Project description:ChIP-on-chip tilling array comparing untreated human SW480 cells vs SW480 cells treated with 2mM H2O2 for 30min. Exposing cells to the reactive oxygen species, hydrogen peroxide, recruits DNA methyltransferase 1 (DNMT1) to damaged chromatin. DNMT1 becomes part of a complex(es) containing DNMT3B and members of Polycomb Repressive Complex 4. The goal was to determine the effect of hydrogen peroxide treatment on chromatin, including changes in histone modifications and binding patterns of chromatin-associated proteins.

Project description:[original Title] Expression profiling of mouse embryonic fibroblasts with or without a deletion in the helicase domain of the Werner Syndrome gene homologue treated with hydrogen peroxide Werner syndrome (WS) is a rare disorder characterized by the premature onset of a number of age-related diseases. The gene responsible for WS is believed to be involved in different aspects of transcription, replication, and/or DNA repair. In addition to genomic instability, WS cells exhibit oxidative stress. In this report, we have examined the impact of exogenous peroxide on the expression profile of mouse embryonic fibroblasts lacking part of the helicase domain of the WRN homologue. This expression profile was compared to hydrogen peroxide treated wild type mouse embryonic fibroblasts. Interestingly, untreated mouse embryonic fibroblasts with a deletion of the helicase domain of Wrn already exhibited down regulation of several biological processes decreased by hydrogen peroxide in wild type cells.

Mutant gshF peroxide treatment

Project description:The goal of this study is to characterize a gshF::cam L. plantarum strain NZ7608. From phenotype characterization we have observed that the gene gshF plays a role in hydrogen peroxide resistance in L. plantarum and therefore we wanted to analyze the global transcriptome response of strain NZ7608 towards peroxide and compared this to wild type response. In this experiment we compared two L. plantarum strains: WCFS1 (wild type) and NZ7618 (gshF::cam). Both strains were grown -in duplo- on CDM containing 0.5% glucose at 37 degrees celsius. When cell density reached OD600=1.0 a hydrogen peroxide stress was given to the cultures to a final concentration of 10mM. Samples were taken at times (0 10 and 30 minutes) upon hydrogen peroxide. All withdrawn samples were used for this microarray experiment. Keywords: peroxide, gshF mutant
