Project description:C4 plants frequently experience damaging high light (HL) and high temperature (HT) conditions in native environments, which reduce growth and yield. However, the mechanisms underlying these stress responses in C4 plants have been under-explored, especially the coordination between mesophyll (M) and bundle sheath (BS) cells. We investigated how the C4 model plant Setaria viridis responded to a four-hour HL or HT treatment at the photosynthetic, transcriptomic, and ultrastructural levels. Although we observed a comparable reduction of photosynthetic efficiency in HL- or HT-treated leaves, detailed analysis of multi-level responses revealed important differences in key pathways and M/BS specificity responding to HL and HT. We provide a systematic analysis of HL and HT responses in S. viridis, reveal different acclimation strategies to these two stresses in C4 plants, discover unique light/temperature responses in C4 plants in comparison to C3 plants, and identify potential targets to improve abiotic stress tolerance in C4 crops.

Project description:Increasing photosynthetic efficiency remains a major goal across the globe given its central role in CO2 assimilation. Here, we identified a nuclear genome-residing orphan gene, comprised of 3 exons, two with apparent endophytic origins and the third being a highly conserved fragment of the RIBULOSE BISPHOSPHATE CARBOXYLASE/ OXYGENASE LARGE SUBUNIT (RuBisCo). This novel gene, here-on referred to as Populus RuBisCo-like (PRL-1) gene localizes in chloroplast, endoplasmic reticulum and nuclear compartments of plant cells, acts as a transcriptional repressor and modulates adaptation to fluctuating light. P. trichocarpa genotypes with high PRL-1 expression levels quickly dissipated non-photochemical quenching (NPQ) upon transitioning from high to low light and rapidly induced NPQ upon returning to high light. The dynamic modulation of NPQ yields high quantum efficiency of linear electron transport (PSII) and 10-20% increased quantum efficiency of CO2 assimilation (CO2). The enhanced photosynthesis efficiency corresponded with increased plant height and biomass in P. trichocarpa with up to 35 % and 100% increases under field and greenhouse conditions. Transgenic Arabidopsis plants heterologously expressing the PRL-1 gene gained up to 200% in biomass and 97% in seed yield under greenhouse conditions. Taken together, PRL-1 presents a novel and trackable target for increasing photosynthetic efficiency in plants.

Project description:Due to global warming, invasive species have spread across the world. We therefore studied the impact of short-term (1 day or 2 days) and longer (7 days) heat stress on photosynthesis and secondary metabolites in Heracleum sosnowskyi, one of the important invasive species in the European Union. H. sosnowskyi leaves exposed to short-term heat stress (35 °C/1 d) showed a decrease in chlorophyll and maximum potential quantum efficiency of photosystem II (Fv/Fm) compared to control, 35 °C/2 d, or 30 °C/7 d treatments. In turn, the high level of lipid peroxidation and increased H2O2 accumulation indicated that the 30 °C/7 d stress induced oxidative damage. The contents of xanthotoxin and bergapten were elevated in the 2 d and 7 d treatments, while isopimpinellin was detected only in the heat-stressed plants. Additionally, the levels of free proline and anthocyanins significantly increased in response to high temperature, with a substantially higher increase in the 7 d (30 °C) treatment. The results indicate that the accumulation of proline, anthocyanins, and furanocoumarins, but not of phenolic acids or flavonols, contributes to protection of H. sosnowskyi plants against heat stress. Further studies could focus on the suppression of these metabolites to suppress the spread of this invasive species.

Project description:Despite the significance of nucleus genes in plant growth and development, little is known of the molecular bases of regulation of photosynthesis in woody plants.Hence discovering the genetic basis for photosynthesis related phenotypic variation and identifying the major genes controlling these complex traits in trees is essential. Combining the microarray technique and bulked segregant analysis strategy, we used poplar as a model to detect the nucleus genes differentially expressed in segregation population of photosynthesis traits. We measured the F1 interspecific population and segregated the stable extrem samples into two groups including three pools containing five incividuals.Use the Affymetrix poplar gene chip to decrypt the gene functions and mechanisms in different photosynthetic rate. Leaves were taken from high and low photosynthetic rate individuals for RNA extraction and hybridization on Affymetrix microarrays. H_A, H_B, H_C are from the high photosynthetic rate individuals. L_D,L_E, L_F are from the low photosynthetic rate incividuals.

Project description:We present experimental results demonstrating that, relative to continuous illumination, an increase of a factor of 3-10 in the photon efficiency of algal photosynthesis is attainable via the judicious application of pulsed light for light intensities of practical interest (e.g., average-to-peak solar irradiance). We also propose a simple model that can account for all the measurements. The model (1) reflects the essential rate-limiting elements in bioproductivity, (2) incorporates the impact of photon arrival-time statistics, and (3) accounts for how the enhancement in photon efficiency depends on the timescales of light pulsing and photon flux density. The key is avoiding "clogging" of the photosynthetic pathway by properly timing the light-dark cycles experienced by algal cells. We show how this can be realized with pulsed light sources, or by producing pulsed-light effects from continuous illumination via turbulent mixing in dense algal cultures in thin photobioreactors.

Project description:We report that the efficiency of reprogramming human somatic cells to induced pluripotent stem cells (hiPSCs) can be dramatically improved in a microfluidic environment. Microliter-volume confinement resulted in a 50-fold increase in efficiency over traditional reprogramming by delivery of synthetic mRNAs encoding transcription factors. In these small volumes, extracellular components of the TGF-β and other signaling pathways exhibited temporal regulation that appears critical to acquisition of pluripotency. The high quality and purity of the resulting hiPSCs (μ-hiPSCs) allowed direct differentiation into functional hepatocyte- and cardiomyocyte-like cells in the same platform without additional expansion.

Project description:Diatoms, considered as one of the most diverse and largest groups of algae, can provide the means to reach a sustainable production of petrochemical substitutes and bioactive compounds. However, a prerequisite to achieving this goal is to increase the solar-to-biomass conversion efficiency of photosynthesis, which generally remains less than 5% for most photosynthetic organisms. We have developed and implemented a rapid and effective approach, herein referred to as intracellular spectral recompositioning (ISR) of light, which, through absorption of excess blue light and its intracellular emission in the green spectral band, can improve light utilization. We demonstrate that ISR can be used chemogenically, by using lipophilic fluorophores, or biogenically, through the expression of an enhanced green fluorescent protein (eGFP) in the model diatom Phaeodactylum tricornutum. Engineered P. tricornutum cells expressing eGFP achieved 28% higher efficiency in photosynthesis than the parental strain, along with an increased effective quantum yield and reduced nonphotochemical quenching (NPQ) induction levels under high-light conditions. Further, pond simulator experiments demonstrated that eGFP transformants could outperform their wild-type parental strain by 50% in biomass production rate under simulated outdoor sunlight conditions. Transcriptome analysis identified up-regulation of major photosynthesis genes in the engineered strain in comparison with the wild type, along with down-regulation of NPQ genes involved in light stress response. Our findings provide a proof of concept for a strategy of developing more efficient photosynthetic cell factories to produce algae-based biofuels and bioactive products.
