ABSTRACT: We established two in vitro cell-free systems to mimic the activation of mTORC1 and autophagy by monitoring the translocation of two readout proteins Raptor and ATG14. Based on the two systems, we further screened the potential regulators involved in the leucine-mediated mTORC1 and autophagy pathways using comparative proteomics strategy.
