Project description:Brown adipose tissue (BAT) plays a central role in energy homeostasis through non-shivering thermogenesis. Besides, recent human studies using 18FDG-PET/CT imaging demonstrated that BAT acts as a significant metabolic-sink for glucose. Notably, these functions in BAT decrease with age: however what regulates this process remains poorly understood. To this end, we employed RNA-seq to identify the transcriptional changes in BAT of young and old mice.

Project description:In our investigation of age-related changes in brown adipose tissue, we have identified two distinct populations of brown adipocytes characterized by high and low expression of uncoupling protein 1 (UCP1). We observed that with aging, the proportion of UCP1-low adipocytes increases, and these cells notably express genes associated with pyroptosis, an inflammatory form of programmed cell death.

This study reports analysis of transcriptome profiles in brown adipose tissue, liver, white adipose of mice in repsonse to 24 hour cold exposure Total RNA was extracted from brown adipose, whie adipose and liver from cold treated and control plants. 3 replicates of brown adipose and white adipose and 2 replicates of liver samples.

Prdm16 is a transcription factor that regulates the thermogenic gene program in brown and beige adipocytes. However, whether Prdm16 is required for the development or physiological function of brown adipose tissue (BAT) in vivo has been unclear. By analyzing mice that selectively lacked Prdm16 in the brown adipose lineage, we found that Prdm16 was dispensable for embryonic BAT development.
