Acetylome profiling of bermudagrass stolons

ABSTRACT: Bermudagrass (Cynodon dactylon L.) is an important warm-season turfgrass species with well-developed stolons, which lay the foundation for fast propagation of bermudagrass plants through asexually clonal growth. However, the growth and development of bermudagrass stolons are still poorly understood at the molecular level. In this study, we comprehensively analyzed the acetylation modifications of proteins in fast growing stolons of bermudagrass cultivar Yangjiang. A total of 4657 lysine acetylation sites on 1914 proteins were successfully identified using liquid chromatography coupled to tandem mass spectrometry.

Bermudagrass stolon phosphoproteomics

Project description:As a widely used turfgrass species, bermudagrass (Cynodon dactylon L.) can be easily propagated by colonial growth of stolons. Previous studies collectively revealed that exotic environmental factors and intrinsic hormones and genes are all involved in the differentiation, development, and diageotropically growth of stolons. However, the detailed molecular mechanism how environmental and hormone signals regulate the gene expression and biochemical activities in bermudagrass stolons remains unclear. In this study, LC-MS/MS analyses of the total protein extracts of bermudagrass stolons without preliminary phosphopeptide-enrichment successfully identified 862 nonredundant phosphorylation sites and 613 phosphoproteins.

Comparative proteomics analyses of rhizomes and stolons in bermudagrass

Project description:As an important perennial warm-season turfgrass species, bermudagrass (Cynodon dactylon L.) form underground-growing rhizomes and aboveground-growing stolons simultaneously, making it a fast propagating clonal plant with strong regeneration ability. However, the intrinsic difference between the two types of specialized stems are still uncharacterized, especially at the molecular level. In the current study, we compared the internode proteomes of rhizomes and stolons at the same developmental stage in the bermudagrass cultivar Yangjiang using isobaric tags for relative and absolute quantitation (iTRAQ). The results indicated that 228 protein species were differentially accumulated in the two specialized stems. These DAPs comprise complex protein networks to finely regulate diverse cellular activities in the two types of specialized stems. Notably, photosynthesis and flavonoid biosynthesis were significantly regulated in stolons, whereas sucrose and starch metabolism were significantly regulated in rhizomes.

In this study, we compared the proteome of the internode section of shoots and stolons at the same developmental stage in bermudagrass cultivar Yangjiang. The results indicated that 376 protein species were differentially accumulated in the two types of stems.

Integrated phosphoproteome, ubiquitinome, and acetylome analyses uncovering the post-translational modifications crosstalk during MSP1-induced signaling in rice

Project description:Magnaporthe oryzae snodprot1 homologous protein (MSP1) has been shown to act as a pathogen-associated molecular pattern (PAMPs) and trigger PAMP-triggered immunity (PTI) response involving programmed cell death and expression of various defense-related genes in rice. The involvement of several post-translational modifications (PTMs) in the regulation of plant immune response, especially PTI, during pathogen infection is well established, however, the information on the regulatory roles of these PTMs in response to MSP1-induced signaling in rice is currently elusive. Here, we report the phosphoproteome, ubiquitinome, and acetylproteome to investigate the MSP1-induced PTMs alterations in MSP1 overexpressed rice. Our analysis identified a total of 4,666 PTM modified sites in rice leaves including 4,292 phosphosites, 189 ubiquitin sites, and 185 acetylation sites. Among these, PTM status of 437 phosphorylated, 53 ubiquitinated, and 68 acetylated peptides were significantly changed by MSP1. Functional annotation of MSP1 modulated peptides by MapMan analysis revealed that these were majorly associated with cellular immune responses such as signaling, transcription factors, DNA and RNA regulation, and protein metabolism, among others. Taken together, this study uncovers the MSP1-induced PTMs changes in rice proteins and identified several novel components of rice-MSP1 interaction.

Project description:Two rounds of TMT relative quantitative proteomics were performed to detect cellular factors involved in p-eIF4E regulation of the synthesis of viral proteins.our first round of screening identified differentially expressed proteins in PEDV-infected cells and mock-infected cells; the cellular pathways involved were mainly the estrogen, cAMP, and calcium signaling pathways. Second round screening identified differentially expressed proteins in the PEDV-infected S209A-Vero cells vs. the PEDV-infected WT-Vero cells; the regulated cellular pathways were found to be mainly in the PI3K-Akt, focal adhesion, and mTOR signaling pathways, and the biological processes and molecular functions in which p-eIF4E played a role were related mainly to metabolism and biogenesis, catalytic activity, and stimuli response.4006 host factors were detected, of which 193 (in brown) were significantly upregulated (ratio ≥1.2, P＜0.05) and 191 (in green) were down-regulated upon PEDV infection (ratio ≤0.83, P＜0.05). 29 of the 191 down-regulated proteins were susceptible to a low level of p-eIF4E . Notably, among the 193 upregulated cellular proteins, 77 were upregulated in the WT-Vero over the S209A-Vero cells , suggesting that the WT-Vero cells are more susceptible to a high level of p-eIF4E.
