Project description:Vibrio alginolyticus is a Gram-negative marine bacterium. A limited population of the organisms causes acute gastroenteritis in humans. In this study, Vibrio alginolyticus wild type strain EPGS is compared with the mutants of Ser-Thr kinase PpkA and phosphatase PppA, after cultured for 7h, in Luria-Bertani containing medium 3 % NaCl at 30 C. Our goal is to determine the ppkA and pppA regulon. Three wild type and five mutant Vibrio alginolyticus samples were compared.

Project description:One of the most distinct features of Pseudoalteromonas sp. SCSIO 11900 is its ability to form a very robust pellicle than most Pseudoalteromonas strains. Thus we want to identify the genes essential for the pellicle formation of SCSIO 11900. We compared transcriptom profiles of planktonic cells, initial pellicle and mature pellicle of coral Pseudoalteromonas sp. SCSIO 11900 and revealed that some unique genes from horizontal gene transfer is involved in the pellicle formation of SCSIO 11900.

Project description:RNA-seq analysis demonstrated that VqsA (18672) controls ~275 genes' expression in V. alginolyticus. Collectively, our data established that VqsA plays essential roles in QS regulation and may facilitate the illumination of the mechanisms bacterial cells sense environmental signals and integrate them into coordinated QS responses.

Project description:HEK293 cells that stably express PHB2 (V1 or V3) fused at C-terminus with the biotin ligase from A. aeolicus were incubated with 100 μM biotin for 18 h. The mitochondrial fractions were lysed in RIPA buffer, and the clarified supernatants were incubated with streptavidin agarose beads for 2 h at 4° C. Proteins on the beads were digested by adding trypsin/Lys-C mix for 16 h at 37° C. Three biological replicates of the samples were individually prepared and analyzed by LC-MS/MS. Label-free quantification was performed using Proteome Discoverer 2.2 software.
