Project description:In nature, plants are frequently subjected to multiple biotic and abiotic stresses, resulting in a convergence of adaptive responses. We hypothesized that hormonal signalling regulating defences to different herbivores may interact with drought response, causing distinct resistance phenotypes. To test this, we studied hormonal and transcriptomic responses of Solanum dulcamara subjected to drought and herbivory by the generalist Spodoptera exigua (BAW) or the specialist Leptinotarsa decemlineata (CPB). Bioassays showed that plants under drought became more resistant to BAW, but not to CPB. While drought did not alter BAW-induced hormonal responses, it enhanced CPB-induced accumulation of jasmonic acid and salicylic acid (SA) as well as supressed ethylene (ET) emission. Microarray analyses showed that under drought BAW herbivory enhanced several herbivore-induced responses, including cell-wall remodelling and metabolism of carbohydrates, lipids and secondary metabolites. In contrast, CPB herbivory enhanced several photosynthesis-related and pathogen responses in drought-stressed plants. This may divert resources away from the production of effective defences and increase tissue nutritive value. In conclusion, while BAW suffers from the drought-enhanced defences, CPB may benefit from effects of the enhanced SA and reduced ET signalling. This suggests that the fine-tuned interaction between the plant and its specialist herbivore is sustained under drought.

Project description:Plant drought stress response and resistance are complex biological processes that merit systems-level analyses to dissect drought stress encountered by crops in the field. We have used gene expression profiling of Arabidopsis plants subjected to a controlled, sublethal, moderate drought (mDr) treatment to characterize early and late response to drought. We have also compared these profiles to those from plants treated with soil water deficit (progressive) drought (pDr) to reveal acclimation responses in plants.

Project description:Water availability is the biggest single limitation on plant productivity worldwide. In Arabidopsis, adjustments to drought stress, involving changes in metabolism and gene expression drive increased drought tolerance and initiate diverse drought avoidance and escape responses. To address regulatory processes that integrate these complex responses we hypothesised that we needed to identify genes that govern early responses to drought. To this end, we produced a high-resolution time series transcriptomics dataset, coupled with detailed physiological and metabolic analyses of plants subjected to a slow transition from well-watered to the onset of drought conditions. 1825 differentially expressed genes (DEGs) were identified which showed no significant enrichment in gene ontology terms associated with dehydration responses and abscisic acid (ABA) regulation, confirming that the gene expression time series had targeted events prior to severe drought stress. Initial changes in gene expression coincided with a drop in carbon assimilation, not the later increase in foliar ABA content. Thus the early physiological and gene expression responses to drought were not driven by changes in leaf ABA content. In order to identify gene regulatory networks (GRNs) linked to early events, we used Bayesian network modelling of differentially expressed transcription factor (TF) genes. This approach identified AGAMOUS-LIKE 22 as key hub gene in a TF GRN. AGL22 is involved in the transition from vegetative state to flowering. Loss of AGL22 expression affected flowering time and drying rate providing a link between early changes in metabolism and the subsequent initiation of developmental responses to stress that govern plant productivity.

Project description:With frequent fluctuations in global climate, plants often experience co-occurring dry-wet cycles and pathogen infection and this combination adversely affects plant survival. In the past, some studies indicated that morpho-physiological responses of plants to the combined stress are different from the individual stressed plants. However, interaction of drought stressed or drought recovered plants with pathogen has not been widely studied at molecular level. Such studies are important to understand the defense pathways that operate as part of combined stress tolerance mechanism. In this study, Arabidopsis plants were exposed to individual drought stress (soil drying at 40% FC, D), Pseudomonas syringae pv tomato DC3000 (PStDC3000), infection and their combination. Plants recovered from drought stress were also exposed to PStDC3000. Beside we have also infiltrated P. syringae pv tabaci (PSta, non-host pathogen) individually or in combination with drought stress. Using Affymetrix WT gene 1.0 ST array, global transcriptome profiling of plants leaves under individual drought stress and pathogen infection was compared with their combination. Results implicate that plants exposed to combined drought and pathogen stress experience a new state of stress where each combination of stressor and their timing defines the plant responses and thus should be studied explicitly. Global transcriptional analysis in Arabidopsis leaves exposed to individual and combined drought and pathogen stress.

Project description:Drought is a stressor for many soil-inhabiting organisms. Although plants have been extensively investigated for drought-adaptive mechanisms, little information is available for fungi. Antioxidants are especially relevant, since desiccation is accompanied by an excessive intracellular production of reactive oxygen species. Riboflavin (vitamin B2) is one antioxidant regulating drought tolerance in plants. A similar function may exist in fungi. Here, we examined the respiratory and transcriptional responses of Agaricus bisporus to drought and the impact of riboflavin. Mesocosm experiments with four groups were established: hyphae were treated with or without 50 µM riboflavin under drought or no drought conditions. Drought increased riboflavin content in hyphae about 5 times with, but also without, addition of riboflavin. Without addition of riboflavin, fungal respiration decreased by more than 50% at water potentials of about -20 MPa. With addition of riboflavin, respiration remained about 2-3 times higher. The transcriptional responses to only drought or only riboflavin strongly overlapped and were mainly based on factors regulating transcription and translation. This was even stronger in combined treatments. Riboflavin induced protective mechanisms in drought-stressed hyphae. Most pronounced was the methylglyoxal (cytotoxic by-product of glycolysis) detoxifying of lactoylglutathione lyase. Thus, our data suggest a stress-priming function and a role of riboflavin in drought responses of A. bisporus.

Project description:We sought to generate a comprehensive proteomic profile for a Populus plant that captures protein abundance behavioral differences in mature leaves under various water deficit conditions. The sudden infliction of severe drought is unlikely to reflect what naturally happens to soil-grown plants. Therefore, rather than exposing plants to severe dehydration via the application of concentrated osmotica (e.g., polyethylene glycol or mannitol), we aimed to monitor plants exposed to prolonged drought as well as plants frequently experiencing cyclic drought. For the progressive drought experiment, herein referred to as acute drought, water will be withheld from the plants for a certain period of time until symptoms of wilting are observed. From this drought treatment, protein abundance changes that occur because of declining water potentials that consequently lead to worsening plant health will provide new insights to biological pathways in response to severe drought conditions. On the other hand, as a consequence of simulating a more natural condition, the cyclic drought treatment will provide novel insights into drought recovery and coping mechanisms. As first noted in a meta-analysis of microarray experiments comparing different water deficit-related treatments3, one consequence of analyzing multiple treatment conditions is that we expect only a few differentially abundant proteins to be common to both treatments. Therefore, we implemented a bioinformatic framework to provide an overall integrative picture of the conserved drought markers but also meaningful divergences in functional behavioral responses between the two experiments.

Project description:We use Solanum dulcamara subjected to drought or flooding and damaged by Spodoptera exigua to analyze such interactions at multiple levels. Drought and herbivory caused comparable effects on S. dulcamara physiological response, which was reflected by a considerable overlap in S. dulcamara transcriptomic profiles. This included many defense responses and genes involved in biosynthesis of secondary metabolites that were induced by drought and herbivory but repressed by flooding. Furthermore, combination of drought and herbivory additively induced a part of these herbivore-induced responses suggesting that drought-stressed plants were more resistant. Our study provides concrete evidence of how abiotic stresses differentially affect the plant complex hormonal interactions to fine-tune plant responses to insects.
