Project description:We have captured 100,000 single cells for single-cell RNAseq from whole mouse embryos during gastrulation and organogenesis, spanning days 6.5 to 8.5 of development, including embryonic and extraembryonic tissues. Cells were sampled every six hours, providing a continuous molecular characterisation of these processes. Cell libraries were prepared using the 10X Genomics Chromium platform.

Gene Expression Differences in p53-Deficient Day 8.5 Mouse Embryos

Project description:Neural tube defects (NTDs) are one of the most common human birth defects, with a prevalence of approximately 1 in 1000 live births in the United States. In animal studies, deletion of p53 leads to a significant increase in embryos that exhibit exencephaly. Whereas several studies have closely investigated the morphological changes of p53-deficient embryos, there is no study that has reported the molecular-level alternations in p53-deficient embryos. Here we use microarray approach to find genes modified by deletion of p53 in day 8.5 mouse embryos to identify genes that may be involved in the mechanisms underlining NTDs and begin to define the developmental role of p53 in the etiology of NTDs. S. erythraea R5 Fall 2002 RNA Timecourse

Project description:Saccharopolyspora erythraea NRRL2338 (wildtype) and K41-135 (overproducer strain from Kosan Biosciences, KOVP) were grown in shake flask cultures in rich medium R5. RNA samples were harvested for each strain over 5 days. 12 h samples taken for each strain were used as their respective reference ("time zero") samples. The genomic DNA control compares NRRL2338 gDNA to NRRL2338 12 h RNA to show relative gene expression at the start of the timecourse. The 12 h RNA control compares initial gene expression between the wildtype and overproducer strains. Groups of assays that are related as part of a time series. Keywords: time_series_design
