Identification of the proteins that stably or transiently interact with the nuclear poly(A)-binding protein Pab2 in the fission yeast Schizosaccharomyces japonicus using the proximity-labeling method TurboID.

ABSTRACT: We looked for the proteins that associate with Pab2, a nuclear poly(A)-binding protein, in Schizosaccharomyces japonicus through the proximity-labeling technology TurboID followed by mass spectrometry. We revealed that Pab2 interacts with two putative RNA-binding proteins, Red5 and Rmn1, and a yeast two-hybrid assay verified these interactions. We also found that Pab2 is associated with the CPSF (mRNA cleavage and polyadenylation specificity factor) component Ppn1, and the Pab2-Ppn1 interaction was confirmed by a yeast two-hybrid assay (and Ppn1 TurboID). These results indicate that Pab2 works with the three proteins mentioned above in Schizosacccharomyces japonicus.
