ABSTRACT: Three studies, including cohousing, Infliximab (anti-TNF)and Tocilizumab (anti-IL6r) and Lachnospiraceae strains treatments were performed to investigate how to restoring the dysbiotic microbiome in the Nlrp12-/- mice.

RNAseq analysis of gene expression in colorectal tumors of wild-type and Nlrp12-/- mice

Project description:Background: NLRP12 is a cytosolic pattern recognition receptor in the family of NOD-like receptor. NLRP12 has been shown to suppress colorectal tumorigenesis. However, the precise mechansim of NLRP12-mediated regualtion of colorectal cancer is unknown. Results: RNAseq data demonstrate higher expression of oncogenes, matrix degrading enzymes such as matrix metaloproteinases (MMPs), and genes involved in tumor invasion in Nlrp12-/- compared to wild-type tumors. Notably, several of the genes highly induced in Nlrp12-/- tumors are regulated by the Wnt/b-catenin pathway. Conclusion: NLRP12 negatively regualtes the Wnt/b-catenin pathway to suppress colorectal cancer progression and invasion.

Project description:Transcriptional analysis of immunological characteristics of petstore mice, C57Bl/6 laboratory mice, and C57Bl/6 laboratory mice cohoused with petstore. We hypothesized that cohousing would confer a basal transcriptional signature of immune activation to laboratory mice from petstore mice. Comparison of these data with existing human adult vs. neonatal PBMC expression profiling data (GSE27272) revealed close concordance of laboratory mice with human neonates, and of cohoused or petstore mice with human adults. Results highlight the impact of environment on the basal immune state and suggest that restoring physiological microbial exposure in laboratory mice could provide a relevant tool for modeling immunological events in free-living organisms, including humans. Total RNA was isolated from PBMC of C57Bl/6 laboratory mice, petstore mice and C57Bl/6 laboratory mice cohoused with petstore mice (at least after 60 days post-cohousing)and transcriptional comparison among 3 groups were performed.

Identification of baseline gene expression signatures predicting therapeutic responses to three biologic agents in rheumatoid arthritis: a retrospective observational study

Project description:The aim of this study was to identify the specific molecular biological features predicting the therapeutic outcomes of three bDMARDs, infliximab (IFX), tocilizumab (TCZ), and abatacept (ABT) by studying blood gene expression signature of patients prior to biologic treatment in a unified test platform. GSEA analyses showed that inflammasome genes were significantly upregulated in IFX’s NON-REM compared with its REM. In TCZ’s REM, B cell-specifically expressed genes were upregulated. RNA elongation, apoptosis-related, and NK cell-specifically expressed genes were upregulated in ABT’s NON-REM.
