Project description:We have generated immune-enhancing neutrophils by culturing murine primary bone marrow derived neutrophils with either super-low dose of LPS. Immune-enhancing neutrophils preferentially express co-stimulatory molecules such as CD74, CD44 and CD86, and exhibit reduced expression of CD11b. Purified bone marrow neutrophils were treated with PBS or 100 pg/ml LPS overnight in the presence of GM-CSF, and harvested for scRNAseq analysis to examine their profiles of gene expression.

Project description:The goals of this study is to identify the differential expressed genes in heart of C57BL/6 mice with or without LPS treatment, and compare the differential expressed genes in the heart of LPS injection C57BL/6 mice after Xijiao Dihuang decoction(XJDHT) treatment. Briefly, the mice were randomly divided into 3 groups: control, LPS and LPS + XJDHT groups (n=4 for each group). Mice in Control and LPS groups were intraperitoneal injection with saline and 10mg/kg of LPS respectively, and orally administrated with saline; the mice in LPS + XJDHT groups were intraperitoneal injection with LPS (10 mg/kg) and orally administrated with 50 mg/kg /day of XJDHT for total 3 days. Then the heart were used to identify differentially expressed genes among different groups.

Project description:MicroRNAs (miRNAs) play important roles in intestinal diseases; however, the role of miRNAs during weaning stress is unknown. In our study, six jejunal small RNA libraries constructed from weaning piglets at 1, 4 and 7 d after weaning (libraries W1, W4 and W7, respectively) and from suckling piglets on the same days as the weaning piglets (libraries S1, S4 and S7, respectively) were sequenced using Solexa high-throughput sequencing technology. Overall, 260 known swine miRNAs and 317 novel candidate miRNA precursors were detected in the six libraries. The results revealed that 16 differentially expressed miRNAs were found between W1 and S1; 98 differentially expressed miRNAs were found between W4 and S4 (ssc-mir-146b had the largest difference and ssc-mir-215 had the highest expression level); and 22 differentially expressed miRNAs were found between W7 and S7. Sequencing miRNA results were validated using RT-qPCR. Approximately 12,819 miRNA-mRNA interactions corresponding to 4,250 target genes were predicted. The biological analyses revealed that the differentially expressed miRNAs regulated small intestinal metabolism, stressful responses, cellular and immune functions and miRNA biosynthesis in piglets. Therefore, the small intestine miRNA transcriptome was significantly different between weaning and suckling piglets; the difference varied with the number of days after weaning.

Project description:The research on alternative and sustainable feed ingredients is a challenge to reduce feed-food competition between humans and monogastrics, in particular pigs. Former food products (FFPs) drop out from the industrial production of food such as pasta, bread, snacks, and chips. They have a high nutritional and energetic value and represent an alternative and sustainable feed ingredient. The aim of this study was to apply omics tools to assess the impact of the dietary inclusion of FFPs in pigs’ diet. For the in vivo trial, thirty-six Swiss Large White male castrated pigs were randomly assigned to three dietary groups: control (CTR), 30% replacement of CTR diet with salty FFPs (SA), 30% replacement of CTR diet with sugary FFPs (SU). The trial lasted from the start of the growing phase (22.4 ± 1.7 kg) until slaughtering phase (110 ± 3 kg). After slaughtering, blood and liver tissue samples were collected and processed according to the omics approaches used. The number of differentially regulated proteins in each comparison matrix (SA/SU vs. CTR) indicated a marginal impact on liver function, as measured by proteomics on tissue samples. The peptidomics investigation on plasma samples showed low variability between the peptidome of the three dietary groups and identified three possible bioactive peptides associated with anti-hypertension in the SA dietary group. To conclude, the safety and the limited impact of the SA and SU diets on liver proteome and plasma peptidome strengthened the idea of recycling FFPs as feed ingredients to make pig production more sustainable.
