Quantification of oxidative stress-related proteins using MRM approach in atherosclerosis-related and non-related to chronic kidney disease

ABSTRACT: The aim of MRM analysis of selected oxidative stress-related proteins, including MPO, CERu, sTNF, SOD3, HMOX1, PON1, GPX3, NOS2, CAT and IFNG was to investigate changes in plasma samples from patients at different stages of atherosclerosis-related to chronic kidney disease, classical atherosclerosis as well as in comparison with healthy controls.

iTRAQ-MRM approach identify plasma biomarkers of atherosclerosis and myocardial injury

Project description:Current methods of stratifying patient risk of myocardial injury and stable angina rely on complex combinations of risk factors that exhibit only limited prognostic power, hence there remains a need to identify biomarkers that can be sampled non-invasively and more accurately predict patient outcomes in the clinic. In the current study, we performed comparative quantitative proteomics on whole plasma sampled from patients with stable angina (NMI), acute myocardial infarction (MI), and healthy control subjects without angina (Ctrl). We detected a total of 371 proteins with high confidence (FDR < 1%, p < 0.05), including 53 candidate biomarkers that displayed ≥ 2-fold modulated expression in patients with cardiovascular diseases (27 associated with atherosclerotic stable angina, 26 with myocardial injury). In the verification phase, we used label-free LC-MRM-MS-based targeted proteomic method to quantify and to verify the candidate biomarkers in pooled plasma, excluded peptides that were poorly distinguished from background, and then performed further validation of the remaining candidates in 49 individual plasma samples. Using this approach, we identified a final panel of 8 proteins that were both reliably and significantly modulated in disease (p < 0.05), including novel biomarkers of atherosclerotic stable angina that have been implicated in endothelial dysfunction (F10 and MST1), and previously unknown biomarkers of myocardial injury reportedly involved in either plaque destabilization (SERPINA3, CPN2, LUM) or tissue protection/repair mechanisms (ORM2, ACTG1, NAGLU). Taken together, our data showed that prognostic markers can be successfully detected in non-depleted human plasma using an iTRAQ/MRM-based discovery-validation approach, and also demonstrate that a novel panel 8 biomarkers can discriminate between the complex pathophysiologies of atherosclerotic stable angina and myocardial injury.

JAG1-NOTCH4 mechanosensing drives atherosclerosis

Project description:Endothelial cell (EC) sensing of fluid shear stress regulates atherosclerosis, a disease of arteries that causes heart attack and stroke. Atherosclerosis preferentially develops at regions of arteries exposed to low oscillatory shear stress (LOSS), whereas high shear regions are protected. We show using inducible EC-specific genetic deletion in hyperlipidaemic mice that the Notch ligands JAG1 and DLL4 have opposing roles in atherosclerosis. While endothelial Jag1 promoted atherosclerosis at sites of LOSS, endothelial Dll4 was atheroprotective. Analysis of porcine and murine arteries and cultured human coronary artery EC exposed to experimental flow revealed that JAG1 and its receptor NOTCH4 are strongly upregulated by LOSS. Functional studies in cultured cells and in mice with EC-specific deletion of Jag1 show that JAG1-NOTCH4 signalling drives vascular dysfunction by repressing endothelial repair. These data demonstrate a fundamental role for JAG1-NOTCH4 in sensing LOSS during disease, and suggest therapeutic targeting of this pathway to treat atherosclerosis.

Clonal Haematopoiesis-related Mutant ASXL1 Promotes Atherosclerosis in Mice via Dysregulated Innate Immunity

Project description:Certain somatic mutations confer a fitness advantage in hematopoietic stem cells, resulting in the clonal expansion of mutant blood cells, known as clonal haematopoiesis (CH). Among the top 3 CH mutations, ASXL1 mutations present the highest risk for developing cardiovascular diseases (CVDs). However, how ASXL1 mutations induce CVDs remains totally elusive. Here we show that haematopoietic cells harbouring C-terminally truncated form of ASXL1 mutant (ASXL1-MT) accelerated development of atherosclerosis in Ldlr–/– mice. Transcriptome analyses of plaque-cells showed inflammatory signatures of monocytes and macrophages expressing ASXL1-MT. Mechanistically, wild-type ASXL1 inhibited innate immune signalling through the inhibition of IRAK1-TAK1 interaction in the cytoplasm, indicating an unexpected non-epigenetic role of ASXL1. In contrast, ASXL1-MT lost this regulatory function, leading to NF-κB activation. Intriguingly, IRAK1/4 inhibition decreased inflammatory monocytes and atherosclerosis driven by ASXL1-MT. The present work connects ASXL1 mutations with inflammation and CVDs, giving a clue to prevent CVDs in ASXL1-CH.

Repeated gestational exposure of mice to chlorpyrifos oxon is associated with paraoxonase 1 (PON1)-modulated effects in maternal and fetal tissues

Project description:Chlorpyrifos oxon (CPO), the toxic metabolite of the organophosphorus (OP) insecticide chlorpyrifos, causes developmental neurotoxicity in humans and rodents. CPO is hydrolyzed by paraoxonase-1 (PON1), with protection determined by PON1 levels and the human Q192R polymorphism. To examine how the Q192R polymorphism influences fetal toxicity associated with gestational CPO exposure, we measured biomarker inhibition and fetal-brain gene expression in wild-type (PON1+/+), PON1-knockout (PON1-/-), and tgHuPON1R192 and tgHuPON1Q192 transgenic mice. Pregnant mice exposed dermally to 0, 0.50, 0.75 or 0.85 mg/kg/d CPO from gestational days (GD) 6 through 17 were sacrificed on GD18. Biomarkers of CPO exposure inhibited in maternal tissues included brain acetylcholinesterase (AChE), RBC acylpeptide hydrolase (APH), plasma butyrylcholinesterase (BChE) and carboxylesterase (CES). Fetal plasma BChE was inhibited in PON1-/- and tgHuPON1Q192, but not PON1+/+ or tgHuPON1R192 mice. Fetal brain AChE and plasma CES were inhibited in PON1-/- mice, but not in other genotypes.
