Project description:Lake Apopka (Fl, USA) experienced heavy uses of organochlorine pesticides (OCPs) in the 1950s-1970 during an intense agricultural period. As a result, the region remains heavily contaminated and was declared a Superfund site by the US Environmental Protection Agency. Aquatic organisms inhabit the freshwater systems in and around Lake Apopka, however the impacts of sub chronic exposure to OCPs in this natural environment are not known. The objectives of this study were to determine the effects of the contamination on the reproductive axis of largemouth bass (LMB) (Micropterus salmoides). In October (2007), healthy LMB were stocked into natural mesocosms and remained in mesocosms before being sampled in January 2008 (at early oogenesis). Additional LMB were placed into mesocosms for 2 months in February (2008) and sampled in April (2008) (oocyte maturation). LMB placed in these mesocosms for four months had a 2-20X higher contaminant load for OCPs (e.g. DDE, dieldrin, methoxychlor) than LMB collected from reference sites. Gonadosomatic index for LMB collected in April from the mesocosms were not different that LMB from reference sites. Vitellogenin levels in LMB collected from the mesocosms in January and April were not significantly different than fish collected from Welaka in late vitellogenin or early maturation respectively. Steroids were depressed in mesocosm fish. Microarray analysis revealed that the expression profiles of genes in the LMB ovary were unique compared to LMB collected form reference sites. Transcripts that showed altered abundance in LMB from the mesocosms were insulin-like growth factor I and steroidogenic acute regulatory protein. Interestingly, differentially expressed transcripts showed a significant and positive correlation for LMB sampled in January and April despite the 3 month period in between samplings. Sub-network enrichment analysis for cellular processes showed that retinoic acid metabolism and germ-cell development were decreased in mesocosm-exposed fish but processes such as vitellogenesis, amino acid catabolism, granulosa cell function, vitamin D metabolism, and hormone biosynthesis were increased in mesocosm-exposed fish. These data suggest that (1) LMB from the mesocosms are exhibiting unique gene profiles that may impair normal reproduction and that (2) microarray analysis in the field can provide site specific information by discriminating LMB from reference and polluted sites.

Project description:Transcriptional profiling of the digestive gland tissue of female mussel Mytilus galloprovincialis exposed to copper along with a temperature gradient Background: Global warming is a major factor that may affect biological organization, especially in marine ecosystems and in coastal areas that are particularly subject to anthropogenic pollution. We evaluated the effects of simultaneous changes in temperature and copper concentrations on lysosomal membrane stability of the blue mussel Mytilus galloprovincialis (Lam.). Temperature and copper exerted additive effects on lysosomal membrane stability, exacerbating the toxic effects of metal cations present in non-physiological concentrations. Mussel lysosomal membrane stability was positively related toscope for growth, indicating possible effects of increasing temperature on mussel populations in metal-polluted areas. To clarify the molecular response to environmental stressors, we used a cDNA microarray with 1,700 sequences to measure the relative transcript abundances in the gills of mussels exposed to copper (40µg/L) and a temperature gradient (16°C, 20°C, and 24°C). In animals exposed only to heat stress, hierarchical clustering of the microarray data revealed three main clusters, which were largely dominated by down-regulation of translation-related differentially expressed genes, drastic up-regulation of folding protein-related genes, and genes involved in chitin metabolism. The response of mussels exposed to copper at 24° C was characterized by an opposite pattern of the genes involved in translation, most of which were up-regulated, as well asthe down-regulation of genes encoding heat shock proteins and microtubule-based movement proteins. Our data provide novel information on the transcriptomic modulations in mussels facing temperature increases and high copper concentrations; these data highlight the risk of marine life exposed to toxic chemicals in the presence of temperature increases due to climate change.

Transcriptomic profiles from laboratory-exposed and wild fish sampled along a contamination gradient were compared. During laboratory experiments, fish were exposed to pollutants or natural factors under levels that were closed to those found in the sampling sites. A strong difference was observed between the transcriptomic patterns of wild and laboratory-exposed animals, suggesting a general stress induced by captivity in laboratory. Surprisingly, animals acclimatized to cold temperature clustered with fish from the most contaminated sites and shared common genes involved notably in epigenetic mechanisms. A possible energy saving strategy was suggested in cold-acclimated animals. This result may suggest that animals chronically exposed to contaminants develop a general defense/adaptation strategy against pollution through a general shunt in their basal metabolism, whatever the type of contaminants.

Seven sampling wild sites were selected in Canada (St. Jean, Sud-Ouest, St. FranM-CM-'ois, St. Pierre) and in France (Dordogne, Garonne, Gironde) on the basis of their known gradient of contamination by metallic and organic pollutants previously described in Baillon et al., in press. Each sample were hybridized with a same reference on array (30 samples from reference site (Certes, France) collected in spring 2011 and 2012. Animals from laboratory exposures were described below.
