Project description:The soft coral Scleronephthya gracillimum is an azooxanthellate octocoral order Alcyonacea. In this study, stress responses to increased seawater temperature and marine acidification were investigated using a microarray. The S. gracillimum microarray was constructed. The S. gracillimum microarray was constructed after RNA-seq. Oligonucleotides were picked from UniGene of S. gracillimum and the clones were annotated using Blast.

Project description:Cultures of Synechococcus (WH8102) were grown in polycarbonate bottles with a continuous culturing method with an artificial seawater medium. The experimental design included two concentration ratios of macronutrients (NO3-:PO43- = 1.7 and 80) and 3 levels of temperature (20, 24 and 28°C). Light was supplied with white fluorescent lamps at ~125 umol quanta m-2 s-1 on a 12h:12h light:dark cycle. Cultures were stable 38 days after initiation and cells were collected on days 38, 43, 47, 50 and 57 for particulate organic matter, nutrient analysis and cellular proteins.

Microcosms exposed to artificial sunlight were compared to dark controls. Three experiments were done on three separate days.
