Project description:Extracellular vesicles (EVs) such as exosomes are key regulators of intercellular communication that can be found in almost all bio fluids. Although studies in the last decade have made great headway in discerning the role of EVs in many physiological and pathophysiological processes, the bioavailability and impact of dietary EVs and their cargo still remain to be elucidated. Due to its widespread consumption and high content of EV-associated microRNAs and proteins, a major focus in this field has been set on EVs in bovine milk and colostrum. Despite promising in vitro studies in recent years that show high resiliency of milk EVs to degradation and uptake of milk EV cargo in a variety of intestinal and blood cell types, in vivo experiments continue to be inconclusive and sometimes outright contradictive. To resolve this discrepancy, we assessed the potential postprandial transfer of colostral EVs to the circulation of newborn calves by analysing colostrum-specific protein and miRNAs, including specific isoforms (isomiRs) in cells, EV isolations and unfractionated samples from blood and colostrum. Our findings reveal distinct populations of EVs in colostrum and blood from cows that can be clearly separated by density, particle concentration and protein content (BTN1A1, MFGE8). Postprandial blood samples of calves show a time-dependent increase in EVs that share morphological and protein characteristics of colostral EVs. Analysis of miRNA expression profiles by Next-Generation Sequencing gave a different picture however. Although significant postprandial expression changes could only be detected for calf EV samples, expression profiles show very limited overlap with highly expressed miRNAs in colostral EVs or colostrum in general. Taken together our results indicate a selective uptake of membrane-associated protein cargo but not luminal miRNAs from colostral EVs into the circulation of neonatal calves.

Project description:Delivery of colostrum within the first several hours after birth is vital for establishing successful passive immunity in neonatal dairy calves. However, it is unclear whether the difference in colostrum feeding strategy can affect the development of the calf gastrointestinal tract. The aim of this study was to evaluate the effect of colostrum feeding time within the first 12 h after birth on the colonic mucosal immune system in neonatal calves using a genome wide transcriptome analysis.RNA sequencing based transcriptome analysis of colon tissues collected from twenty-seven male Holstein calves which were randomly assigned to one of three colostrum feeding strategies (immediately after birth (TRT0); 6 h after birth (TRT6); 12 h after birth (TRT12)) and were euthanized at 51 h of age detected 15935 ± 210, 15332 ± 415, and 15539 ± 440 expressed genes in groups, respectively. The core transcriptome of the colon in dairy calves included 12,678 genes, with enriched “cellular process” and “metabolic process” as the top three biological functions. Expression of 802 immune related genes were detected in the colon tissue. Principal component analysis of the transcriptomes did not display a clear separation by colostrum feeding strategy, and differential abundance analyses showed no significant difference in the expression of immune related genes among the treatments.Transcriptome analysis indicates that the development of the colonic mucosal immune system in neonatal calves may be independent of the timing of initial colostrum meal within 12 h after birth.

Project description:In cattle, the in vitro production (IVP) of embryos is becoming more relevant than embryos produced in vivo, i.e., after ovarian stimulation and embryo transfer (MOET). However, the effects of IVP on the developmental programming of specific organs in the postnatal calves are yet unknown. The objective of this study was to compare the hepatic and muscular transcriptomic modifications between IVP and MOET male calves of three months of age (n=4 per group). Tissue samples from liver and semitendinosus muscle were obtained at 3 months of age, and the extracted RNA was sequenced through RNAseq to determine differentially expressed genes (DEG; FDR<0.05). KEGG pathways enrichment analysis showed that DEG up-regulated in the liver and the muscle of the IVP calves were involved in oxidative phosphorylation and the tricarboxylic acid cycle. In contrast, ribosome and translation were up-regulated in the liver but down-regulated in the muscle of the IVP calves compared to the MOET calves (FDR<0.05). In conclusion, our findings indicated an altered hepatic and muscular energy regulation in phenotypically normal IVP calves compared to MOET calves.

