Purification of Human β and γ Actin from Budding Yeast

Project description:To facilitate biochemical studies of human cytoplasmic actins, Saccharomyces cerevisiaestrains were developed that express either human β-actin or γ-actin as their sole actin. Actins purified from these strains have biochemical properties similar to mammalian skeletal muscle α-actin, but display differential binding to some key actin regulators. Mass-spectrometry analysis shows that the human actins made from yeast generally retain their N-terminal methionines, which are modified by acetylation.

Project description:Actin is one of the most essential and abundant intracellular proteins, playing an essential physiological role as the major constituent of the actin cytoskeleton. Two cytoplasmic actins, beta- and gamma-actin, are encoded by different genes, but their amino acid sequences differ only by four conservative substitutions at the N-terminus, making it very difficult to dissect their individual regulation in vivo. The majority of actins are N-terminally acetylated, following the removal of N-terminal Met. Here, we analyzed beta and gamma cytoplasmic actin N-termini in different actin preparations.

Human Neuron Proteome Altered by Disrupted in schizophrenia 1 (DISC1) Mutations

Project description:Disrupted in schizophrenia 1 is a protein that is encoded by the DISC1 gene in humans. In coordination with a wide array of interacting partners, DISC1 has been shown to participate in the regulation of cell proliferation, differentiation, migration, neuronal axon and dendrite outgrowth, synapse formation and maturation, synaptic transmission, mitochondrial transport, fission and/or fusion, and cell-to-cell adhesion. Several studies have shown that unregulated expression or altered protein structure of DISC1 may predispose individuals to the development of schizophrenia, major depression, bipolar disorder, and other psychiatric conditions. The cellular functions that are disrupted by permutations in DISC1, which lead to the development of these disorders, have yet to be clearly defined and are the subject of current ongoing research.

Hai1 Mediates Inflammation Through MMP9

Project description:Adults heterozygous for the hi2217 retroviral insertion within the HAI locus (Mathias et al 2007 JCS) were in-crossed for RNA from hi2217 mutants and corresponding WT siblings. Adults heterozygous for the hi1520 retroviral insertion within the Clint1 locus (Dodd et al, 2009) were in-crossed for RNA from hi1520 mutants and corresponding WT siblings. Both hi2217 and hi1520 are mutants with epidermal defects and showing symptoms of chronic inflammation. In this study we aimed to compare the RNA expression changes between Wild-type siblings (a non-inflammatory status) and homozygous mutants (a chronic inflammatory status) for each mutant background (hi2217 and hi1520). Pools of 30-50 zebrafish embryos of WT siblings and hi2217 or hi1520 mutants were collected for RNA isolation. RNA from the hi2217 mutant embryos or hi1520 mutant embryos (test samples)was labelled with Cy5 and hybridized against Cy3-labelled RNA from the corresponding WT sibling embryos (reference samples) . These experiments were performed in biological triplicate.

Furan warheads for covalent trapping of weak protein-protein interactions: specific cross-linking of thymosin β4 to actin

Project description:We describe furan as a triggerable ‘warhead’ for site-specific cross-linking using the actin and thymosin β4 (Tβ4)-complex as model of a weak and dynamic protein-protein interaction with known 3D structure and with application potential in disease contexts. The presented in vitro validation of covalently acting ‘furan-armed’ Tβ4-variants provides initial proof to further exploit furan-technology for covalent drug design.

Proteomics identifies ubiquitin-proteasome targets and new roles for chromatin-remodelling in the Arabidopsis response to phosphate starvation

Project description:In order to identify new regulators of the phosphate (Pi) starvation signaling pathway in plants, we analyzed variation in the abundance of nuclear-enriched nuclear proteins isolated from Arabidopsis roots that depends on Pi supply. We used 2-D Fluorescence Difference Gel Electrophoresis and MALDI-TOF/TOF techniques for nuclear proteome separation, visualization and relative protein abundance quantification and identification. Pi-controlled proteins identified in our analysis included components of the chromatin remodeling, DNA replication, and mRNA splicing machineries. In addition, by combining Pi starvation conditions with proteasome inhibitor treatments, we characterized the role of the ubiquitin- (Ub)-proteasome system (UPS), a major mechanism for targeted protein degradation in eukaryotes, in the control of the stability of Pi-responsive proteins. Among Pi-responsive proteins, the histone chaperone NAP1;2 was selected for further characterization, and was shown to display differential nucleo-cytoplasmic accumulation in response to Pi deprivation. We also found that mutants affecting three members of the NAP1 family accumulate lower Pi levels and display reduced expression of Pi starvation-inducible genes, reflecting a regulatory role for these chromatin-remodelling proteins in Pi homeostasis.
