Project description:G. uraniireducens was isolated from a subsurface site in Rifle, CO undergoing in situ uranium bioremediation. Sediments from the Rifle site were heat-sterilized, amended with acetate to simulate in situ bioremediation conditions, and inoculated with G. uraniireducens. Gene transcript abundance in these cells using sediment Fe(III) and Mn(IV) oxides as the electron acceptor were compared with transcript levels in cells grown with fumarate as the electron acceptor. Additional comparisons were made between cells grown on synthetic Fe(III) or Mn(IV) oxides and cells grown on fumarate.

Comparative transcriptome analysis of manganese transportation in different leaves in soybean under manganese stress

Project description:Manganese (Mn) stress is known to be a major limitation for development of soybean, and legume crop productivity globally. However, very little information is available on the adaptive mechanisms, particularly in the important legume crop soybean (Glycine Max L.), which enable leaves to respond to high-Mn availability. Thus, to elucidate these mechanisms in soybean leaves at molecular level, we used an RNA sequencing approach to investigate transcriptomes of the leaves under Mn-sufficient and Pi-excessive conditions. Our investigation revealed that more genes showed altered expression patterns in old leaf than in young leaf under Mn excess, suggesting that the Mn excess-more-sensitive old leaf required expression change in a larger number of genes to cope with high-Mn stress than the Mn excess-less-sensitive young leaf. The functional classification of differentially expressed genes (DEGs) was examined to gain an understanding of how leaves respond to Mn stress, caused by soil Mn excess. As a result, more DEGs involved in nodulation, detoxification, nutrient/ion transport, transcriptional factors, key metabolic pathways, Mn remobilization and signalling were found in Mn-excessive induced old leaves than in Mn-excessive induced young leaves. Our findings have enabled the identification of molecular processes that play important roles in the acclimation of leaves to Mn excess, ultimately leading to the development of Mn-efficient soybean suitable for Mn-excessive soils.

Manganese homeostasis modulates fungal virulence and stress tolerance in Candida albicans

Project description:As transition metal availability is very limited inside the human host, fungal pathogens have evolved sophisticated mechanisms to uptake and utilize these micronutrients at the infection interface. While considerable attention was turned into iron, copper and zinc acquisition mechanisms and their importance in fungal fitness, less was done regarding either the role of Mn in infectious processes or the cellular mechanism by which fungal cells achieve their Mn-homeostasis. Here, we undertook a transcriptional profiling of the pathogenic fungus Candida albicans experiencing both Mn starvation and excess to comprehensibly capture biological processes that are modulated by Mn. We uncovered that Mn scarcity influence diverse biological processes associated with fungal fitness including morphogenetic switch, invasion of host cells and antifungal sensitivity. We also uncovered that Mn levels influence the abundance of iron and zinc emphasizing the complex crosstalk between ions metals. Deletion of SMF12, a member of Mn Nramp transporters confirmed its contribution to Mn uptake. In accordance with the RNA-seq data, smf12 was unable to form hyphae and damage host cells, and exhibited sensitivity to azole antifungals. We also found that unfolded protein response (UPR), likely activated by decreased glycosylation under Mn limitation, was essential to promote C. albicans growth. RNA-seq profiling of cells exposed to Mn excess uncovered that the UPR signaling was also essential to bypass Mn toxicity. Collectively, this study underscores the importance of Mn homeostasis in fungal virulence, and comprehensively provides a transcriptional portrait of biological functions that are modulated by Mn in a fungal pathogen.

Physiological and transcriptomic analyses of manganese toxicity response in Stylosanthes leaves

Project description:Despite its necessity, manganese (Mn) can causes phytotoxicity when present in excess. Stylo (Stylosanthes) is a dominant tropical legume with high Mn adaptability, but its Mn tolerance mechanisms are not well documented. This study integrated both physiological and transcriptome analyses of stylo in response to excess Mn toxicity. Results showed that stylo growth was decreased by excess Mn higher than 200 µM, as reflected by reductions in leaf chlorophyll and plant dry weight. Increases in enzyme activities of peroxidase (POD), ascorbate peroxidase (APX) and phenylalanine ammonia-lyase (PAL) and concentrations of secondary metabolites, including total phenols, flavonoids, tannins and anthocyanidins, were observed in stylo leaves at high Mn stress. Transcriptome analysis in stylo leaves resulted in identification of 2,471 up-regulated and 1,623 down-regulated genes under excess Mn toxicity. Among them, a set of differentially expressed genes (DEGs) involved in secondary metabolism, including PAL, chalcone synthase (CHS), chalcone isomerase (CHI) and flavonol synthase (FLS), were up-regulated in stylo under Mn toxicity, which were closely associated with the accumulation of secondary metabolites, suggesting that activation of secondary metabolism and corresponding gene expression might be important for stylo adaptation to Mn toxicity. Furthermore, a group of DEGs encoding transcription factors, such as genes belonged to C2H2 zinc finger transcription factor, WRKY, MYB and AP2 family, may be involved in stylo responses to Mn toxicity. Taken together, this study suggests that enhancing defense response and secondary biosynthesis pathway is adaptive strategy of stylo during Mn exposure, which might be regulated by complex transcriptional regulatory networks.

Combined Process of Biogenic Manganese Oxide and Manganese-Oxidizing Microalgae for Improved Diclofenac Removal Performance: Two Different Kinds of Synergistic Effects.

Project description:Biogenic manganese oxides (Bio-MnOx) have attracted considerable attention for removing pharmaceutical contaminants (PhCs) due to their high oxidation capacity and environmental friendliness. Mn-oxidizing microalgae (MnOMs) generate Bio-MnOx with low energy and organic nutrients input and degrade PhCs. The combined process of MnOMs and Bio-MnOx exhibits good prospects for PhCs removal. However, the synergistic effects of MnOMs and Bio-MnOx in PhCs removal are still unclear. The performance of MnOMs/Bio-MnOx towards diclofenac (DCF) removal was evaluated, and the mechanism was revealed. Our results showed that the Bio-MnOx produced by MnOMs were amorphous nanoparticles, and these MnOMs have a good Mn2+ tolerance and oxidation efficiency (80-90%) when the Mn2+ concentration is below 1.00 mmol/L. MnOMs/Bio-MnOx significantly promotes DCF (1 mg/L) removal rate between 0.167 ± 0.008 mg/L·d (by MnOMs alone) and 0.125 ± 0.024 mg/L·d (by Bio-MnOx alone) to 0.250 ± 0.016 mg/L·d. The superior performance of MnOMs/Bio-MnOx could be attributed to the continuous Bio-MnOx regeneration and the sharing of DCF degradation intermediates between Bio-MnOx and MnOMs. Additionally, the pathways of DCF degradation by Bio-MnOx and MnOMs were proposed. This work could shed light on the synergistic effects of MnOMs and Bio-MnOx in PhCs removal and guide the development of MnOMs/Bio-MnOx processes for removing DCF or other PhCs from wastewater.
