Project description:Identification of molecular determinants underlying the firming effect and protection gainst senescence of high CO2 using diploid strawberries: One of the greatest threats to strawberries is rapid softening, however firmness increases during or following high CO2 levels. Firmness at consumption is an obvious target for preventing fruit loss and to gain eating quality. Therefore, we performed RNA-seq analysis, construcing a weighted gene co-expression network analysis (WGCNA) to identify which molecular determinants play a role in cell wall integrity , using strawberries under storage conditions. Differential gene expression (DEG) analysis showed that cell wall structural architecture of firmer CO2 -treated strawberries is characterised by xyloglucans stabilisation attributed mainly to a down-regulation of Csl-like E1, β-glc, XTH15 and maintenance of expression levels of FUT and GMP as well as improved lamella integrity linked to a down-regulation of RG-lyase and PL-like. The preservation of cell wall elasticity together with the up-regulation of LEA, EXPA4, and MATE transporters required to maintain cell turgor, are the mechanisms controlled by high CO2. In stressed air-cold stored strawberries, in addition to an acute softening, there is a preferential transcript accumulation of genes involved in lignin and raffinose pathways. The oxidative stress involving jasmonate and H2O 2 is characteristic of senescent non-cold stored samples. The results are fundamental and practical for breeding in strawberry industry.

Photorespiratory stress low CO2 conditions

Project description:Six weeks old Arabidopsis plants were transferred to a low CO2 (100 ppm) environment during 24 hours and compared to control plants kept under ambient CO2 conditions. Limited CO2 availability will cause higher rates of photorespiration and affect the plant redox homeostasis. We studied the transcriptomic impact of exposing plants to a lower CO2 environment to further eliculidate the signaling pathways during photorespiratory stress.

Multiplexed proteomics of autophagy deficient macrophages reveals enhanced antimicrobial immunity via the oxidative stress response

Project description:Macrophages play a critical role in clearance of cytosolic pathogens. Autophagy functions at the intersection of antimicrobial innate immunity and protein quality control; however, the impacts of infection and autophagy on shaping the macrophage proteome are poorly understood. Here, we describe a deep multi- dimensional proteomic analysis of primary murine macrophages infected with Shigella flexneri (S.flexneri). Tandem mass tagging (TMT) revealed dynamic genotype- and infection-dependent differences in host and pathogen proteins, phosphorylation and ubiquitination. These data catalogue the complex circuitry connecting autophagy, inflammatory signaling and the oxidative stress response. Loss of the autophagy gene Atg16l1 induced basal oxidative stress, activated the compensatory glutathione biosynthetic machinery, and enhanced clearance of S.flexneri. Pathogen clearance was similarly enhanced in wild type macrophages upon pharmacological inhibition of cysteine import. Our study provides a resource for innate immunity research and reveals that ATG16L1 dampens antimicrobial immunity by regulating oxidative stress.

Project description:Cyanobacteria are oxygenic photoautotrophs notable for their ability to utilize atmospheric CO2 as the major source of carbon. The prospect of using cyanobacteria in converting solar energy and high concentrations of CO2 (e.g. flue gas from coal power plants) efficiently into biomass and renewable energy sources is of interest to many research fields. In order to guide further advances in this area, a better understanding about the metabolic changes that occur under conditions of high CO2 is important. The objective of this study is to utilize genome-wide microarray expression profiling in the unicellular diazotrophic cyanobacterium Cyanothece 51142 grown in 8% CO2-enriched air and to determined the impact of high CO2 on cyanobacterial cell physiology and growth.
