Project description:To characterize plant growth in net zero energy greenhouse models covered with semi-transparent organic solar cells (OSCs), RNA was extracted from tomato leaf tissue. Six experimental treatments were created with three OSC filters: FTAZ:IT-M, PTB7-Th:IEICO-4F, FTAZ:PCBM. Boxes with either a clear glass cover or a spectrally-neutral cover were used as the high light (HC) control and the comparative light intensity (PC) control, respectively. These treatments were divided into 2 experiments. 4 treatments (3 OSC boxes and control) had a consistent light intensity and were called the PPFD Controlled (PC) experiment. The other 4 treatments were the Height Controlled (HC) experiment and had more variation in light intensity between treatments due to a consistent distance from the growth chamber light source (height). Plants in the FP_PC treatment flowered one day later than the PC control. FI_PC flowered one day earlier. Gene expression in samples from the FP_PC treatment had a distinct pattern related to flowering initiation.

Project description:To characterize plant growth in net zero energy greenhouse models with semi-transparent organic solar cell (OSC) filter roofs, RNA was extracted from red oak leaf lettuce leaves grown under 3 OPV filters (FTAZ:IT-M, PTB7-Th:IEICO-4F, FTAZ:PCBM) an clear and shaded glass controls. 2 light intensity experiments were used. PPPFD Controlled (PC) treatments had a constant light intensity and different light spectra created by the filters. Height Controlled (HC) treatments varied in both light intensity and spectra, according to the light transmission of each filter. When light intensity was controlled, plants grown under FTAZ:IT-M had a DEG profile distinct from plants grown under the other 2 filters. Key genes were up or down-regulated that indicate changes in anthocyanin accumulation, nitrogen metabolism and pest defense.

Project description:Arabidopsis HMA4 transformation facilitates Zn root-to-shoot translocation, but in Zn-supply dependent manner. It implies that distinct effects of HMA4 expression on Zn root:shoot distribution in transgenics grown under different Zn supply regimes results from the interaction between the transgene activity and the molecular background of the host plant. Microarray analysis was performed to compare expression profiles in leaves of transgenic and WT-plants grown in the presence of 5 µM Zn to identify the molecular mechanism underlying the difference in Zn root:shoot distribution between 35S:AtHMA4-expressing and wild-type tomato. To recognize tissue specific alterations in gene transcription in tomato leaves invoked by AtHMA4 expression, spongy paerenchyma + lower epidermis and palisade paerenchyma + upper epidermis were isolated using LCM technique. Understanding the interplay between the transgene and the endogens is crucial for planning genetic modification for Zn-biofortification purpose. 10-day old tomato plant (35S:AtHMA4 expressing and WT) were subjected to 5 µM Zn (added as ZnSo4) for 15 days. Three independent experiments were performed. At the end of each experiment the 3rd and 4th leaf (counting upwards) were collected, 1-2 mm long and 2-3 mm wide fragments were cut out from between the vascular bundles in the middle part of each leaf, embedded in NEG-50 Frozen Section Medium (Thermo Scientific), and frozen in liquid nitrogen. Total RNA isolated from three batches of leaves was amplified and used for three independent microarray analysis.

Response of tomato leaves to tobacco mosaic virus (TMV)

Project description:TMV-resistant (N), -susceptible (n), and enhanced susceptible mutant sun1-1 (N) tomato were grown in asceptic conditions in growth chambers (25º, 16hr/8hr light dark). 4-5 week old tomato were treated with TMV leaf sap or mock leaf sap, and leaf tissue was collected after 3, 9, and 27 hours. RNA was isolated from each sample (leaf tissue from one plant/one treatment/one timepoint), using standard TIGR protocols, and 25ug of total RNA was labeled using TIGR indirect labeling protocols. High-throughput single-cell transcriptome profiling of tomato leaf

Project description:The occurrence of Tomato chlorosis virus (ToCV) disease seriously damages tomato growth and yield, and there is no effective way to control ToCV transmission. So far, no studies have reported exploring the interaction between ToCV and tomato at the single cellular level. In this study, single cell RNA sequence was performed on a total of 26720 individual cells from healthy and ToCV-infected tomato leaves. We through identifying cell types, the first tomato leaf cell atlas was successfully constructed. In situ hybridization experiments identified specific marker genes that can be used to identify tomato cell types. Moreover, we have characterized transcription factors that may play a key role in tomato response to ToCV infection, and described the trichome differentiation trajectory during ToCV infection through pseudotime analysis. In conclusion, we proved the feasibility of single-cell sequencing to study the response of plants to biotic stress, and put forward new insights into the interaction between ToCV and tomato from the cellular level. Our data will lay the foundation for following studies between ToCV and plants, and will also provide a valuable reference for future research on non-model plant single cells.
