Project description:The endoplasmic reticulum (ER) is the organelle of nucleated cells that produces lipids, sugars and proteins. More than 20 ER-resident members of the Protein Disulfide Isomerase (PDI) family regulate formation, isomerization and disassembly of covalent bonds in newly synthesized polypeptides. The PDI family includes few membrane-bound members. Among these, TMX1, TMX2, TMX3, TMX4 and TMX5 belong to the thioredoxin-related transmembrane (TMX) protein family. TMX5 is the least known member of the family. Here, we establish that TMX5 covalently engages via its active site cysteine residue at position 220 a subset of secretory proteins, mainly single- and multi-pass Golgi-resident polypeptides. TMX5 also interacts non-covalently, and covalently, via non-catalytic cysteine residues, with the PDI family members PDI, ERp57 and ERp44. The association of TMX5 and ERp44 requires formation of a mixed disulfide between the catalytic cysteine residue 29 of ERp44 and the non-catalytic cysteine residues 114 and/or 124 of TMX5 and controls the ER retention of TMX5. Thus, TMX5 belongs to the family of proteins including Ero1, Ero1, Prx4, ERAP1, SUMF1 that do not display ER retention sequences and rely on ERp44 engagement for proper inter-compartmental distribution. The client-specificity of TMX5 in cellulo was assessed by expressing mutant forms of the enzymes, where the last cysteine residue of the TMX’s CXXC catalytic sites has been mutated to alanine, which stabilizes the mixed disulfide that oxidoreductases establish with clients. Clients remain disulfide-bonded to the oxidoreductase and are identified upon co-immunoprecipitation and mass spectrometry analyses.

Project description:Social insects frequently engage in oral fluid exchange – trophallaxis – between adults, and between adults and larvae. Although trophallaxis is normally considered a simple food-sharing mechanism, we hypothesized that endogenous components of this fluid might underlie a novel means of chemical communication that directly influences colony organization. Through protein and small-molecule mass spectrometry and RNA sequencing in the ant Camponotus floridanus, we found that trophallactic fluid contains a set of specific proteins, hydrocarbons, microRNAs, and Juvenile Hormone, an important insect growth regulator. Comparison of trophallactic fluid proteins across social insect species (ants Camponotus fellah and Solenopsis invictis, honeybees Apis mellifera) revealed that many are regulators of growth and development. These results raise the possibility that, in addition to its role in food transfer, trophallaxis is a mode of communication that enables communal control of colony phenotypes.

Project description:The endoplasmic reticulum (ER) is a multifaceted organelle that plays an essential role in cellular processes such as protein folding, modification, and trafficking. It also acts as a proteostasis sensor and contributes to adaptation responses to maintain cellular homeostasis. The transcription factor NRF1 resides in the ER and is constantly transported from the ER to the cytosol for proteasomal degradation. However, when the proteasome function is defective, NRF1 escapes degradation and undergoes proteolytic cleavage by the protease DDI2, generating a transcriptionally active form that restores proteostasis, including proteasome function. Despite the importance of NRF1 in these processes, the mechanisms that regulate its proteolytic activation and transcriptional potential remain poorly understood. In this study, we utilized a functional molecular approach to elucidate the critical steps involved in NRF1 cleavage. Our results demonstrate that the ER is a crucial regulator of NRF1 function, orchestrating its ubiquitination by the ER-localized E3 ubiquitin ligase HRD1, a component of the ER-associated degradation (ERAD) pathway. Furthermore, we show that HRD1-mediated NRF1 ubiquitination is necessary for DDI2-mediated processing in cells. Notably, we found that the fusion of a ubiquitin moiety to NRF1 was sufficient to promote its proteolytic maturation by DDI2, thus bypassing the requirement for its trafficking to the ER. Our findings highlight the intricate mechanism by which NRF1 is activated by DDI2 and coordinates the transcriptional activity of an adaptation response in cells and suggest potential avenues for therapeutic interventions in conditions associated with proteasome impairment.

