Project description:Mycobacterium tuberculosis and M. smegmatis form drug-tolerant biofilms through dedicated genetic programs. In support of a stepwise process regulating biofilm production in mycobacteria, it was shown elsewhere that lsr2 participates in intercellular aggregation, while groEL1 was required for biofilm maturation in M. smegmatis. Here, by means of RNA-Seq, we monitored the early steps of biofilm production in M. bovis BCG, to distinguish intercellular aggregation from attachment to a surface. Genes encoding for the transcriptional regulators dosR and BCG0114 (Rv0081) were significantly regulated and responded differently to intercellular aggregation and surface attachment. Moreover, a M. tuberculosis H37Rv deletion mutant in the Rv3134c-dosS-dosR regulon, formed less biofilm than wild type M. tuberculosis, a phenotype reverted upon reintroduction of this operon into the mutant. Combining RT-qPCR with microbiological assays (colony and surface pellicle morphologies, biofilm quantification, Ziehl-Neelsen staining, growth curve and replication of planktonic cells), we found that BCG0642c affected biofilm production and replication of planktonic BCG, whereas ethR affected only phenotypes linked to planktonic cells despite its downregulation at the intercellular aggregation step. Our results provide evidence for a stage-dependent expression of genes that contribute to biofilm production in slow-growing mycobacteria.

Aggregation adaptive routes revealed by transcriptomic and ultrastructural signatures in Phytophthora parasitica

Project description:Within oomycetes, Phytophthora is a genus of eukaryotic microorganisms encompassing some of the most damaging plant pathogens. It spreads by airborne sporangia or waterborne zoospores. Zoospores perceive plant signals through chemotaxis and electrotaxis, and produce signals to attract other zoospores (autotaxis), resulting in auto-aggregation or biofilm formation on the plant surface. The mechanisms underlying intercellular communication and consequent attraction, adhesion and aggregation are largely unknown. Recent studies demonstrated that in Phytophthora parasitica the perception of K+ gradient induced coordinated motion and rapid aggregation. Based on this model, we combined data mining and ultrastructural microscopy analyses to propose a first definition of molecular events leading to oomycete aggregation. Results indicate that the transcriptome repertoire required for aggregation is fully functional already at this single zoospore stage, before aggregation or encystment. Freely swimming zoospores secrete or harbor proteins implied in aggregation through adhesion and extracellular matrix elaboration. They secrete vesicular and fibrillary material, including fibronectin-like proteins, resulting in intercellular structure formation. Consistently, the signature of transcriptome dynamics during transition from single-cell to aggregate is an upregulation of genes contributing to vesicular trafficking, possibly involved in adhesive proteins and extracellular matrix components mobilization. Moreover, the transcriptome signature suggests that pH homeostasis may contribute to aggregation by acting on both zoospore movement and adhesion.

Identification and characterization of conceptus gene expression patterns during rapid trophoblastic elongation and attachment in the pig will provide a better understanding of the events required for successful implantation and embryonic survival.

Developmental gene regulation by an ancient intercellular communication system in social amoebae

Project description:In the social amoebae (Dictyostelia) quorum sensing system mediates aggregation of single cells into multicellular aggregates by chemotactic movement towards gradients of diffusible molecules known as acrasins. The acrasin of P. violaceum is the unusual dipeptide N-propionyl-gamma-L-glutamyl-L-ornithine-delta-lactam-ethylester, known as glorin. Phylogenetic analysis has indicated that P. violaceum is more related to the most derived group 4 dictyostelids than to the ancient group 2 polysphondylids such as P. pallidum. Nevertheless it has been reported that P. pallidum cells respond to glorin in chemotaxis assays. This has led to the assumption that glorin-based communication may be the most ancient form of intercellular communication that Dictyostelia invented to organize early steps of multicellular development. In this study we show that glorin mediates rapid changes in gene expression at the transition from vegetative growth to aggregation, apparently without pronounced cross-talk with the cyclic AMP-based communication system that coordinates post-aggregation events in this species. We describe glorin-mediated changes in gene expression in the social amoeba Polysphondylium pallidum at the transition from unicellular growth to multicellular development. Comparison of gene expression in growing cells versus cells starving for 2 or 3 hours in the presence or absence of glorin.

Project description:Gene expression changes during biofilm formation processes were investigated. The gene expression was compared at attachment, colony formation and maturation during biofilm formation. At the same time, the gene expressions were also compared with exponential phase and stationary phase in planktonic cells. The gene expression pattern at attachment and colony formation processes showed similar pattern with those in planktonic exponential phase, and the gene expression pattern at maturation process showed similar pattern with those in planktonic stationary phase. During the maturation process, metabolic activities of the cells in the biofilms decreased, and the genes involved in the anaerobic respiration and efflux pumps were induced. The analysis revealed that gene expression pattern was changed and the physiological states were changed dramatically during maturation process in the biofilms. Keywords: time course Affymetrix E. coli antisense genome array was used to compare the gene expression among biofilm formation processes (attachment, colony formation and maturation) and planktonic cells (exponential phase and stationary phase). All samples were grown in MOPS minimal media with 0.2% glucose at 37ºC. Biofilms were grown on glass surface in flow cells (1 x 4 x 40 mm), and samples were taken at 2 h, 24 h and 72 h. Planktonic cell were grown for 6 h (exponential phase) and 24 h (stationary phase). Experiments were repeated 3 times, which resulted in 3 replicates of 5 different samples.
