Proteome analysis of TRANSLOCON OF THE OUTER CHLOROPLAST membrane 75 (TOC75) knockdown mutants

Surveying the chloroplast protein kinase preferences for cation cofactors in Arabidopsis thaliana

Project description:Survival of plants depends on their ability to adapt to ever-changing environmental conditions. Chloroplasts sense different stimuli and respond to ion fluxes in the cytosol triggering systemic responses under stress conditions. Indeed, changes in calcium and magnesium ion concentration in the chloroplast stroma impact the immune response against pathogens and adapt the photosynthetic machinery under fluctuating light conditions. Here we investigated the link between di-valent cations, calcium, magnesium and manganese and the protein kinase activity in Arabidopsis chloroplasts. Our results show that overall, manganese ions are the strongest activator of kinase activity in chloroplasts followed by magnesium ions, whereas calcium ions have seemingly no effect. Additionally, when comparing the phosphorylation pattern from Arabidopsis wild type and a cmt1 mutant, which is defective in manganese import into chloroplasts, the phosphorylation of specific protein bands is strongly reduced in the mutant chloroplasts supporting the notion that chloroplasts contain manganese-dependent protein kinases. These findings provide insights for the future characterization of chloroplast protein kinases activity regarding in-vitro assays and potential target proteins.

Structural characterizations of the chloroplast translocon protein Tic110.

Project description:Tic110 is a major component of the chloroplast protein import translocon. Two functions with mutually exclusive structures have been proposed for Tic110: a protein-conducting channel with six transmembrane domains and a scaffold with two N-terminal transmembrane domains followed by a large soluble domain for binding transit peptides and other stromal translocon components. To investigate the structure of Tic110, Tic110 from Cyanidioschyzon merolae (CmTic110) was characterized. We constructed three fragments, CmTic110A , CmTic110B and CmTic110C , with increasing N-terminal truncations, to perform small-angle X-ray scattering (SAXS) and X-ray crystallography analyses and Dali structural comparison. Here we report the molecular envelope of CmTic110B and CmTic110C determined by SAXS, and the crystal structure of CmTic110C at 4.2 Å. Our data indicate that the C-terminal half of CmTic110 possesses a rod-shaped helix-repeat structure that is too flattened and elongated to be a channel. The structure is most similar to the HEAT-repeat motif that functions as scaffolds for protein-protein interactions.

Characterization of the translocon of the outer envelope of chloroplasts.

Project description:The protein translocon of the outer envelope of chloroplasts (Toc) consists of the core subunits Toc159, Toc75, and Toc34. To investigate the molecular structure, the core complex was purified. This core complex has an apparent molecular mass of approximately 500 kD and a molecular stoichiometry of 1:4:4-5 between Toc159, Toc75, and Toc34. The isolated translocon recognizes both transit sequences and precursor proteins in a GTP-dependent manner, suggesting its functional integrity. The complex is embedded by the lipids phosphatidylcholine and digalactosyldiacylglyceride. Two-dimensional structural analysis by EM revealed roughly circular particles consistent with the formation of a stable core complex. The particles show a diameter of approximately 130 A with a solid ring and a less dense interior structure. A three-dimensional map obtained by random conical tilt reconstruction of electron micrographs suggests that a "finger"-like central region separates four curved translocation channels within one complex.

The comparison for the transcript levels of chloroplast-encoded genes between wild type (Col-0) and pbf8 mutants

Project description:We found that thylakoid-anchored protein PBF8 is a key regulator for Photosystem I (PSI) biogenesis. To explore the role of PBF8 in regulating chloroplast gene expression, we performed the RNA-seq to compare the the transcript levels of chloroplast-encoded genes between wild type (Col-0) and pbf8 mutants. To this end, we isolated the total RNA form 12-day-old wild type and pbf8 seedlings grown on the MS medium under long-day conditions (14 h light, 10 h dark) at 22 ºC and with a light intensity of 80 µmol m-2 s-1. The rRNAs were deleted using the Ribo-Zero Kit (Epicentre). The resulting rRNA-depleted RNA was used for preparing the sequencing library with NEBNext Single Cell/Low input library Prep Kit. The libraries were pooled and sequenced on an Illumina Nova 6000 system with 150-bp pair-end reads. Finally, our results show that the transcript accumulation for chloroplast-encoded PSI subunit and assembly factor genes between the wild type (Col-0) and pbf8 samples, suggesting PBF8 may not affect the transcript levels of chloroplast-encoded PSI subunits and assembly factors in chloroplasts.