Project description:Calves are highly susceptible to gastrointestinal infection with Cryptosporidium parvum (C. parvum), which can result in watery diarrhea and eventually death or impaired development. With little to no effective therapeutics, understanding the host’s microbiota and pathogen interaction at the mucosal immune system has been critical to identify and test novel control strategies. We used an experimental model of C. parvum challenge in neonatal calves to describe the clinical signs and mucosal innate immune and microbiota hallmarks in the ileum and colon during cryptosporidiosis and investigated the impact of supplemental colostrum feeding on C. parvum infection. The C. parvum challenged calves experienced clinical signs including pyrexia and diarrhea 5 days post challenge. These calves showed ulcerative neutrophil ileitis with a proteomic signature driven by inflammatory effectors, including reactive oxygen species and myeloperoxidases. Colitis was also noticed with an aggravated mucin barrier depletion and lack of full filled mucin granule in goblet cells. The C. parvum challenged calves also displayed a pronounced dysbiosis with a high prevalence of Clostridium species (spp.) and number of exotoxins, adherence factors, and secretion systems related to Clostridium spp. and other enteropathogens, including Campylobacter spp., Escherichia sp., Shigella spp., and Listeria spp. Daily supplementation with a high-quality bovine colostrum product mitigated some of the clinical signs and modulated the gut immune response and concomitant microbiota to a pattern more similar to that of healthy unchallenged calves.

Project description:In cattle, two of the most common technologies for embryo production are “in vivo”, after multiple ovulation and embryo transfer (MOET), or “in vitro” (IVP). IVP can sometimes impact fetal growth, resulting in large offspring, although most IVP calves are born apparently healthy. However, gene regulation changes in the organs involved in the developmental process in phenotypically normal postnatal calves are unknown and could indicate physiological programming. The objective was to compare the epigenomic and transcriptomic modifications in the liver, muscle, and organs of the hypothalamus-pituitary-gonadal and adrenal (HPGA) axes between IVP and MOET male calves of three months of age. Pregnancies were monitored until delivery. Male calves were maintained in similar conditions during the postnatal period until euthanasia at around 100 days of age. There were no differences regarding body weight at birth and growth rate among the animals in both groups at this age.

Project description:Extracellular vesicles (EVs) and their microRNA (miRNA) cargo have been suggested as potential biomarkers for mammary gland health in cattle. However, milk is a dynamic fluid, and its biologically active components, including miRNAs, could be subject to changes throughout the day. The current study aimed to evaluate the circadian fluctuation of milk EVs miRNA cargo to assess the feasibility of milk EVs as future biomarkers for mammary gland health management. Milk from four healthy dairy cows was collected manually from one quarter during four consecutive days in the two daily milking sessions in the morning and the afternoon. The SCC was determined, and the milk EVs were isolated from skimmed milk. The presence of EVs was confirmed by transmission electron microscopy (TEM), tunable resistive pulse sensing (TRPS), and western blot (WB). Small RNA libraries were produced from 10 ng of extracted RNA and sequenced in two lanes of a HiSeq2500. The heterogeneity and integrity of EVs and the protein EV markers CD9, CD81, and TSG101 were confirmed by TEM and WB. The sequencing revealed that despite daily fluctuation in other milk components, like the somatic cells during milking sessions, the miRNA cargo abundance in milk EVs stayed constant. Our results show that the miRNA cargo of milk EVs is very stable regardless of the hour of the day, supporting their potential use as diagnostic markers for mammary gland health.

Project description:In this study, we show that muscle-specific inactivation of FBXW7 elicits striking defects in postprandial glucose metabolism and failure to maintain skeletal muscle mass in adult mice. Further, mice lacking FBXW7 exhibited impaired endurance capacity and exacerbated HFD-induced insulin resistance and postprandial hyperglycemia. At the mechanistic level, RNAseq and quantitative proteomic analysis revealed global effects of FBXW7 deficiency on skeletal muscle transcriptome and proteome. This work illustrates a prominent role of FBXW7 in integrating postprandial nutritional signals to coordinate glucose metabolism and muscle mass maintenance.
