Project description:The foraging gene in Drosophila, encoding PKG, is known for its importance as a natural variant affecting behavioral plasticity. Here, we demonstrate that it also affects metabolic plasticity: Rovers show a greater response to changes in their food environment than do sitters. Using metabolic profiling, we define foraging- and food-dependent changes in metabolites indicating that Rover store energy predominantly as lipids, whereas sitter variants store it as carbohydrates, Such changes are reflected in gene expression patterns, indicating an influence of foraging on regulators of insulin signaling. Combining for alleles with mutants of positive insulin signaling regulators makes Rover responses sitter-like, but does not change sitter responses. Collectively these findings suggest that the effect on metabolic plasticity of foraging works through the insulin signaling pathway. Project description:Social insect foragers may specialize on certain resource types. Specialization on pollen or nectar among honeybee foragers is hypothesized to result from associations between reproductive physiology and sensory tuning that evolved in ancestral solitary bees (the Reproductive Ground-Plan Hypothesis; RGPH). However, the two non-honeybee species studied showed no association between specialization and ovary activation. Here we investigate the bumblebee B. impatiens because it has the most extensively studied pollen/nectar specialization of any bumblebee. We show that ovary size does not differ between pollen specialist, nectar specialist, and generalist foragers, contrary to the predictions of the RGPH. However, we also found mixed support for the second prediction of the RGPH, that sensory sensitivity, measured through proboscis extension response (PER), is greater among pollen foragers. We also found a correlation between foraging activity and ovary size, and foraging activity and relative nectar preference, but no correlation between ovary size and nectar preference. In one colony non-foragers had larger ovaries than foragers, supporting the reproductive conflict and work hypothesis, but in the other colony they did not.

Treatments were leaving flies on lab food (control) or food-depriving them overnight with access to water. Three genotypes differed in foraging alleles: Rovers are homozygous for[R] (strain: BB), sitters homozygous for[s] (strain: ee, mutant sitters homozygous for[s2] (strain:s2) . Adult flies were 6-7 days post emergence. Three samples of equal numbers of male and females were collected per combination, frozen in liquid nitrogen, and heads were sieved for RNA extraction. This Gene by Environment design tests how presence or absence of food for a short period (overnight) affects gene expression in different foraging genotypes.

Project description:Bumblebees (Hymenoptera: Apidae) are important pollinating insects that play pivotal roles in crop production and natural ecosystem services. To achieve a comprehensive profile of accessible chromatin regions and provide clues for all possible regulatory elements in the bumblebee genome, we did ATAC-seq for Bombus terrestris samples derived from its four developmental stages: egg, larva, pupa, and adult, respectively. The sequencing reads of ATAC-seq were mapped to B. terrestris reference genome, and its accessible chromatin regions were identified and characterized using bioinformatic methods. Our study will provide important resources not only for uncovering regulatory elements in the bumblebee genome, but also for expanding our understanding of bumblebee biology.
