Project description:Introduction: The flavonoid myricetin has been shown to induce cell cycle arrest and mitochondrial-dependent apoptosis in preclinical cancer models. We hypothesised that myricetin-derived flavonoids with redox properties and improved physicochemical attributes that enhance cell uptake and mitochondrial targeting might have increased potential as antitumour agents, since mitochondria are the main site of production of reactive oxygen species (ROS) and redox status is known to regulate the development and advancement of certain cancers. In this study we assessed a small library of novel flavonoids, then focussed on the lead compound, second-generation analogue OncamexTM, its mechanism of action and structure-activity relationships as an anti-proliferative agent. Methods: With this aim, we studied the effect of OncamexTM in a panel of 7 breast cancer cell lines using proliferation, cytotoxicity and apoptosis assays. The redox properties and mitochondrial delivery of the drug were studied using cyclic voltammetry and fluorescence microscopy, respectively. The mechanism of action was further studied using western blotting, gene expression analysis and immunohistochemistry (IHC) of treated xenograft tissue from in vivo mice models. Results: Sulforhodamine B (SRB) proliferation assays demonstrated strong, anti-proliferative properties of OncamexTM, with IC50 values in the low micromolar range. Treatment for 8 h exerted concentration-dependent reduction in cell viability and induction of cytotoxicity and apoptosis, with increased caspase activation. Microarray analysis suggested that OncamexTM regulates changes in cell cycle and apoptosis at gene expression level. Fluorescence microscopy was used to investigate mitochondrial targeting and ROS regulation in treated cells. OncamexTM was found to induce production of superoxide at concentrations which exerted anti-proliferative effects. Initial in vivo studies in mice implanted with a MDA-MB-231 breast cancer xenograft showed that OncamexTM inhibited tumour growth, reducing tissue viability and Ki-67 proliferation, with no overall systemic toxicity. Conclusions: OncamexTM is a novel flavonoid capable of specific delivery to the mitochondria and induction of ROS production. We have shown its antitumor activity in preclinical models of breast cancer, both in vitro and in initial in vivo models, where tumour growth was arrested without inducing toxicity, support the potential of this novel drug for its continued development as an anticancer agent. Project description:This study has identified a unique susceptibility that capitalizes on a cancer cell’s reliance on key metabolic pathways, not only to meet increased energetic demands, but also to maintain the critical balance between reactive oxygen species (ROS) and cellular antioxidant levels, necessary for cancer cell survival and growth. Specifically, we explored the use of phenformin, a metabolic drug that is being repurposed as a cancer treatment in clinical trials. We find that several breast cancer cell lines, across molecular subtypes, were sensitized to phenformin when combined with the pro-inflammatory cytokine, interferon-gamma (IFNγ), revealing a previously unidentified anti-tumor effect. Similarly, we find that an elevated inflammatory response (stimulated by PolyIC) synergizes with phenformin to attenuate tumor growth, in three different mouse models of breast cancer. Through inhibiting complex I of the electron transport chain, phenformin directly inhibits tumor growth by suppressing mitochondrial-dependent energy generation, and consequently increases levels of superoxide. Functionally, the mitochondrial ROS scavenger, MitoTempo, reverses the ability of, IFNγ and PolyIC to synergize with phenformin. Through RNA sequencing, by comparing Wild-Type and STAT1 KO cells, we identify Nqo1 as a target gene that decreases upon IFNγ treatment in a STAT1 dependent manner. Nqo1 is a 2-electron reductase that is upregulated in many tumors and has important roles in ROS scavenging and cellular stress responses. In support, targeting Nqo1 either by shRNA-mediated gene silencing or through its bioactivatable inhibitor, β-lapachone, sensitized established human and mouse cell lines, cells from breast cancer patient-derived xenografts (PDX), and in vivo xenografted tumor growth to phenformin.

