furthermore, the extract treatment reduced the autofluorescence, which is a known biomarker of ageing. Thus, we concluded that C. chinensis is an overall healthspan enhancer. In contrast, the E. ulmoides extract specifically enhanced the lifespan and stress resistance of aged C. elegans, but did not improve any other health aspect. To reveal the mechanism behind the healthspan enhancing effects of C. chinensis, the transcriptome of treated and untreated C. elegans on the 12th day of adulthood was analysed. For comparison purposes, E. ulmoides treated nematodes were included in the analysis.

Drug synergy slows ageing and improves healthspan through IGF and SREBP lipid signaling

Project description:Pharmacological interventions directly modulating ageing would extend human healthspan, resulting in dramatic medical and economic benefits. Such interventions should be efficacious in adults and ideally repurpose drugs known to be safe. Here we show that dramatic lifespan extension can be achieved by targeting multiple, evolutionarily conserved ageing pathways using drugs already used in humans. Using this approach in C. elegans, we were able to slow ageing, improve healthspan and minimize trade-offs. We show that daf-2, daf-7 and sbp-1 (mammalian SREBP-1c) interact upstream of changes in lipid metabolism, resulting in increased monounsaturated fatty acid content and lifespan extension. For one combination, we observed evolutionary conservation in fruit flies. To the best of our knowledge, this is the largest lifespan effect ever reported for any adult-onset drug treatment in C. elegans. Drug repurposing approaches, by leveraging drugs already approved for humans, could lead to rapid translation of experimental results to clinical applications.

Project description:Hedgehog morphogen GRD-1 regulates C. elegans growth and metabolism downstream of TOR complex 2. Both Hedgehog (Hh) signaling and target of rapamycin complex 2 (TORC2) are evolutionarily conserved pathways that regulate growth and metabolism. In C. elegans, loss of Hh morphogens often leads to developmental arrest, while loss of essential TORC2 component RICTOR (rict-1) causes delayed development, reduced brood, small size, increased fat, and shortened lifespan. Here we report that knockdown of Hh-related morphogen grd-1 causes developmental acceleration rather than arrest by speeding up molting transitions. Further, we show that RNAi to grd-1 not only suppresses the slow growth of rict-1 mutants, but also normalizes multiple abnormalities downstream of TORC2 including restoration of lifespan to near normal and partial rescue of low brood, small body size, and increased fat. Mechanistically, grd-1 slows growth downstream of TORC2 at least in part by increasing transcription of paraquat mediator 1 (PQM-1) target genes. Given the important role of TORC2 in governance of development, these data implicate grd-1 and pqm-1 as critical executors of organismal growth slowing in response to unfavorable growth conditions downstream of the nutrient sensor TORC2 in C. elegans.
