Project description:The present study aims to characterise proteins bound to circuits collected from children on extracorporeal membrane oxygenation (ECMO). ECMO circuits were collected from 6 patients. Quantification of concentrations for proteins bound to the ECMO circuit samples was performed using bicinchoninic acid (BCA) protein assay, whilst characterisation of the bound proteome was performed using Data-independent acquisition Mass Spectrometry (DIA-MS). Reactome Over-representation Pathway Analyses tool was used to identify functional pathways corresponding to the common proteins bound to circuits across all patients.

The characterisation of protein and cellular binding in paediatric extracorporeal membrane oxygenation (ECMO) circuits using proteomics, immunofluorescence, and scanning electron microscopy

Project description:Extracorporeal membrane oxygenation (ECMO) has been increasingly applied to both adult and paediatric patients worldwide. However, continuous contact between blood and foreign surface of the ECMO circuit may contribute to haemostatic, inflammatory and other physiological disturbances during ECMO. Although previous studies have extensively investigated blood samples from patients on ECMO, the protein and cellular binding to the ECMO circuit has largely been overlooked as an additional factor that could contribute to differences in clinical outcomes. The detailed protocol described in this paper enables the comprehensive characterisation of protein and cellular binding in paediatric ECMO circuits, which could extend our knowledge of the pathophysiology of circuit binding and provide guidance for improved ECMO circuit design.

Transciptomic signature of chronic antibody-mediated kidney rejection traited by extracorporeal photopheresis

Project description:Background: The benefit of extracorporeal photopheresis on the course of kidney transplant rejection is unknown. The aim of our study was to investigate the variations in transcriptomics on graft biopsies when extracorporeal photopheresis was used to treat chronic humoral rejection after kidney transplantation. Methods: we retrospectively analyzed the mRNA expression of 770 genes of interest in graft biopsies performed before and after treatment. Eight patients received an average of 23 extracorporeal photopheresis sessions over 4 months between the two biopsies. Results: Transcriptomic analysis of the graft biopsies identified a significant (adjusted p-value< 0.05) increase in CAV1 mRNA in all patients and a significant decrease in CD19, IL21, PAX5, and SFTPA2 mRNAs in 7 of 8 patients. Conclusions: In patients treated with extracorporeal photopheresis for chronic humoral rejection after renal transplantation, omic analysis of repeated biopsies shows a reduction in fibrotic and inflammatory transcriptomic signatures.

Project description:BackgroundCardiac tamponade may present with very different signs and clinical consequences in patients who are supported with venoarterial extracorporeal membrane oxygenation. Failure to recognize cardiac tamponade in this setting can cause failure to wean from venoarterial extracorporeal membrane oxygenation, and even lead to death.Case presentationWe present a 44-year-old Caucasian female in whom cardiac tamponade manifested as venoarterial extracorporeal membrane oxygenation weaning failure. After discovering the contribution of cardiac tamponade, it was possible to wean the patient from venoarterial extracorporeal membrane oxygenation support. No clear signs of cardiac tamponade had existed beforehand.ConclusionsThe diagnosis of cardiac tamponade can be very challenging in venoarterial extracorporeal membrane oxygenation supported patients due to (patho)physiological particularities related to the parallel blood flow.
