Project description:We tested the hypothesis that differential gene expression in whole blood will reveal candidate blood biomarkers for exposure to agricultural pesticides and herbicides. Blood gene expression in male Latino farmworkers, where chronic pesticide exposure is occupational, was compared to blood gene expression in age and gender matched Latino manual workers. We identified an expression signature for farmwork, differential expression in genes that correlated with levels of urinary pesticide metabolites, alterations in axonal guidance pathways and statistical models that link farmworker differential expression to Parkinson's disease.

Project description:Fungal group III histidine kinases are the molecular targets of some classes of fungicides. In contrast to the yeast Saccharomyces cerevisiae, the fungal pathogen Candida albicans possesses a group III histidine kinase, CaNik1p, also called Cos1p. To investigate the function of CaNIK1, the gene was expressed in S. cerevisiae. The transformants became susceptible to antifungal compounds to which the wild-type strain is resistant. The susceptibility was related to the activation of the MAP kinase Hog1p of the osmotic stress response pathway. Gene expression analysis revealed a strong overlap of the responses to osmotic stress and to fludioxonil at early time points. While the response to fludioxonil persisted, the response to osmotic stress was diminished with time.

Project description:Plant food production is severely affected by fungi; to cope with this problem, farmers use synthetic fungicides. However, the need to reduce fungicide application has led to a search for alternatives, such as biostimulants. Rare-earth elements (REEs) are widely used as biostimulants, but their mode of action and their potential as an alternative to synthetic fungicides have not been fully studied. Here, the biostimulant effect of gadolinium (Gd) is explored using the plant-pathosystem Arabidopsis thaliana–Botrytis cinerea . We determine that Gd induces local, systemic, and long-lasting plant defense responses to B. cinerea, without affecting fungal development. The physiological changes induced by Gd have been related to its structural resemblance to calcium. However, our results show that the calcium-induced defense response is not sufficient to protect plants against B. cinerea, compared to Gd. Furthermore, a genome-wide transcriptomic analysis shows that Gd induces plant defenses and modifies early and late defense responses. However, the resistance to B. cinerea is dependent on JA/ET-induced responses. These data support the conclusion that Gd can be used as a biocontrol agent for B. cinerea. These results are a valuable tool to uncover the molecular mechanisms induced by REEs.

Project description:Gene expression changes and polymorphisms were investigated in genetically related mosquito lines maintained for 30 generations under four different selection regimes. All mosquito line derived from the same parental line (Anopheles gambiae Tiassale line from Ivory coast). Selection regimes were as follows: i) no selection pressure (Control line), ii) selection at each generation at larval stage with a mixture of insecticides used against agricultural pests (Ins line); iii) selection at each generation at larval stage with a mixture of herbicides and fungicides used on crops (Herb line), iv) selection at each generation at larval stage with a mixture of both insecticides and non-insecticide compounds (compounds ii + iii, Mix line). The transcriptome of each line was sequenced using RNA-seq with four replicates per condition.

Project description:Septoria leaf blotch is a worldwide threat for wheat and mainly controlled by the application of synthetic fungicides. The fungal pathogen responsible for this disease, Zymoseptoria tritici, was shown as highly adaptable to its host plant, but also to fungicide challenge. Over the past decades it developed resistance to most fungicides due to target site modifications. Recently isolated strains showed cross-resistance to diverse fungicides and to unrelated drugs, suggesting a resistance mechanism that seems rarer in phytopathogenic fungi, known as multidrug resistance (MDR) in other organisms. In this study we show for two Z. tritici MDR strains, MDR6 and MDR7, enhanced prochloraz efflux sensitive to the modulators amitryptiline and chlorpromazine. Efflux was also inhibited by verapamil in the MDR7strain. Transcriptomics revealed several overexpressed transporter genes in both MDR strains, out of which the expression of the MgMFS1 transporter gene was the strongest and constitutively high in tested MDR field strains. Its inactivation in the MDR6 strain abolished resistance to fungicides with different modes of action revealing its involvement in the MDR phenomenon in Z. tritici.

Project description:The entomopathogenici fungus B. bassiana is of great imporatnce in biological control of insect pests. In integrated pest management (IPM) program, unsaturated fatty acids and their derivatives are used as fungicides. The compatibility of mycoinsecticides with fungicides has attracted more and more attention. The genome-wide exprssion analysis involved in fungal resonse to unsaturated fatty acids was analyzed by using high throughput sequencing (RNA-Seq) at different concentrations of linoleic acid. Our transcriptional profiles revealed that numerous differentially expressed genes (DEGs), of which involved in metabolism, cell transport and cell rescue, were significantly involved in the tolerance of B. bassiana to linoleic acid stress.

Project description:Wheat leaf rust is a serious fungal disease of wheat that causes annual losses and necessitates using fungicides for effective disease management. It is caused by Puccinia triticina which spreads by means of airborne urediniospores. When these germinate on the leaf surface, they form germ-tubes which enter the leaf through open stomates. Spores and germ-tubes represent the first fungal structures that the host can perceive during a rust infection. They therefore contain proteins that could potentially trigger early host defense responses. Using 2-DE to separate this proteome, we produced gels containing 173 spots in the pI range of 4-7 and identified 123 proteins. These were predominantly proteins involved in metabolic and cellular processes, but with a large number (77%) of novel proteins that could not be identified through homology matching Twenty four of these showed no homology to wheat sequences, making them good candidate PAMPs.
