Project description:In the ex situ conservation of chondrycthyan species, successful reproduction in aquaria is essential. However, aquatic species often exhibit reduced reproductive success under human care. Different factors, including water temperature, nutrition, and intrinsic genetic and epigenetic elements, influence their fertility. Conventional sperm analyses do not provide insights into the functional competence of semen. Therefore, proteomics is gaining relevance, increasing a better understanding of male physiology. In this study, we investigated the proteomic profiles of seminal plasma and spermatozoa from small-spotted catsharks in two different environments: natural environment and aquarium environment.

Project description:The Spodoptera genus is defined as the pest-rich genus because it contains some of the most destructive lepidopteran crop pests, characterized by a wide host range. During feeding, the caterpillars release small amounts of oral secretion (OS) onto the wounded leaves. This secretion contains herbivore-induced molecular patterns (HAMPs) that activate the plant defense response, as well as effectors that may inhibit or diminish the plant’s anti-herbivory response. In this study, we explored the protein components of the OS of two Spodoptera species, Spodoptera exigua and Spodoptera littoralis.

Proteomic and pathway profiling of four Breast Cancer cell lines exosomes

Project description:Biomarker discovery in breast cancer (BC) is a clinical need for therapeutics and non-invasive diagnostics. In a single study, we provide a comparative differential proteomic profile of four invasion, metastasis, angiogenesis and drug resistance. Exosome proteome profile reflects their different BC cell origin suggesting potential indicators of BC subtype. Further, our pathway inference analysis reveals a differential role of exosomes in BC signalling pathways in recipient cells, according to their protein cargo and cell origin. Project description:The specific aim of the present work is to expand the data on the salivary proteome of O. moubata adult ticks, particularly of female ticks, by solving the aforementioned drawbacks, as part of the in depth characterization and analysis of the O. moubata sialome. Knowing this sialome will allow identifying and selecting novel salivary antigens as targets for tick vaccines, and their subsequent testing in animal immunization trials. We pay special attention to female ticks because they are a key developmental stage in the tick life cycle and vaccines may exert on them double deleterious effects, for instance an increase of mortality and a reduction/inhibition of the reproductive performance. With that aim, in the current work we have analysed samples of female and male saliva separately using two different mass spectrometry approaches: data-dependent acquisition (DDA) LC-MS/MS and Sequential Window Acquisition of all Theoretical fragment ion spectra Mass Spectrometry (SWATH MS). This last technique is a specific variant of data-independent acquisition (DIA) methods and is emerging as a technology that combines deep proteome coverage capabilities with quantitative consistency and accuracy.

A 3D printed plastic frame deeply impacts yeast cell growth

Project description:Solid State Fermentation (SSF) processes have been explored for yeast growth and protein and metabolites production. However, most of these processes lack standardization. In this work, we present a polylactic acid (PLA) 3D printed matrix that dramatically enhances yeast growth when embedded in liquid media compared to equivalent static cultures, and changes yeast expression patterns at the proteome level. Moreover, differences in sugar assimilation and ethanol production, as the main product of alcoholic fermentation, are observed. Our results suggest that these matrixes may be useful for a vast range of biotechnological applications based on yeast fermentation.

Project description:Extracellular vesicles (EVs) have been characterized from many species of parasitic helminths, and recent experimental evidence supports important functions for their cargo in host-parasite relationships. Here, we carried out a detailed proteomic analysis of two populations of EVs – exosome like vesicles (ELVs) and microvesicles (MVs) – from Schistosoma mansoni. We profiled the proteins present in these EVs (including external trypsin-liberated peptides, cargo proteins, integral membrane proteins (IMPs) and peripheral membrane proteins (PMPs) using LC-MS/MS.

Project description:Prediction of male or semen fertility potential remains a persistent challenge that has yet to be fully resolved. This work analyzed several in vitro parameters and proteome of spermatozoa in bulls cataloged as high (HF; n=5) and low field (LF; n=5) fertility after more than a thousand artificial inseminations. Sperm motility was evaluated by Computer-Assisted Sperm Analysis. Sperm viability, mitochondrial membrane potential (MMP), and reactive oxygen species (mROS) of spermatozoa were assessed by flow cytometry. Proteome was evaluated by SWATH-MS procedure. Spermatozoa of HF bulls showed significantly higher total motility than the LF group (41.4% vs. 29.7%). Rates of healthy sperm (live, high MMP, and low mROS) for HF and LF bull groups were 49% and 43%, respectively (p > 0.05). Spermatozoa of HF bulls showed higher presence of differentially abundant proteins (DAPs) related to both energy production (COX7C), mainly OXPHOS pathway, and to the development of structures linked with the motility process (TPPP2, SSMEM1 and SPAG16). Furthermore, we observed that EQTN, together with other DAPs related to the interaction with the oocyte, were overrepresented in HF bull spermatozoa. The biological processes related to protein processing, catabolism, and protein folding were found to be overrepresented in LF bull sperm in which the HSP90AA1 chaperone was identified as the most DAP
