Analysis of circ101093 binding proteins in A549 cells after RNA pull-down.

ABSTRACT: We used circular RNA 101093 and its antisense to perform RNA pull-down, and analyzed the peptides in blank group, antisense group and circular RNA 101093 group. The object of this group is to analyze circ101093-binding protein. Blank and antisense group are used as negative control group.

Project description:The neonatal mammalian heart is able to regenerate after injury by inducing cardiomyocyte proliferation. However, this regenerative capacity is virtually lost in the adult mammalian heart. Extracellular vesicles (EVs) have been shown to play an important cardioprotective role in heart repair. Here, we performed proteomic analysis of EVs from neonatal mouse heart tissues (Neo-EVs), EVs regenerated from neonatal heart tissues after apicoectomy (AR-Neo-EVs), and EVs from adult mouse hearts (Adu-EVs), to compare the differential changes in proteins among them.

Transcription profiling of human bronchioloalveolar lavage (BAL) not used for diagnostic purposes taken from patients with LF syndrome and SOS of stage II active disease

Project description:Cells from bronchioloalveolar lavage (BAL) not used for diagnostic purpose were taken from patients with LF syndrome and SOS of stage II active disease. The SOS samples were divided in two groups by the means of CD4+-/CD8+-cell ratio < 5 or > 5, respectively. At least five samples of each group were pooled. In all cases clinical diagnosis was established according to the criteria of the ATS-ERS consensus statement {Costabel, 1999 #68}. In addition diagnosis was confirmed by transbronchial biopsies and lymphocytic, T-helper dominated alveolitis assessed in BAL. An informed consent was obtained from the patients. The patient data were anonymized prior to use in the laboratory.

The YbjN regulon in Escherichia coli

Project description:YbjN, an enterobacteria-specific protein, is a multicopy suppressor of ts9 temperature sensitivity in Escherichia coli. Microarray study revealed that the expression level of ybjN was inversely correlated with the expression of flagellar, fimbrial and acid resistance genes. Over-expression of ybjN significantly down-regulated genes involved in the citric acid cycle, glycolysis, the glyoxylate shunt, oxidative phosphorylation, and amino acid and nucleotide metabolism. On the other hand, over-expression of ybjN up-regulated toxin-antitoxin modules, the SOS responsive pathway, cold shock proteins and starvation-induced transporter genes. Our results collectively suggest that YbjN may play important roles in regulating bacterial multicellular behaviors, metabolism and survival under various stress conditions in Es. coli. A total of 8 samples were analyzed: E. coli wild type strain (2 replicates); E. coli ybjN mutant strain (3 replicates); E. coli ybjN over-expression strain (3 replicates).

Insulin reverses impaired alveolar fluid clearance in ARDS by inhibiting LPS-induced autophagy and inflammatory

Project description:Until now, acute respiratory distress syndrome (ARDS) has been a difficult clinical condition with a high mortality and morbidity rate, and is characterized by a build-up of alveolar fluid and impaired clearance. The underlying mechanism is not yet fully understood and no specific treatment available. Autophagy activation is associated with ARDS caused by different pathogenic factors. It represents a new direction of prevention and treatment of ARDS to restrain autophagy to a reasonable level through pharmacological and molecular genetic methods. Na, K-ATPase is the main gradient driver of pulmonary water clearance in ARDS and could be degraded by the autophagy-lysosome pathway to affect its abundance and enzyme activity. As a normal growth hormone in human body, insulin has been widely used in clinical for a long time. To investigate the association of insulin with Na, K-ATPase, autophagy and inflammatory markers in LPS-treated C57BL/6 mice by survival assessment, proteomic analysis, histologic examination, inflammatory cell counting, myeloperoxidase, TNF-α, IL-1β activity analysis etc. This was also verified on mouse alveolar epithelial type Ⅱ (AT Ⅱ) and A549 cells by transmission electron microscopy. We found that insulin restored the transport efficiency of Na, K-ATPase, inhibited the activation of autophagy and reduced the release of inflammatory factors caused by alveolar epithelial damage. The regulation mechanism of insulin on Na, K-ATPase by inhibiting autophagy function may provide new drug targets for the treatment of ARDS.

Project description:Listeria monocytogenes is a food-borne pathogen which causes listeriosis. It is an intracellular parasite invading the epithelial cells where it escapes from the vacuole into the host cytoplasm to replicate, using actin-based motility to move within and between cells. The intracellular life cycle is well documented whereas the time spent in the lumen of the intestine is poorly understood. The aim of this study was to investigate the mechanism by which L. monocytogenes adapts to the environment of the small intestine prior to invasion. Specifically, to determine if the PrfA regulon, that encodes the virulence factors of L. monocytogenes, is switched on by signals within the intestinal lumen. L. monocytogenes were grown under aerobic or microaerobic conditions with glucose or glycerol as carbon source.
