Project description:Natural Helper cells constitute a unique lineage of Th2-cytokine producting innate lymphocytes, here we characterize the gene expression profile of non-stimulated or PMA/ionomycin-stimulated Natural Helper cells from naive C57Bl/6 mouse lungs. We used microarrays to detail the gene expression profile of stimulated and unstimulated lung Natural Helper cells in mice. Lineage(-)Sca-1(+)c-Kit(-/low)CD127(+)CD25(+) Lung Natural Helper cells were purified from naive 6-8 week old B6 mice by FACS. RNA was extracted immediately from un-stimulated Natural Helper cells after FACS. Stimulated Natural Helper cells were cultured with PMA/ionomycin for 3 days followed by RNA extraction.

Project description:Post-transcriptional gene regulation by mRNA-binding proteins (RBPs) ensures an important layer to control protein outcome. It includes many fundamental processes, ranging from splicing to transport, decay and translation. Although recent technical developments allow the identification of mRNA-binding proteins, the total RBPome of different primary immune cells remains unknown. Here, we performed an RBP capture of the two major primary T helper cell types in mouse: T helper effector-like and regulatory-like (Foxp3+) cells. The two datasets show a high overlap of ~80% to reveal a high confidence common T helper cell RBPome of 240 proteins. 60% of those proteins have not been previously associated with any immune function. Most importantly, 11 RBPs were not previously identified through any mammalian mRNA-capture approach. These novel RBPs include some well-known T cell proteins associated with non-immune functions such as Stat1 and Vav1. Overall our data suggest an underestimated complexity of gene regulatory pathways.

IL 12R expression is required for Th1, IL-4RM-NM-1 for Th2, and IL-6RM-NM-1/gp130 for Th17 differentiation to allow responsiveness to IL-12, IL-4, and IL-6, respectively. IL-2 via STAT5 controls Th2 differentiation by regulating the Th2 cytokine gene cluster and Il4ra expression. Here we show that IL-2 regulates Th1 differentiation, inducing STAT5-dependent IL-12RM-NM-22 and T-bet expression, with impaired human Th1 differentiation when IL-2 was blocked. Th1 differentiation was also impaired in mouse Il2-/- T cells but restored by IL-12RM-NM-22 expression. Consistent with IL-2M-bM-^@M-^Ys inhibition of Th17 differentiation, IL-2 decreased Il6ra and Il6st/gp130 expression, and Il6st augmented Th17 differentiation even when IL-2 was present. Thus, IL-2 influences T-cell differentiation by modulating cytokine receptor expression to help specify/maintain differentiated states. Project description:Post-transcriptional gene regulation is complex, dynamic and ensures proper T cell function. The targeted transcripts can simultaneously respond to various factors as evident for Icos, an mRNA regulated by several RNA binding proteins (RBPs), including Roquin. However, fundamental information about the entire RBPome involved in post-transcriptional gene regulation in T cells is lacking. Here, we applied global RNA interactome capture (RNA-IC) and orthogonal organic phase separation (OOPS) to human and mouse primary T cells and identified the core T cell RBPome. This defined 798 mouse and 801 human proteins as RBPs, unexpectedly containing signaling proteins like Stat1, Stat4 and Vav1. The data represent an atlas of RNA-binding proteins in human and mouse T helper cells as a resource for studying higher order post-transcriptional gene regulation.

Project description:The severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2 or CoV-2) is the agent of a major global outbreak of respiratory tract disease known as COVID-19. CoV-2 infects the lungs and may cause several immune-related complications such as lymphocytopenia and cytokine storm which are associated with the severity of the disease and predict mortality. The mechanism by which CoV-2 infection may result in immune system dysfunction is not fully understood. Here we showed that CoV-2 infects human CD4+ T helper cells, but not CD8+ T cells. CoV-2 is found in the blood and bronchoalveolar lavage T helper cells of severe COVID-19 patients. We demonstrated that CoV-2 spike glycoprotein (S) (sCoV-2) directly binds to the CD4 co-receptor, which in turn mediates the entry of CoV-2 in T helper cells in a mechanism that also requires ACE2 and TMPRSS2. Once inside T helper cells, CoV-2 assembles viral factories, impairs cell function and may cause cell death. CoV-2 infected T helper cells express higher amounts of IL-10, which has been associated with viral persistence and disease severity. Thus, CD4-mediated CoV-2 infection of T helper cells may explain the poor adaptive immune response of many COVID-19 patients.

Project description:Pathogenesis of atherosclerosis results from the interactions between disrupted lipid homeostasis and immune response but the molecular bridges between the major players are still a matter of controversy. We investigated the systemic effect of inflammatory cytokine tumor necrosis factor alpha (TNF-α), a well established anorexia agent, in livers; of mice exposed to 24h-cytokine/starvation. 10% of genes of the Agilent 10kb cDNA array is detected above level of detection in the mouse liver. 12% of these is modulated upon starvation while 22% is altered by TNF-α. Experiment Overall Design: Twelve mice (C57BL/6, Harlan) were randomly assigned into three groups. Three animals were i.v. injected by human recombinant TNF-α (30 µg/animal), a dose that is known to have a significant effect in Experiment Overall Design: mice without leading to mortality. Food was withdrawn after application of TNF-alpha (TNF/starvation group). Further, for three animals which were treated with corresponding volume (200µl) of saline a food was withdrawn (starvation group). Three animals from control group had free access to food throughout the study. Mice were sacrificed 20 hours later. Livers for RNA analysis were stored in RNAlater (Quiagen) according to manufacture's instruction. RNA of individual treated mouse was hybridized versus control sample which is obtained by pooling of control RNAs.