Project description:Endolysins are peptidoglycan hydrolases produced at the end of the bacteriophage (phage) replication cycle to lyse the host cell. Gram-positive phages endolysins come in a variety of multi-modular forms that combine different catalytic domains and may have evolved to adapt to their bacterial hosts. However, the reason why phage can adopt endolysin with such complex multidomain architecture is for the moment not well understood. We used the Streptococcus dysgalactiae phage endolysin PlySK1249 as a model to study the implication of multi-domain architecture in phage-induced bacterial lysis and lysis regulation. The activity of the enzyme relied on a bacteriolytic amidase (Ami), a non-bacteriolytic L-Ala-D-Ala endopeptidase (CHAP) acting as a de-chaining enzyme and central LysM cell wall binding domain (CBD). Ami and CHAP synergized for peptidoglycan digestion and bacteriolysis in the native enzyme or when expressed individually and reunified in vitro. This cooperation could be modulated by bacterial cell wall-associated proteases, which specifically cleaved the two linkers connecting the different domains. While both catalytic domains were observed to act coordinately to optimize bacterial lysis, the CBD is expected to delay diffusion of the enzyme until proteolytic inactivation is achieved. As for certain autolysins, PlySK1249 cleavage by bacterial cell wall associated proteases might be an example of dual phage-bacterial regulation and mutual coevolution.

Project description:At dawn of a scorching summer day, land plants must anticipate upcoming extreme midday temperatures by timely establishing molecular defenses that can maintain heat-labile membranes and proteins in a functional state. A gradual morning pre-exposure to increasing sub-damaging temperatures induces the accumulation of heat-shock proteins (HSPs) that are central to the onset of plant acquired thermotolerance (AT). In an attempt to gain knowledge on the mechanisms of AT in the model land plant Physcomitrium patens, we used label-free LC-MS/MS proteomics to quantify the accumulated and depleted proteins before and following a mild heat-priming treatment.

Project description:Socially exchanged fluids, such as seminal fluid and milk, are a direct means through which an organism can influence conspecifics. When orally feeding larval offspring via trophallaxis, workers of the carpenter ant Camponotus floridanus were recently shown to transfer Juvenile Hormone III (JH), a key developmental regulator, as well as paralogs of JH esterase (JHE), an enzyme that catalyzes degradation of JH. Here we combine proteomic, phylogenetic and selection analyses to investigate the evolution of this esterase subfamily. We show that members of the Camponotus JHE-like protein family have undergone multiple duplications, experienced positive selection, and changed tissue localization to become abundantly and selectively present in trophallactic fluid. The Camponotus trophallactic esterases have maintained their catalytic triads but contain a number of positively-selected amino acid changes distributed throughout the protein, which possibly reflect an adaptation to the highly acidic trophallactic fluid of formicine ants. To determine whether these esterases might regulate larval development, we fed workers with a JHE-specific pharmacological inhibitor to introduce it into the trophallactic network. This inhibitor increased the likelihood of pupation of the larvae reared by these workers, similar and complementary to supplementation with JH. Together, these findings suggest that JHE-like proteins have evolved new roles in the Camponotus genus in inter-individual regulation of larval development.

Project description:Here we made an attempt to obtain partial structural information on the topology of multispan integral membrane proteins of yeast by isolating organellar membranes, removing peripheral membrane proteins at pH 11.5 and introducing chemical crosslinks between vicinal amino acids either using homo- or hetero-bifunctional crosslinkers. Proteins were digested with specific proteases and the products analysed by mass spectrometry. Dedicated software tools were used together with filtering steps optimized to remove false positive crosslinks. In proteins of known structure, crosslinks were found only between loops residing on the same side of the membrane. As may be expected, crosslinks were mainly found in very abundant proteins. Our approach seems to hold to promise to yield low resolution topological information for naturally very abundant or strongly overexpressed proteins with relatively little effort.

Project description:SNARE (soluble N-ethylmaleimide-sensitive factor attachment protein receptor) proteins drive vesicle fusion and contribute to homoeostasis, pathogen defence, cell expansion, and growth in plants. In Arabidopsis thaliana, the two homologous Qa-SNAREs, SYP121 and SYP122 facilitate the majority of secretory traffic to the plasma membrane. In the absence of stress, the single mutants are indistinguishable from wild-type plants, implying a redundancy in their functions. Nonetheless, several studies suggest differences among the secretory cargo of these SNAREs. To address this issue, we conducted an analysis of the proteins secreted by cultured wild-type, syp121 and syp122 mutant Arabidopsis seedlings. Here, we report that a number of cargo proteins are associated differentially with traffic mediated by SYP121 and SYP122. The data also point to important overlaps between the SNAREs. The findings lead us to conclude that the two Qa-SNAREs mediate distinct, but complementary secretory pathways during vegetative plant growth.
