Enterococcus faecalis EfaR mutant transcriptome profile

Project description:Metal ions are important cofactors of inumerous enzymes and can interfere in many biological processes. Manganese is a particularly relevant metal as it is also involved in protection from oxidative stress and several Mn-dependent regulators have been shown to have a role in bacterial pathogenicity. A putative DNA-binding motif, here called efa motif was discovered in the promoter regions of genes previously shown to be differentially expressed in the presence of excess zinc, manganese and copper ions. This motif led to the finding and study of the genes putatively involved in the maintenance of manganese homeostasis in E. faecalis. In these genes is included the efaCBA operon, regulated by the Mn-dependent regulator EfaR. Other genes with this efa motif are shown to be regulated by EfaR. Regulation of these efa motif genes by EfaR is discussed as well as some clues on metal interaction with EfaR. Experiments mimicking conditions occurring on biological processes in the host showed that an EfaR plays an important role in biological processes crucial for E. faecalis colonization and virulence. Project description:Efficient horizontal gene transfer of the conjugative plasmid pCF10 from Enterococcus faecalis depends on the expression of its Type 4 Secretion System (T4SS) genes, controlled by the PQ promoter. The PQ promoter is strictly regulated, partially to limit cell toxicity caused by overproduction of PrgB, a T4SS adhesin. PrgU plays an important role in regulating this toxicity by decreasing PrgB levels. PrgU has an RNA-binding fold, prompting us to test whether PrgU exerts its regulatory control through binding of prgQ transcripts. We used a combination of in vivo methods to quantify PrgU effects on prgQ transcripts at both single cell and population levels.

Project description:Transcriptional profiling to investigate the effect of drug treatment on the E. faecalis cells. For microarray analysis, E. faecalis OG1RF was grown in FMC medium supplemented with 10 mM glucose to an optical density at 600 nm (OD600) of 0.3 and the cultures were divided in 3 aliquots. One aliquot was collected by centrifugation and immediately frozen (untreated control cells). The other aliquots were treated for 30 or 60 min with 1.25 X the minimum inhibitory concentration (MIC) of vancomycin (10 M-NM-<g ml-1). After an exposure time of 30 or 60 minutes, each of these cultures was also centrifuged and the pellets frozen. RNA was then isolated from each pellet for microarray analysis. This process was repeated 3 additional times, for a total of four replicates of each condition. RNA was extracted from four replicate samples of each condition of interest (control cells grown to OD600 = 0.3 in FMC medium supplemented with 10mM glucose, then treated with vancomycin for 30 or 60 minutes) and labeled with Cy3. For each replicate, labeled RNA was hybridized to slides along with Cy5-labeled reference RNA, extracted from E. faecalis OG1RF cells grown in BHI medium to mid-log.
