Project description:To understand how cells communicate with each other, it is essential to define the cellular secretome, a collection of proteins including soluble secreted, unconventionally secreted and proteolytically-shed proteins. Quantitative methodologies to decipher the secretome are challenging, due to the requirement of large cell numbers and abundant serum proteins that interfere with the detection of low-abundant cellular secretome proteins. Here, we have use the highe perfomance secretome-protein-enrichment-with-click-sugars method (hiSPECS) for glyco-secretome analysis. We applied this method to investigate differences of hippocampal and cortical murine neurons. Additionally, we have inhibited the Alzheimer related protease BACE1 to identify potential substrates in the secretome of hippocampal neurons.

Project description:The hippocampus is important for memory formation and is severely affected in the brain with Alzheimer's disease (AD). Since AD brain tissue is available postmortem, our understanding of early pathogenic processes occurring in hippocampi remains speculative. Here, an MS-based proteomic approach was used to analyze free-floating hippocampal spheroids (HSs) enriched in PROX1-positive granule neurons, from induced pluripotent stem cells (iPSCs) of healthy individuals and AD patients. HSs generated from two AD patients carrying variations in amyloid precursor protein (APP) or presenilin 1 (PS1) genes, and their age and gender matched controls.

Project description:The goal of this study is to understand how the specific TERT inhibitor BIBR1532 preconditioning regulates transcriptional reprogramming in mouse neuronal cells. We use CUT-Tag approach to comprehensively map the genomic binding sites of TERT in primary neurons and determine the alteration of is binding profile after BIBR1532 preconditioning. By obtaining over 10 million reads per sample, we generated genome-wide TERT chromatin-binding maps of mouse primary neurons. Under normal conditions, we show that more than 50% of the TERT binding sites were located at the promoter regions, many of them are previously unknown genomic loci. Intriguingly, BIBR1532 preconditioning significantly alters TERT-chromatin binding profile. Out of the total promoter binding sites by TERT, 30.4% (2,837 peaks) and 31.7% (2,954 peaks) are respectively upregulated and downregulated in TERT-binding after BIBR1532 preconditioning. Interestingly, we find that BIBR1532-preconditioned neurons show significant upregulation of promoter binding of TERT to the mitochondrial antioxidant genes. Together, these results identify the previously unknown TERT-binding sites in the mouse primary neurons and demonstrate that BIBR1532 preconditioning confers neuronal ischemic tolerance through TERT-mediated transcriptional reprogramming.

Project description:Exercise has emerged as a powerful variable that can improve cognitive function and delay age-associated cognitive decline and Alzheimer’s disease (AD), however, the underlying mechanisms are poorly understood. To determine if protective mechanisms may occur at the transcriptional level, we used microarrays to investigate the relationship between physical activity levels and gene expression patterns in the cognitively-intact aged human hippocampus. In parallel, hippocampal gene expression patterns associated with Aging and AD were assessed using publicly available microarray data profiling hippocampus from young (20-59 yrs), cognitively-intact aging (73-95 yrs) and age-matched AD cases. To identify “anti-Aging/AD” transcription patterns associated with physical activity, probesets significantly associated with both physical activity and Aging/AD were identified and their directions of expression change in each condition were compared. Remarkably, of the 2138 probesets significant in both datasets, nearly 95% showed opposite transcription patterns with physical activity compared to Aging/AD. The majority (>70%) of these anti-Aging/AD genes showed increased expression with physical activity and decreased expression in Aging/AD. Enrichment analysis of the anti-Aging/AD genes showing increased expression in association with physical activity revealed strong overrepresentation of mitochondrial energy production and synaptic function, along with axonal function and myelin integrity. Synaptic genes were notably enriched for synaptic vesicle priming, release and recycling, glutamate and GABA signaling and synaptic spine plasticity. Anti-Aging/AD genes showing decreased expression in association with physical activity were enriched for transcription-related function. These data suggest that physical activity preserves a more youthful profile in the hippocampus across multiple biological processes, providing a potential molecular foundation for how physical activity can delay age- and AD-related decline of hippocampal function.

Project description:Alzheimer’s disease (AD) is a progressive neurodegenerative disorder that mostly strikes the elderly. However, the exact molecular and cellular pathogenesis of AD, especially the dynamic changes of neurons during disease progression, remains poorly understood. Here we used single-nucleus RNA sequencing (snRNA-seq) to access the transcriptional changes of hippocampal neurons in APP23 mouse model of AD. We performed snRNA-seq using a modified Smart-seq2 technique on 3,280 neuronal nuclei from the hippocampus of young and aged APP23 and control mice and identified four distinct subpopulations. Comparative transcriptional analysis showed multiple changes in different subtypes of hippocampal neurons of APP23 mice in comparison to control mice, as well as the transcriptional changes in these neurons during disease progression. Our findings revealed multiple neuronal subtype-specific transcriptional changes that may lead to targets for future studies of AD.