Project description:Comparison af adherent growing breast cancer cell lines versus mammospheres under serum-free conditions Background: In patients with breast cancer, subsets of long-lived cells tolerate chemotherapies. These chemoresistant cells can remain dormant at secondary sites, such as bone and lung, for years and decades. Cancer cells endowed with drug resistance are maintained in vivo in a quiescent slow-growing state that preserves them from anti-proliferative cancer drugs. The mechanism of conversion from dormancy to growth remains poorly understood. We aimed to identify microRNAs (miRNAs) as master regulators of cancer stem cells (CSCs) maintainance, because one of the characteristics of CSCs is their slow proliferation or dormancy. MiRNA targeting CSCs may be an effective therapy to improve the prognosis of breast cancer patients. Methods: We performed miRNA array analysis to identify differences miRNA expression profiles between adherent cells and mammospheres that contain higher number of breast cancer stem cells (BCSCs). In this approach, we focused on expression of hsa-miR-27a. Further, roles of hsa-miR-27a target genes in maintaining BCSC properties were analysed . Results: Here, we showed that hsa-miR-27a was downregulated in mammosphere cells. The formation of BCSCs was attenuated by transfection of hsa-miR-27a. We found that hsa-miR-27a targets F-box and WD-40 domain protein 7 (FBW7), a tumor suppressor gene, thereby inducing tumor dormancy. Additionally, we found that hsa-miR-27a targets genes involved in GSH synthesis, including xCT (encoded by SLC7A11), a heterodimeric protein of a transporter subunit of the xC(-) system, cystathionine gamma-lyase (CTH/CSE), and nuclear factor-erythroid 2-related factor 2 (Nrf2/NFE2L2), thereby increasing intracellular reactive oxygen species (ROS) levels. Upregulation of these genes by antisense (as)-miR-27a induced autophagy. Conclusions: Previous studies showed that expression of hsa-miR-27a is elevated in breast cancer. Here, we showed that expression of hsa-miR-27a was reduced in BCSC. CSCs in the dormant state are resistant to chemotherapy due to inhibition of ROS accumulation in these cells. These results demonstrate that hsa-miR-27a plays an essential role in maintaining the BCSCs phenotype via regulation of FBW7, ROS related genes and the Nrf2 pathway, as well as in the dormant-to-proliferative switch of breast cancer cells. Thus, hsa-miR-27a represents a novel strategy for various type of breast cancer.

Project description:Malignant carcinomas that recur following therapy are typically de-differentiated and multi-drug resistant (MDR). De-differentiated cancer cells acquire MDR by upregulating reactive oxygen species (ROS)-scavenging enzymes and drug efflux pumps, but how these genes are upregulated in response to de-differentiation is not known. Here, we examine this question by using global transcriptional profiling to identify ROS-induced genes that are already upregulated in de-differentiated cells, even in the absence of oxidative damage. Using this approach, we found that the Nrf2 transcription factor, which is the master regulator of cellular response to oxidative stress, is pre-activated in de-differentiated cells. In de-differentiated cells, Nrf2 is not activated by oxidation but rather through a non-canonical mechanism involving its phosphorylation by the ER membrane kinase PERK. In contrast, differentiated cells require oxidative damage to activate Nrf2. Constitutive PERK-Nrf2 signaling protects de-differentiated cells from chemotherapy by reducing ROS levels and increasing drug efflux. These findings are validated in therapy-resistant basal breast cancer cell lines and animal models, where inhibition of the PERK-Nrf2 signaling axis reversed the MDR of de-differentiated cancer cells. Additionally, analysis of patient tumor datasets showed that a PERK pathway signature correlates strongly with chemotherapy resistance, tumor grade, and overall survival. Collectively, these results indicate that de-differentiated cells upregulate MDR genes via PERK-Nrf2 signaling, and suggest that targeting this pathway could sensitize drug-resistant cells to chemotherapy. Differentiated human breast epithelial cells (HMLE-shGFP) or de-differentiated cells (HMLE-Twist) were treated in culture with 40uM menadione for 2 hours or 1uM PERK inhibitor for 48 hours and compared to control DMSO-treated cells.

Project description:Triple-negative breast cancer is a difficult breast cancer subtype to treat that lacks targeted therapies. Specific miRNAs have been reported to be involved in cancer progression and their significance in cancer diagnosis and anti-cancer drug development has been highlighted; however, whether redox-associated miRNAs are potential targets of anti-cancer drugs or bioactive phytocompounds, which deregulate cancer cell activity through modulating ROS dynamics is unclear. This study addressed the anti-TNBC cell effect of phytosesquiterpene lactones deoxyelephantopin (DET) and its derivative DETD-35 by which redox-associated miRNAs were responsive and regulated as demonstrated by comparative miRNA microarray analysis and ROS scavenging approaches.

Project description:Despite advances in breast cancer screening and treatment, prognosis for metastatic disease remains dismal at 30% five-year survival. This is due, in large, to the failure of current therapeutics to target properties unique to metastatic cells. One of the drivers of metastasis is miR-10b, a small noncoding RNA implicated in cancer cell invasion, migration, viability, and proliferation. We have developed a nanodrug termed MN-anti-miR10b that delivers anti-miR10b antisense oligomers to cancer cells. In mouse models of metastatic triple-negative breast cancer, MN-anti-miR10b has been shown to prevent onset of metastasis and eliminate existing metastasis in combination with chemotherapy even after treatment has been stopped. Recent studies have implicated miR-10b in conferring stem cell-like properties onto cancer cells, such as chemoresistance. In this study, we show transcriptional evidence that inhibition of miR-10b with MN-anti-miR10b activates developmental processes in cancer cells and that stem-like cancer cells have increased miR-10b expression. We then demonstrate that treatment of breast cancer cells with MN-anti-miR10b reduces their stemness confirming that these properties make metastatic cells susceptible to the nanodrug actions. Collectively, these findings indicate that inhibition of miR-10b functions to impair breast cancer cell stemness, positioning MN-anti-miR10b as an effective treatment option for stem-like breast cancer subtypes.
